Low-temperature fermentations (13 degrees C) are considered to improve wine aromatic profiles. However, because the risk of stuck and sluggish fermentations is high, these fermentations are not common. The aim of this paper was to analyze the effect of different preadaptation protocols in two commercial wine strains on the fermentation and some wine parameters. Preadaptation is understood to be the process between the rehydration of active dry yeast and the inoculation. In this study, it consisted of preparing a fermentation starter (addition of yeast grown at 25 degrees C) or inocula preadapted at low temperatures (as before, but grown at a fermentation temperature of 13 or 17 degrees C). These results were compared with those of rehydrated active dry yeast, and a commercial "cryotolerant" yeast was used as a reference. General fermentation kinetic parameters, yeast imposition, nitrogen consumption, and main wine products were analyzed. The results showed that the preadaptation of a yeast could improve the fermentation performance, although this improvement was strain-dependent. Low-temperature fermentations also had some general effects: reduction of acetic acid and fusel alcohol production and increased concentrations of glycerol. When the yeast performed better in fermentation because of preadaptation, nitrogen consumption was faster and the wine's "negative" attributes (acetic acid, fusel alcohols) were significantly reduced. Thus, in some strains, preadaptation could be an effective mechanism for improving low-temperature fermentation, which also significantly reduces detrimental wine attributes.  相似文献

The composition of the essential oil from ripe and unripe berries and leaves of Juniperus oxycedrus L. ssp. oxycedrus, Juniperus phoenicea ssp. turbinata and Juniperus communis ssp. communis was analyzed by GC-MS, and microbiological assays were carried out. Samples were collected in different localities (Sardinia, Italy) and hydro distilled. The yields ranged between 2.54% +/- 0.21 (v\w dried weight) and 0.04% +/- 0.00. A total of 36 components were identified. The major compounds in the essential oils were alpha-pinene, beta-pinene, delta-3-carene, sabinene, myrcene, beta-phellandrene, limonene, and D-germacrene. Both qualitative and quantitative differences between species and between different parts of the plant were observed. The essential oils and their major compounds were tested against Candida albicans, Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa, and the minimum inhibitory concentration and minimum bactericidal concentration were determined. The results obtained led to a nonsignificant inhibitory effect, although all the essential oils from Juniperus phoenicea ssp. turbinata and the essential oil from leaves of Juniperus oxycedrus ssp. oxycedrus exhibited rather good or weak activity against Candida albicans and Staphylococcus aureus.  相似文献

The influence of enzyme mixtures containing amylase and lipase activities on straight dough bread staling was studied. Amylopectin retrogradation, crumb firming, amylose-lipid complexes, and dextrin production were analyzed in bread samples supplemented with two enzyme mixtures. The addition of enzyme mixtures to bread formula causes a beneficial effect on bread keeping properties and the formation of a more thermostable amylose-lipid complex than the one found in control bread. Amylopectin retrogradation was inhibited by the use of the enzyme; the effect was accompanied by reduced crumb-firming rates. The enzymatically generated water-soluble dextrins (maltose and DP3, DP4, DP5, and DP6 dextrins) are the most effective in preserving crumb softness during bread storage.  相似文献

In this work an HPLC method for the determination of azadirachtin residues on olives was developed, and the field degradation kinetics of the pesticide was studied. In field trials the active ingredient (a.i.) decay had a half-life time of 0.8 days, which was too short to show a good efficacy of treatment. The mechanism of disappearance of the pesticide studied with model systems showed that it was unrelated to evaporation, thermodegradation, and co-distillation, but it was related to photodegradation. The high photodegradation rate of commercial formulations calls for the need to test different formulates in order to increase the persistence of the residue and thus the pesticide's efficacy.  相似文献

