Understanding existing levels of genetic diversity of sheep breeds facilitates in situ and ex situ conservation activities. A comprehensive evaluation of US sheep breeds has not been previously performed; therefore, we evaluated the genetic diversity among and within 28 US sheep breeds. Both major and minor breeds were included in the analysis and consisted of 666 animals from 222 producers located in 38 states. The level of within-breed genetic diversity was variable and not dependent upon status of a breed as a major or minor breed. Bayesian cluster analysis indicated the breeds were grouped more by physiological differences (meat vs. wool production) rather than geographic origin. Results suggest several actionable items to improve in situ and ex situ conservation. The results clearly identify breeds in need of increased in situ and ex situ management (e.g., Hog Island and Karakul) and allow several suggestions for in situ management of flocks. Conversely, several of the breeds appear genetically similar and therefore require less emphasis on collecting germplasm samples for the gene bank. Commercially important breeds (e.g., Rambouillet and Suffolk) were found to have substantial variation, which should enable breeders to proceed, unencumbered by genetic diversity concerns, with selection strategies that maximize profit. 相似文献

This study aimed to test the hypothesis that maintenance energy requirement (MEm) can be estimated from continuous heat production measurements during a change from a near maintenance feeding level to far below maintenance for two consecutive days. The MEm of eight Hereford steers weighing 286 +/- 5 kg (mean +/- SE) was determined in a balance trial. In addition, during the 10-d collection period, the animals were kept in open-circuit respiration chambers to measure 24-h gas exchange continuously at 10-min intervals. During the balance trial, the animals were fed dried chopped grass twice daily at an estimated level of 1.2 x MEm. After termination of the collection period on the 11th d of the balance trial, the steers were offered 2 kg/d of wheat straw while only gas exchange was measured. Estimates of MEm were derived from heat production (HP) data. The analyses included values of 24-h HP, HP of the nocturnal period (0000 to 0630), HP of the nocturnal period (excluding HP caused by standing) during the grass-feeding period and 24-h HP, nocturnal HP, and nocturnal HP (excluding HP caused by standing) during the straw feeding period. The MEm predicted from estimates of HP measurements were 536 +/- 9, 470 +/- 8, 441 +/- 8, 435 +/- 8, 393 +/- 9, and 373 +/- 9 kJ.kg of BW(-0.75).d(-1), respectively, whereas MEm calculated from data of the balance trial were 416 +/- 9 kJ.kg of BW(-0.75).d(-1). Values predicted for nocturnal HP (excluding HP caused by standing) of grass fed animals, 24-h HP, and nocturnal HP during straw feeding did not differ significantly from MEm. The differences in MEm among animals were reflected by all estimates of HP, whereas the correlation with the 24-h HP during straw feeding reached 0.9 (P = 0.002). Reasons for performing study: To describe the clinical symptoms, treatment, and outcome of meniscal cysts in horses. These structures have not been previously described in the literature as a potential cause of lameness in the horse. Hypothesis: Meniscal cysts are an uncommon condition of the femorotibial joint but can be a significant cause of lameness. Symptoms can be resolved by arthroscopic excision. Methods: Records of horses diagnosed with meniscal cysts and treated by cyst excision and meniscal debridement at 2 surgical practices were reviewed. Clinical outcome was determined by repeat veterinary examination and contact with owner. Results: Seven cases of meniscal cyst were treated with arthroscopic cyst excision and meniscal debridement. Five of 7 horses had lameness attributable to femorotibial joint pathology, while the remaining 2 horses had meniscal cysts found incidentally during diagnostic arthroscopy for the treatment of osteochondritis dissecans of the lateral trochlear ridge of the femur. Five of 6 horses with long‐term follow‐up were sound and a 7th horse was improved 11 months after surgery. Conclusions and potential relevance: Meniscal cysts, while uncommon, can be associated with progressive lameness in the horse. Surgical excision of the cysts results in resolution or improvement of symptoms, without evidence of recurrence on follow‐up examination. 相似文献

An isolate of Eimeria meleagridis Tyzzer, 1927 was obtained by harvesting oocysts from the ceca of a turkey from northwest Arkansas and a pure line was established by infecting birds with a single oocyst. Oocysts were first produced in the ceca of infected birds from 102 to 108 hr after inoculation and were of similar size (mean length X width, 24.9 X 17.0 microm) to those of Eimeria adenoeides Moore and Brown, 1951 and Eimeria gallopavonis Hawkins, 1952. The line was identified as E. meleagridis based upon the development of large schizonts in the midintestine, and small schizonts in the ceca. Two generations of large schizonts were found 48 and 72 hr after infection, and at least two generations of small schizonts were found from 60 to 108 hr after infection. An inoculum of 2 X 10(5) oocysts was found to cause a significant reduction in weight gain from days 0-3 and 0-6 after infection, suggesting that the significance of this species of Eimeria as a pathogen of turkeys should be reassessed. 相似文献

