High-performance liquid chromatography-electrospray ionization mass spectrometry (HPLC-ESI-MS) and tandem mass spectrometry (MS(n)) were used to investigate the phenolic constituents in methanol, water, and methanol/water extracts of Eucalyptus globulus Labill. bark. Twenty-nine phenolic compounds were identified, 16 of them referenced for the first time as constituents of E. globulus bark, namely, quinic, dihydroxyphenylacetic, and caffeic acids, bis(hexahydroxydiphenoyl (HHDP))-glucose, galloyl-bis(HHDP)-glucose, galloyl-HHDP-glucose, isorhamentin-hexoside, quercetin-hexoside, methylellagic acid (EA)-pentose conjugate, myricetin-rhamnoside, isorhamnetin-rhamnoside, mearnsetin, phloridzin, mearnsetin-hexoside, luteolin, and a proanthocyanidin B-type dimer. Digalloylglucose was identified as the major compound in the methanol and methanol/water extracts, followed by isorhamnetin-rhamnoside in the methanol extract and by catechin in the methanol/water extract, whereas in the water extract catechin and galloyl- HHDP-glucose were identified as the predominant components. The methanol/water extract was shown be the most efficient to isolate phenolic compounds identified in E. globulus bark. 相似文献

The "maize white spot" foliar disease is a problem of increasing importance to Brazilian maize crops. A bacterium isolated from water-soaked lesions from infected maize leaves was pathogenic in biological assays in vivo. It was identified as a Gram-negative, nonsporulating, facultative anaerobic bacterium, belonging to the genus Pantoea. Chemical study of the extracts from bacterial cultivation media allowed the identification of (S)-(-)-N-butanoyl-homoserine lactone and trace amounts of N-hexanoyl-homoserine lactone, widely recognized quorum-sensing signaling substances employed in cell-to-cell communication systems. The absolute configuration of natural (S)-(-)-N-butanoyl-homoserine lactone was determined by gas chromatography-flame ionization detection with a chiral stationary phase and by comparison of circular dichroism spectroscopic data with enantiopure synthetic substances. Biological evaluations with reporter Agrobacterium tumefaciens NTL4(pZLR4) were carried out with synthetic and natural products and also with extracts from maize leaves contaminated with the isolated bacterium, as well as from healthy leaves. 相似文献

Biologically active compounds with different modes of action, such as, antiproliferative, antioxidant, antimicrotubule, have been isolated from marine sources, specifically algae and cyanobacteria. Recently research has been focused on peptides from marine animal sources, since they have been found as secondary metabolites from sponges, ascidians, tunicates, and mollusks. The structural characteristics of these peptides include various unusual amino acid residues which may be responsible for their bioactivity. Moreover, protein hydrolysates formed by the enzymatic digestion of aquatic and marine by-products are an important source of bioactive peptides. Purified peptides from these sources have been shown to have antioxidant activity and cytotoxic effect on several human cancer cell lines such as HeLa, AGS, and DLD-1. These characteristics imply that the use of peptides from marine sources has potential for the prevention and treatment of cancer, and that they might also be useful as molecular models in anticancer drug research. This review focuses on the latest studies and critical research in this field, and evidences the immense potential of marine animals as bioactive peptide sources. 相似文献

A cytologic study of the choroid plexi of animals and humans was carried out using impression smears (imprints, imp) to understand better the cellular changes that occur in the cerebrospinal fluid in the case of disease. The samples, totaling 756 imp were from 11 dogs (239 imp), 10 horses (219 imp), 1 mule (23 imp), 3 cattle (69 imp), 1 sheep (19 imp), 2 pigs (39 imp), 1 deer (20 imp), 4 monkeys (22 imp), and 7 humans (106 imp). The samples came from individuals clinically free of neurologic disease, as well as from a few abnormal cases. Six of the 7 humans had no history of neurologic disease and had a complete necropsy with brain histopathology. The seventh human case had mild neurologic signs at the time of death and only a partial necropsy with histopathological examination of the brain, in which a few small leptomeningeal lymphocytic infiltrates and polymicrogyria were found. One of the human brains without neurologic disease had arteriosclerosis. In the 40 individuals studied, several features and some unique cell types were found, for which little or no information is available. Four different morphologic cell types were consistently found in all the species studied. The first 3 types were arbitrarily named alpha (with deeply basophilic cytoplasm), beta (with neutral to weakly acidophilic cytoplasm), and gamma or vesicle-bearing cells. The third type, gamma, was a cell bearing unique inclusions (vesicles) filled with many tiny light tan to pale pink granules. The fourth type was the Kolmer cell found in very low numbers. Immature lymphocytes were found in all of 3 newborn foals, in 1 pig, and in the only stillborn calf and deer studied. The results suggest that the choroid plexi contain more than 1 epithelial cell type and that knowledge of their morphology is far from complete because other unusual cells and structures are also present in small numbers. Imprints are excellent for studying the choroid plexi, especially for tiny changes that are too subtle to be seen in hematoxylin and eosin sections. 相似文献