The level of compaction induced on cultivated fields through trafficking is strongly influenced by the prevailing soil-water status and, depending on the attendant soil degradation, vital soil hydraulic processes could be affected. Therefore, understanding the relationship between field soil-water status and the corresponding level of induced compaction for a given load is considered an imperative step toward a better control of the occurrence of traffic-induced field soil compaction. Pore size distribution, a fundamental and highly degradable soil property, was measured in a Rhodic Ferralsol, the most productive and extensively distributed soil in Western Cuba, to study the effects of three levels of soil compaction on soil water characteristic parameters. Soil bulk density and cone penetration index were used to measure compaction levels established by seven passes of a 10 Mg tractor at three soil-water statuses corresponding to the plastic (Fs), friable (Fc) and relatively dry soil (Ds) consistency states. Pore size distribution calculated from soil water characteristic curves was classified into three pore size categories on the basis of their hydraulic functioning: >50 μm (f>50 μm), 50–0.5 μm (f50–0.5 μm) and <0.5 μm (f<0.5 μm). The greatest compaction levels were attained in the Fs and Fc soil water treatments, and a significant contribution to compaction was attributed to the existing soil water states under which the soil compaction was accomplished. Average cone index (CI) values in the range of 2.93–3.70 MPa reflected the accumulation of f<0.5 μm pores, and incurred severe reductions in the volume of f>50 μm pores in the Fs and Fc treatments, while an average CI value of 1.69 MPa indicated increments in the volume of f50–0.5 μm in the Ds treatment. Despite the differential effects of soil compaction on the distribution of the different pore size categories, soil total porosity (fTotal) was not effective in reflecting treatment effects. Soil water desorption at the soil water potentials evaluated (0.0 to −15,000 cm H2O) was adversely affected in the f<0.5 μm dominated treatments; strong soil water retention was observed with the predominance of f<0.5 μm, as was confirmed by the high water content at plant wilting point. Based on these findings, the use of field capacity water content as the upper limit of plant available soil water was therefore considered inappropriate for compacted soils.  相似文献

Abstract

New studies are needed to optimize the nitrogen (N) amount that can be applied to utilize the Azospirillum brasilense benefits. In addition, information regarding the interaction between the urease inhibitor and biological nitrogen fixation (BNF) and how they affect the macronutrients accumulation are also needed. We evaluate the effect of N sources and doses associated with A. brasilense regarding the macronutrients accumulation in straw and grains and wheat grain yield in tropical conditions. A randomized block experimental design was used with four replications in a 2?×?5?×?2 factorial arrangement as follows: two N sources (urea and urea with urease enzyme inhibitor NBPT; five N doses (0, 50, 100, 150, and 200?kg ha?1) applied in topdressing; with and without A. brasilense inoculation. We found that an increase in N doses positively influenced the accumulation of macronutrients in straw and grains and the wheat grain yield. N sources have similar effects. Inoculation with A. brasilense increased accumulation of Mg and S in straw and P, Ca, and Mg in grains, regardless of the N dose. The inoculation with A. brasilense associated with 140?kg ha?1 of N increased wheat grain yield. The inoculation can contribute in a more sustainable way to wheat nutrition and optimizing N fertilization.  相似文献

Mutational analysis of the tyrosine phosphatome in colorectal cancers   总被引:1,自引:0,他引:1  
Tyrosine phosphorylation, regulated by protein tyrosine phosphatases (PTPs) and kinases (PTKs), is important in signaling pathways underlying tumorigenesis. A mutational analysis of the tyrosine phosphatase gene superfamily in human cancers identified 83 somatic mutations in six PTPs (PTPRF, PTPRG, PTPRT, PTPN3, PTPN13, PTPN14), affecting 26% of colorectal cancers and a smaller fraction of lung, breast, and gastric cancers. Fifteen mutations were nonsense, frameshift, or splice-site alterations predicted to result in truncated proteins lacking phosphatase activity. Five missense mutations in the most commonly altered PTP (PTPRT) were biochemically examined and found to reduce phosphatase activity. Expression of wild-type but not a mutant PTPRT in human cancer cells inhibited cell growth. These observations suggest that the mutated tyrosine phosphatases are tumor suppressor genes, regulating cellular pathways that may be amenable to therapeutic intervention.  相似文献

Two beams of light can be quantum mechanically entangled through correlations of their phase and intensity fluctuations. For a pair of spatially extended image-carrying light fields, the concept of entanglement can be applied not only to the entire images but also to their smaller details. We used a spatially multimode amplifier based on four-wave mixing in a hot vapor to produce twin images that exhibit localized entanglement. The images can be bright fields that display position-dependent quantum noise reduction in their intensity difference or vacuum twin beams that are strongly entangled when projected onto a large range of different spatial modes. The high degree of spatial entanglement demonstrates that the system is an ideal source for parallel continuous-variable quantum information protocols.  相似文献

Organelle inheritance is an essential feature of all eukaryotic cells. As with other organelles, the Golgi complex partitions between daughter cells through the fission of its membranes into numerous tubulovesicular fragments. We found that the protein CtBP3/BARS (BARS) was responsible for driving the fission of Golgi membranes during mitosis in vivo. Moreover, by in vitro analysis, we identified two stages of this Golgi fragmentation process: disassembly of the Golgi stacks into a tubular network, and BARS-dependent fission of these tubules. Finally, this BARS-induced fission of Golgi membranes controlled the G2-to-prophase transition of the cell cycle, and hence cell division.  相似文献
