The application of disinfectants through drip irrigation could be a feasible practice against verticillium wilt (Verticillium dahliae) of olive. OX-VIRIN (activated peroxide) and OX-AGUA AL25 (quaternary ammonium compounds) are two disinfectants that have shown efficacy against V. dahliae in irrigation water and potential for reducing the disease in young olive plants. In this work, various post-planting application strategies incorporating OX-VIRIN (once a month, or twice a month on alternate or successive weeks) or OX-AGUA AL25 (once a month, or twice a month on alternate weeks) were assessed for their effect on V. dahliae in soil, disease in olive trees, and olive yield, in a 2-year pot-experiment under natural environmental conditions. The disinfectants were injected via metering pumps into a drip irrigation system that irrigated olive trees planted in V. dahliae-inoculated soil. All the application strategies significantly reduced the total inoculum density in soil compared to controls with no disinfectants and noninoculated soil. The microsclerotia density was also significantly reduced in disinfested soils by 73.6–86.8%, depending on the strategy. The symptoms and infection incidence were always lower in treatments subjected to disinfestation. The treatment with OX-AGUA AL25 applied twice a month on alternate weeks most reduced the symptoms (by 53.0%) and colonization index (by 70.8%) with respect to untreated water control. This soil disinfestation also significantly strengthened the symptom remission. Tree growth and production were negatively affected by soil inoculation (reduced by 45.6% and 88.7%, respectively), but not so by disinfectants, which even relieved the reduction in inoculated soils, especially when OX-AGUA AL25 was applied. 相似文献

The shape of the lipid vesicle (the ratio between the maximal and the minimal diameter and the shape coefficient) of the eggs of Diplodus puntazzo and Sparus aurata is correlated with larval survival rate (%) and can be used as an egg quality marker as reported previously by Lahnsteiner and Patarnello (2005) . In the present study, this method for egg quality determination has also been validated for common dentex, Dentex dentex, after introducing several modifications in the calculation and evaluation procedure in order to obtain higher sensitivity. In the present study, it could be demonstrated that the shape of the lipid vesicle is not only correlated with the hatching rate but also with the survival rate of 1, 2 and 3 days post hatch larvae and therefore it can also be used as an indicator of larval viability at early stages of development. 相似文献

A flow cytometric virus-binding assay that directly visualizes the binding and entry of infectious pancreatic necrosis virus (IPNV), infectious haematopoietic necrosis virus (IHNV) and virus haemorrhagic septicaemia virus (VHSV) to several cell lines was established. The highest efficiency of binding was shown by the BF-2 cell line and this was used to study, at the attachment level, the interactions of these cells with salmonid fish viruses in coinfections, and to further determine if the earliest stage of the viral growth cycle could explain the previously described loss of infectivity of IHNV when IPNV is present. Our results demonstrated that IPNV binds to around 88% of cells either in single or dual infections, whereas IHNV attachment always decreased in the presence of any of the other viruses. VHSV binding was not affected by IPNV, but coinfection with IHNV reduced the percentage of virus-binding cells, which suggests competition for viral receptors or co-receptors. Internalization of the adsorbed IHNV was not decreased by coinfection with IPNV, so the hypothetical competence could be restricted to the binding step. Treatment of the cells with antiviral agents, such as amantadine or chloroquine, did not affect the binding of IPNV and VHSV, but reduced IHNV binding by more than 30%. Tributylamine affected viral binding of the three viruses to different degrees and inhibited IPNV or IHNV entry in a large percentage of cells treated for 30 min. Tributylamine also inhibited IHNV cytopathic effects in a dose-dependent manner, decreasing the virus yield by 4 log of the 50% endpoint titre, at 10 mm concentration. IPNV was also inhibited, but at a lower level. The results of this study support the hypothesis that IHNV, in contrast to VHSV or IPNV, is less efficient at completing its growth cycle in cells with a simultaneous infection with IPNV. Accumulation and depletion of soil phosphorus (P) was studied in a long‐term (37 y) field experiment in Southern Finland. The loam soil had a high pH (7.5–7.7) due to an earlier liming. Spring barley, spring wheat, oat, and ryegrass, grown in rotation, were annually fertilized with 0, 32, or 67 kg P ha?1 y?1 (P0, P1, and P2K) and sufficient N. The average dry matter grain yield 2,600 kg ha?1 of the P0 plots increased by about 500 kg ha?1 at P1 treatment and another 600 kg ha?1 by P2K. Soil samples were collected in 1978 (beginning), 1995, 2005, and 2015. According to the Chang and Jackson sequential extraction, the P2K and P1 treatments increased the inorganic soil P by 732 and 32 kg P ha ?1 in 37 years, respectively, while the P0 plots were depleted by –459 kg P ha ?1. The P2K treatment increased all four P fractions, extracted with NH4Cl (easily soluble), NH4F (Al‐P), NaOH (Fe‐P), and H2SO4 (Ca‐P). Continuous depletion (P0) decreased the NH4Cl‐P and NH4F‐P pools, NaOH‐P and H2SO4‐P pools remaining stable. None of the P pools changed significantly at P1. The remarkable gap between the measured change and the balance for the P2K and P1 treatments cannot be explained solely by lateral soil movement, meaning that a significant proportion of the applied P was lost either in surface runoff or transported below the investigated depth of 40 cm. Despite large P applications, the degree of P saturation reached only 20% in the P2K topsoil, assuming a 50% reactivity of Fe and Al oxides. As derived from sorption isotherms, a high EPC0 (i.e., equilibrium P concentration at zero net P sorption or desorption) of 1.30 mg L?1 had been built up in the P2K treatment, while in the P1 treatment EPC0 (0.33 mg L?1) had remained unchanged and P depletion (P0) had caused a decrease to 0.12 mg L?1. These results demonstrate that P sorption and desorption properties respond strongly to both P fertilization and null fertilization treatments and that in a long‐term field experiment only a low proportion of the residual fertilizer P can be recovered from soil. 相似文献

