A study is made of the morphological characteristics of the endodermic cells of the stage 5 chick embryo by means of in vitro cell culture techniques. The scanning electron microscope revealed that the endodermic cells in cultures were rounded, tended to be smooth and had few blebs and microvilli. Regarding cell projections typical of culture cells, such as filopodia, lamellipodia and pseudopodia, there was a noteworthy scarcity after 12 h growth, although greater cellular activity was observed at 24 h, characterized by the presence of filopodia and an ability of the cells to form clusters on the substratum. These facts show the morphological and adhesion movements of the endodermal cells studied to be related mainly with the presence of filopodia as the most abundant cell projections.  相似文献

The negative effects of nematodes in small ruminants can be reduced by use of dewormers but their effectiveness is increasingly limited by the emergence of anthelmintic resistance. The efficacy of benzimidazole (BZ) anthelmintics in the Philippines was estimated by an in vitro larval development assay using worm eggs recovered from faeces collected from goats and sheep. Two hundred and eighteen farms were selected to represent areas of the country with high goat and sheep populations and the full range of farm sizes, from smallholders with just a few animals to commercial and institutional farms with several hundred. Initial surveys of worm control advisers indicated that BZs have been in continuous widespread use for up to 20 years with little use of other chemical groups. A larval development assay (LDA: DrenchRite) was modified for use with BZs alone to allow up to five samples to be analysed on a single microtitre plate. The assay was validated by comparison with the faecal egg count reduction test (FECRT). The dominant nematode genera were Haemonchus and Trichostrongylus with small numbers of Oesophagostomum. The range of BZ efficacy estimated from the LDA results was 0-100% and the distribution of efficacy levels was continuous, with mean efficacy of 82 and 64% for goats and sheep, respectively. There were significant associations between efficacy and parameters measured to characterize the sampled farms: size of animal management group, FEC of sample, recent importation of stock and no access to common grazing were all correlated with decreased efficacy. Likewise, low efficacy was associated with reported frequency and number of years that BZ drenches had been used. The LDA was found to be highly suited to estimate efficacy in nematode populations from small farms where performance of a FECRT for even one chemical would be impractical. Using a larval development assay, we have demonstrated a wide efficacy range for BZs against nematodes from all sizes of goat and sheep farms in the tropics.  相似文献

ABSTRACT A field experiment was designed to test the hypothesis of for increased reproductive ability on different host genetic backgrounds within a wheat powdery mildew population. Studies have suggested that, in host mixtures, such selection could increase the reproduction rate of simple patho-types that always develop on the same host genetic background, whereas complex pathotypes should not be affected because they infect different host genotypes. In our experiment, the Erysiphe graminis population reproduced for successive generations on cvs. Orkis and Etecho, either grown as pure stands or in a mixture. In an additional treatment, the host cultivar changed after each generation. Isolates were sampled in April and, after seven pathogen generations, in July. At the second sampling date and for pure stands only, mean spore efficacy was greater on the host from which isolates were sampled than on the other one. This was attributed to selection within the pathogen population for better spore efficacy on the host genetic background. This selection was independent of the virulence genes carried by the isolates. The possibility of a phenotypic plasticity effect was tested and rejected.  相似文献

For Holm oak (Quercus ilex L.), a flush growing species, nutrient loading during the growing season is difficult and can lead to a low nutrient status of the seedlings. To provide insights about Holm oak nutrient dynamics during fall in the nursery and subsequent planting performance, a factorial nursery experiment was conducted in a mild fall–winter area testing the effects of timing of fertilization (early and late fall) and rate (two doses of a NPK fertilizer that applied 28 and 56 mg N per seedling), followed by an experimental plantation.  相似文献

Soil organic-N dynamics, its controlling factors and its relationships with stand quality were studied in the 0-15 cm soil layer of 24 pinewoods with contrasting age, productivity and parent material (granite; acid schists), searching for N variables useful to predict stand growth and site quality. No significant differences were found between young and old stands for any of the N variables considered, nor two- or three-order interactions among stand age, site quality and parent material. The soil total-N content, which was correlated positively with the Al oxides content (a soil organic matter (SOM) stabilizing agent), did not vary significantly according to parent material, but it was lower (P≤0.02) in stands with high than with low site index (2.68±1.11 and 3.97±1.13 g N kg−1 soil, respectively). The soil δ15N ranged from +3.5 to +6.5 δ, without significant differences among stand groups, and it was negatively correlated with water holding capacity, exchangeable bases, Al oxides and N content, suggesting that: (i) N losses by NO3 leaching are the most important controlling factor of δ15N in these temperate humid region soils; and (ii) soil N richness is related with limited N losses, which discriminate against 15N. At any incubation time, no significant differences were found in soil inorganic-N content among stand groups (7.78±4.57, 39.33±16.20 and 67.80±26.50 mg N kg−1 soil at 0, 42 and 84 d, respectively). During the incubation, the relative importance of ammonification decreased and that of the nitrification increased. The net N mineralization rate (NNMR, in percentage of organic N) was significantly higher in granite than in schists soils at both 42 d (1.24±0.34 and 0.75±0.37%, respectively) and 84 d (2.18±0.56 and 1.53±0.66%, respectively). In high quality pinewoods, the NNMR at 42 and 84 d (1.16±0.45 and 2.12±0.79%, respectively) were significantly higher than in low quality stands (0.83±0.35 and 1.59±0.45%, respectively). This result, together with those on soil total-N and inorganic-N supply, suggests that soil N dynamics in low and high quality stands are different: in the former there is a bigger N pool with a slower turnover, whereas in the latter there is a smaller N pool with a faster turnover, both factors being nearly compensated, making the soil available N supply in both types of stand similar. After 42 and 84 d of incubation, the NNMR and the nitrification rates were higher in the coarse textured soils, likely due to the low physical and chemical protection of their SOM; both rates were positively correlated with available P, exchangeable K+ and CEC base saturation, suggesting strong relationships among the availabilities of the main plant nutrients, and they increased with SOM quality (low C-to-N ratio). The strong negative correlation of site index with soil total-N (r=−0.707; P≤0.005), and its positive correlations with NNMR after 42 and 84 d of incubation, suggested that site quality and potential productivity are closely related to soil organic-N dynamics. Half of the site index variation in the stands studied could be predicted with a cheap and easy analysis of soil N content, the prediction being slightly improved if soil δ15N is included and, more significantly, by including N mineralization measurements.  相似文献