In 1910, H. B. Fantham described the life cycle of a coccidian parasite in birds. Fantham was a parasitologist at Cambridge University in the United Kingdom working for an enquiry into diseases affecting the red grouse. Despite the growing importance of the poultry industry and the realization that coccidiosis was an important disease of the fowl, little further work was carried out in the United Kingdom until coccidiosis research was initiated at the Veterinary Laboratory, Weybridge almost 30 yr later. Further progress depended upon research carried out at academic and agricultural institutions in the United States. E. E. Tyzzer at Harvard University provided the solid foundation upon which our present knowledge of coccidiosis, and the species of Eimeria involved in the disease, is based. Agricultural experiment stations (AESs) throughout the nation played an important role in communicating advances to the agricultural community. W. T. Johnson at Western Washington and, subsequently, Oregon AES made significant contributions to our understanding of the disease, as did C. A. Herrick and coworkers at Wisconsin AES, J. P. Feces were collected from two female and one male Siberian tigers, Panthera tigris altaica. Steroid hormones were extracted from lyophilized feces and quantified by enzyme immunoassay. The fecal contents of estradiol-17beta (E(2)) and testosterone in the females and male, respectively, changed markedly throughout the year. The fecal E(2) contents of females Nos. 179 and 238 increased at 26.4 +/- 8.0 and 28.0 +/- 14.2 day intervals, respectively. However, the fecal contents of progesterone (P(4)) in the female kept alone did not change. In contrast, the other female, which was kept with a male, had increased fecal P(4) contents after copulation. In order to evaluate the variability of encephalomyocarditis virus (EMCV), field isolates originating from different European regions and inducing different clinical pictures in pigs have been molecularly characterised. The regions targeted were the poly(C) tract, a part of the 5'-UTR (360 nucleotides), the Leader gene (201 nucleotides), the complete capsid coding region (2502 nucleotides), the 2A gene (403 nucleotides), the end of the 3D polymerase gene (305 nucleotides) and the 3'-UTR (123 nucleotides). Analyses have also been performed on a virulent field isolate, which had been subjected to serial passages in vivo and in vitro resulting, in the case of the in vitro passaged virus, in attenuation, as demonstrated by animal experiments. The present study shows that different clinical pictures, such as acute fatal myocarditis or reproductive failure, may not only be caused by EMCV isolates which are genetically diverse but also by the same isolate. Thus no correlation could be demonstrated between genotype and clinical disease. However, the European isolate which showed the highest genetic divergence also gave rise to a more complex clinical picture. Despite EMCV having been isolated from cases of acute fatal myocarditis in pigs in certain areas of the world for many years, clinical disease, including a variety of clinical pictures and pathogenicity, has only been recognised in Europe since 1986 and thus it can be considered an emerging disease in this region. These findings, associated with the reported phenotype changes of the virus under environmental changes (passages), along with its wide distribution among vertebrate species (including higher primates), shows the validity of considering EMCV as a potential pathogen for recipients in xenotransplantation. 相似文献

The acquisition of immunity to Eimeria maxima by chicks infected 18 hr after hatch with a single dose of 100 oocysts was investigated. In the first experiment, birds were moved each day to clean cages in order to prevent the possibility of secondary infection resulting from ingestion of oocysts passed in their feces. Immunity was measured at 4 wk of age by calculation of oocyst production following challenge with 500 oocysts or weight gain following challenge with 100,000 oocysts. Large numbers of oocysts were produced by infected birds following challenge, although numbers were significantly less than those from birds that had been reared in the absence of infection (susceptible controls). The weight gain of infected birds following challenge was significantly greater than that of susceptible controls but less than that of unchallenged controls. Thus, only partial protection had been acquired, whether parasite replication or body weight gain was used to assess the extent of immunity development. In a second experiment, acquisition of immunity at 4 wk by chicks infected 18 hr after hatch with 100 oocysts of E. maxima and reared in floor pens in contact with their droppings was investigated. Infected birds produced no oocysts following challenge, and weight gains were not significantly different from the unchallenged controls, which indicates that full immunity had developed by 4 wk. It is concluded that if oocysts of Eimeria species are used to vaccinate day-old chicks, reinfection by oocysts present in the litter is necessary for the establishment of protective immunity. 相似文献