In this paper we present surface dynamic properties (interfacial tension and surface dilational properties) of a whey protein isolate with a high content of beta-lactoglobulin (WPI) adsorbed on the oil-water interface as a function of adsorption time. The experiments were performed at constant temperature (20 degrees C), pH (5), and ionic strength (0.05 M). The surface rheological parameters and the interfacial tension were measured as a function of WPI concentration (ranging from 1 x 10(-)(1) to 1 x 10(-)(5)% w/w) and different processing factors (effect of convection and heat treatment). We found that the interfacial pressure, pi, and surface dilational modulus, E, increase and the phase angle, phi, decreases with time, theta, which should be associated with WPI adsorption. These phenomena have been related to diffusion of the protein toward the interface (at short adsorption time) and to the protein unfolding and/or protein-protein interactions (at long-term adsorption) as a function of protein concentration in solution and processing conditions. 相似文献

Supercritical fluid carbon dioxide extraction (SFE) has been evaluated for the extraction of 17 organohalogen and organophosphate pesticides in gazpacho (a table-ready food composite containing crude vegetables, white bread, vegetable oil, water, and other minor components) using anhydrous magnesium sulfate as drying agent. The effects of different parameters, such as fat content in gazpacho composites, magnesium sulfate/gazpacho ratio, supercritical fluid volume, pressure, temperature, and static modifier additions, on SFE recoveries from spiked gazpacho samples have been studied. Analyses were performed by gas chromatography (GC) with flame photometric (FPD), electron capture (ECD), and mass spectrometry (MSD) detectors. In most experiments, recoveries obtained for the nonpolar organohalogen pesticides were lower than those obtained for the most polar organophosphate pesticides, but overall pesticide recoveries determined by using the optimal SFE conditions indicate that SFE could be used to determine pesticide residue levels in gazpacho. 相似文献

Irrigation scheduling based on the daily historical crop evapotranspiration (ETh) data was theoretically and experimentally assessed for the major soil-grown greenhouse horticultural crops on the Almería coast in order to improve irrigation efficiency. Overall, the simulated seasonal ETh values for different crop cycles from 41 greenhouses were not significantly different from the corresponding values of real-time crop evapotranspiration (ETc). Additionally, for the main greenhouse crops on the Almería coast, the simulated values of the maximum cumulative soil water deficit in each of the 15 consecutive growth cycles (1988–2002) were determined using simple soil-water balances comparing daily ETh and ETc values to schedule irrigation. In most cases, no soil-water deficits affecting greenhouse crop productivity were detected, but the few cases found led us to also assess experimentally the use of ETh for irrigation scheduling of greenhouse horticultural crops. The response of five greenhouse crops to water applications scheduled with daily estimates of ETh and ETc was evaluated in a typical enarenado soil. In tomato, fruit yield did not differ statistically between irrigation treatments, but the spring green bean irrigated using the ETh data presented lower yield than that irrigated using the ETc data. In the remaining experiments, the irrigation-management method based on ETh data was modified to consider the standard deviation of the inter-annual greenhouse reference ET. No differences between irrigation treatments were found for productivity of pepper, zucchini and melon crops. 相似文献

Conventional tillage creates soil physical conditions that may restrict earthworm movement and accelerate crop residue decomposition, thus reducing the food supply for earthworms. These negative impacts may be alleviated by retaining crop residues in agroecosystems. The objective of this study was to determine the effects of various tillage and crop residue management practices on earthworm populations in the field and earthworm growth under controlled conditions. Population assessments were conducted at two long-term (15+ years) experimental sites in Québec, Canada with three tillage systems: moldboard plow/disk harrow (CT), chisel plow or disk harrow (RT) and no tillage (NT), as well as two levels of crop residue inputs (high and low). Earthworm growth was assessed in intact soil cores from both sites. In the field, earthworm populations and biomass were greater with long-term NT than CT and RT practices, but not affected by crop residue management. Laboratory growth rates of Aporrectodea turgida (Eisen) in intact soil cores were affected by tillage and residue inputs, and were positively correlated with the soil organic C pool, suggesting that tillage and residue management practices that increase the soil organic C pool provide more organic substrates for earthworm growth. The highest earthworm growth rates were in soils from RT plots with high residue input, which differed from the response of earthworm populations to tillage and residue management treatments in the field. Our results suggest that tillage-induced disturbance probably has a greater impact than food availability on earthworm populations in cool, humid agroecosystems. 相似文献