Bovine tuberculosis (bTB) is an established zoonotic disease which affects cattle and wildlife worldwide and new strategies are required to control and eradicate the disease. The European wild boar (Sus scrofa) is a major reservoir of bTB in Spain. The objective of this paper was to review tissue-specific response and function of mandibular lymph nodes (MLN) and oropharyngeal tonsils (OT) in European wild boar naturally infected with Mycobacterium bovis. Genomics and proteomics data were used to compare differential gene expression and global protein patterns in OT and MLN of M. bovis-infected and uninfected European wild boar and the results were analyzed considering previous reports of experimental infections in laboratory and domestic animals. The results showed tissue-specific differences in OT and MLN in response to M. bovis infection. Tissue-specific differences in gene expression and protein profiles suggested different functions for OT and MLN during mycobacterial infection and provided information to characterize the pathobiology of M. bovis infection in European wild boar with important implications for the control of bTB in Spain. The characterization of molecular events in tissues that play different roles during mycobacterial infection in naturally infected individuals may be relevant to understand the pathobiology of M. bovis infection and to design effective strategies for the control of bTB in wildlife reservoirs.  相似文献

N mineralization capacity and its main controlling factors were studied in a large variety (n=112) of native (forest, bush) and agricultural (pasture, cultivated) soils from several climatic zones in Spain. The available inorganic N content, net N mineralization, and net N mineralization rate were determined after 6 weeks of aerobic incubation. NH inf4 sup+ –N largely predominated over NO inf3 sup- -N (ratio near 10:1) except in some agricultural soils. Net N mineralization predominated (83% of soils) over net N immobilization, which was more frequent in agricultural soils (25%) than in native soils (9%). In forest soils, both net N mineralization and the net N mineralization rate were significantly higher than in the other soil groups. The net N mineralization rate of pasture and cultivated soils was similar to that of bush soils, but available inorganic N was lower. The net N mineralization rate decreased in the order: soils over acid rocks>soils over sediments>soils over basic rocks or limestone; moreover, the highest net N mineralization and available inorganic N were found in soils over acid rocks. The highest N mineralization was found in soils with low C and N contents, particularly in the native soils, in which N mineralization increased as the C:N ratio increased. N mineralization was higher in soils with a low pH and base saturation than in soils with high pH and base saturation values, which sometimes favoured N immobilization. Soils with an Al gel content of >1% showed lower net N mineralization rates than soils with Al gel contents of <1%, although net N mineralization and available inorganic N did not differ between these groups. The net N mineralization rate in silty soils was significantly lower than in sandy and clayey soils, although soil texture only explained a low proportion of the differences in N mineralization between soils.  相似文献

The effect on productive and vegetative behavior and on the quality of oil from Olea europaea L. when applying two distinct irrigation techniques, full irrigation (FI) and regulated deficit irrigation (RDI), was studied. A total of five wet soil volumes (WSVs, 12, 24, 35, 47 and 59%) expressed in terms of the potential root exploration volume were established for each strategy. The experiment was performed on cv. ‘Arbequina’ in an olive grove in Tarragona (Spain). Results obtained suggest that a 20% reduction in the irrigation dose (RDI) had no significant effect either on olive fruit and oil production or on oil content. Likewise, no significant increase in irrigation water-use efficiency was observed for FI with respect to RDI. A tendency for olive and oil production per hectare to increase with increased WSV percentage was observed, although there were no significant differences between FI and RDI except for 59% WSV in the RDI strategy, producing the best response.  相似文献

Generally, soil compaction is a stress factor affecting negatively the plant growth, but its effects vary between species and with the soil compaction range. The objective of this study is to know the different growth responses of 17 woody species subjected to moderate soil compaction, because most of the studies about this subject compare the effects in treatments with a wide and discrete compaction range. We explore the effects of moderate soil compaction on a continuous scale (0.1–1.0 MPa) on seedling growth. Seedlings of 17 woody species (deciduous and evergreens) mainly from Mediterranean ecosystems were grown in near optimal conditions (light, temperature and water) in a greenhouse with a sandy substrate. In general, there was a great variability of the responses depending on the species and the studied variable. About 53% of the species showed a higher total biomass with a moderate increase in soil compaction possibly being due to a greater root–soil contact. In the same way, 41% of species increase the relative growth rate and 35% the total area. Nevertheless, in spite of these positive effects on growth, for some species (23%) there was a decrease in the root proportion with soil resistance, as result of soil strength. These effects of moderate soil compaction could sum up in two general responses of woody plants: growth increment and architecture distortion. This might be relevant as a lower root investment may be a disadvantage under drought conditions. Finally, a simple conceptual model is proposed to understand the general effects of soil compaction on growth and biomass allocation.  相似文献