Juvenile (5.5–5.7 g) white leg shrimp were exposed to mycotoxin in one indoor trial by feeding fumonisin B1 (FB1) at levels from 0.2 to 2.0 μg g?1 FB1 for 30 days. Shrimp growth was affected after exposure to more than 0.6 μg g?1 FB1. Soluble muscle protein concentration decreased, and changes in myosin thermodynamic properties were observed in shrimp after 30 days of exposure to FB1. Marked histological changes in tissue of shrimp fed a diet containing FB1 at 2.0 μg g?1 were also observed. Shrimp fed diets containing more than 0.6 μg g?1 FB1 showed greater decrease in shear forces after 12 days of ice storage. 相似文献

Magnetic resonance imaging (MRI) of the distal extremities of the standing, sedated horse would be desirable if diagnostic quality images could be obtained. With the availability of extremity and special purpose magnet designs on the market, a system to safely accommodate the standing horse may gain increasing popularity. This paper considers the issue of motion that will need to be addressed to achieve successful, diagnostic quality images. The motion of the carpus and tarsus of five standing, sedated horses was quantified. The obtained motion records were then used to induce motion in cadaveric joint specimens during several MRI scans. The measured dorsal-palmar/plantar, medial-lateral, and proximal-distal random wobbling motions in the standing sedated horse were several centimeters in magnitude and generated severe motion-artifacts during axial MRI of the cadaveric specimens. Two retrospective motion-correction techniques (autocorrection and navigator-based adaptive correction) were used to correct the corrupted images. The motion artifacts were nearly eliminated with the use of both techniques in series. Although significant hurdles remain, these results suggest promise for allowing diagnostic quality MRI of the carpus and tarsus in the standing horse. 相似文献

The Enzyme-linked Immunoelectrotransfer Blot (EITB) has been used widely as a screening test for Taenia solium cysticercosis in swine. However, the relation between seropositivity and infection in pig populations from endemic areas has not been well defined. The aim of this study is to relate EITB seropositivity with infection and infection burden, analyse the trade-off between sensitivity and specificity with various cut-off points for the EITB assay, and finally describe the serology changes in a cohort of rural pigs raised under natural conditions. A group of 107 pigs that were used as controls during a vaccination field trial in Peru was our study population. The prevalence of porcine cysticercosis determined by necropsy examination was 16.82% (18/107) in these animals. Using EITB reactivity to ≥1 band as a cut-off point for the assay, the sensitivity was 88.89% (65.29–98.62, 95% CI) and the specificity was 48.31% (37.59–59.16, 95% CI). Comparing other cut-off points, involving up to as many as 7 reactive bands, a reactivity of ≥3 bands provided the best trade-offs in sensitivity and specificity. Using this cut-off point for the assay, the sensitivity was 77.77% (52.36–93.59, 95% CI) and the specificity was 76.40% (66.22–84.76, 95% CI). A significant association was found between cyst counts over 100 cysts and reactivity to ≥3 bands in the EITB assay (Fisher's exact test, p < 0.05). The results of this study suggest that the use of the EITB assay to study porcine cysticercosis may require setting different cut-offs under field and experimental conditions, and depending upon the objective of the screening process. 相似文献

Elbow joint incongruity is recognized as an important factor in the development, treatment, and prognosis of canine elbow dysplasia. Elbow incongruity has been measured based on radiographic joint space widths, however these values can be affected by the degree of elbow joint flexion. Recent studies have reported radiographic curvature radii as more precise measures of humeroulnar congruity in dogs. The aim of this prospective observational study was to describe radiographic curvature radii measured from flexed and extended elbow radiographs for a sample of dogs representing a medium breed (Portuguese Pointing Dog) and a large breed (Estrela Mountain Dog). The curvature radii from the ulnar trochlear notch and humeral trochlea were measured in 114 mediolateral elbow extended radiographic views (30 Portuguese Pointing Dog and 27 Estrela Mountain Dog), and 84 mediolateral flexed views (22 Portuguese Pointing Dog and 20 Estrela Mountain Dog). The sampled animals' ages ranged from 12 to 84 months (34.6 ± 17.8 months). Good agreement was observed between curvature radii measurements for flexed vs. extended views in both breed groups. Ulnar trochlear notch curvature radii measurements were greater than humeral trochlea curvature radii measurements in both breed groups. Both curvature radii were greater in the large‐breed dog group vs. the medium‐breed dog group. Both breed groups had ulnar and humeral curves with similar typology. However, the large breed group had greater intermediate differences between the humeroulnar surface curvature radii. Results from this study supported the use of curvature radii as measures of humeroulnar congruity in mediolateral flexed elbow radiographs of medium and large breed dogs. 相似文献
