Rates of leaf extension and senescence were measured over three-day intervals on neighbouring sets of caged and uncaged tillers on several occasions during summer 1983, both when grazing animals were present and also when they were absent. The weather was dry and drought conditions developed as the season advanced. Comparison of data from caged and uncaged tillers indicated that some consistent differences occurred. Loss of information through grazing (affecting 30% of uncaged, larger than average, tillers), contributed to the significantly reduced estimates of leaf extension of ungrazed, uncaged tillers compared with caged tillers. Senescence rates were higher in the open plot than under the cages; old leaves were grazed on only 8% of uncaged tillers so that data loss was less important as a cause of bias. In the drought conditions prevailing, it was thought that caging in the presence of grazing animals resulted in reduced moisture stress and contributed to the above results. Comparisons of caged and uncaged tillers when grazing animals were absent were made to examine the environmental effects of caging and the slight difference in tiller marking procedure. No differences were found in leaf extension rates, though a trend towards higher senescence rates for the uncaged populations was evident. It was concluded that the criteria limiting the use of cages (a period of protection less than the average interval between both leaf appearance and repeated defoliation) should be extended to include a time limit set by the need to avoid bias associated with the environmental effects of caging and cessation of grazing. To define this time limit better in particular circumstances, the extent and direction of bias in the estimation of leaf extension and senescence rates as affected by period of caging, tiller handling procedures, sward and climatic conditions requires further investigation. In the case of vegetative tillers, estimates of leaf extension rates can be made considerably more precise by using tiller size variables (pseudostem length, total green leaf length, leaf width) as covariates. A suggestion for the number of tillers to be measured per transect is given. However, no such increase in precision can be made for estimates of senescence rates.  相似文献

It is believed that milk production is determined by the number and activity of mammary secretory cells. Secretory activity, as assessed by milk volume, depends on secretion of the major osmole in milk, lactose, which is produced by lactose synthase. The amount of either of the two proteins in lactose synthase may regulate milk production. The objective of this study was to determine whether the concentrations in milk of the two components of lactose synthase, α‐lactalbumin (α‐LA) and β1,4‐galactosyltransferase (B4GALT), were related to genetic background, stage of lactation, breed or parity of dairy cows. α‐Lactalbumin and B4GALT concentrations were measured by ELISA and by enzyme assays, respectively, from single milk samples. Two herds with a total of 279 cows were used in the analysis. One herd contained Ayrshire, Brown Swiss, Holstein and Jersey cows; the second herd contained two groups of cows; Holsteins selected for high milk production and Holsteins with 1960s genetics. The α‐LA concentration in milk was greater in Jerseys and Ayrshires than in Holsteins and Brown Swiss. However, no difference in α‐LA concentration was observed in milk from high and low genetic merit cows in the Minnesota herd or among different genetic backgrounds in the Illinois herd. β1,4‐Galactosyltransferase concentrations were similar for all groups that were analyzed. α‐Lactalbumin concentrations were positively correlated with milk protein concentration, milk fat concentration and lactose concentration. β1,4‐Galactosyltransferase concentration in milk exhibited a strong positive correlation with number of days in milk. Although the concentration of B4GALT increased as lactation progressed, the values did not show any correlation with persistency of lactation or late lactation milk production. In conclusion, this survey shows that the two components of lactose synthase are each correlated to protein concentration and individually correlated to the concentration of other milk components and stage of lactation.  相似文献

The objective of this study was to define the patterns of organogenesis and foetal haemodynamics during the normal gestation of healthy agoutis (Dasyprocta prymnolopha) kept in captivity. Thirty pregnant agoutis that ranged in size from small to medium and weighed between 2.5 and 3 kg underwent B‐mode and Doppler ultrasonography for the biometric evaluation of the foetal organs. The foetal aortic blood flow proved to be predominantly systolic, and the measured flow velocity was 78.89 ± 2.95 cm/s, with a maximum pressure gradient of 2.12 ± 0.27 mmHg. The liver was characterized by its large volume, occupying the entire cranial aspect of the abdominal cavity, and it was associated cranially with the diaphragm and caudally with the stomach. The flow velocity in the portal vein was estimated to equal 12.17 ± 2.37 cm/s, with a resistivity index of 0.82 ± 0.05. The gallbladder was centrally located and protruded cranially towards the diaphragm. The spleen was visualized as an elongated structure with tapered cranial and caudal extremities, and the foetal kidneys were visualized bilaterally in the retroperitoneal region, with the right kidney positioned slightly more cranially than the left. The morphological characterization and hemodynamic analysis of the foetal organs of black‐rumped agoutis via B‐mode and Doppler ultrasonography allow determination of the vascular network and of reference values for the blood flow required for perfusing the anatomical elements essential for maintaining the viability of foetuses at different gestational ages.  相似文献

The sperm‐mediated gene transfer method is applicable to transgenesis in many species that use spermatozoa for reproduction recently, which has been shown various results. In the current study, we show that transgenic porcine embryos can be efficiently produced by employing a simple transfection method that uses magnetic nanoparticles (MNPs). The complexes formed between plasmid DNA and MNPs were bounded on ejaculated boar spermatozoa at a higher efficiency compared to methods using DNA alone or lipofection. Using confocal microscopy, rhodamine fluorophore‐labelled MNPs were detected on external surfaces of the spermatozoa membrane, which were bounded on zona pellucida of in vitro maturated oocyte during in vitro fertilization. Electron microscopy revealed that clusters of MNPs were detected in inside of plasma membrane and nucleus of the spermatozoa head. Additionally, we found that magnetofected boar spermatozoa could be fertilized with oocytes in vitro and that the resulting gene of green fluorescent protein was detected in fertilized eggs by genomic PCR analysis. Taken together, these results suggest that MNPs can be used to efficiently introduce a transgene into embryo via spermatozoa.  相似文献

In a range of perennial ryegrass/white clover swards, variation in the surface heights of the grass and clover components, the rates of increase of these surface heights and the specific leaf areas (SLAs) of ryegrass and white clover were described for 1 year. The swards were of an early- or late-flowering (Aurora or Melle respectively) perennial ryegrass variety growing with either a small- or a medium-leaved (Kent or Milkanova) white clover and were either continuously stocked by sheep or continuously stocked apart from a rest period in April-May (Aurora) or May-June (Melle).
The surface heights of grass and clover were not affected by the variety of their companion species, and the surface heights of the two clover varieties were similar. The grass was always taller than the clover, although the magnitude of the difference between the species varied with time of year and the timing of the rest period.
Before the summer solstice the rate of increase in height of grass was greater than that of clover except at cool temperatures (5°C) and warm temperatures (16°C), and in the unrested Melle sward. After the solstice the rates of increase in height, particularly of clover, were lower than the rates seen at similar temperatures before the solstice.
Overall, the SLAs of both clover varieties were greater than those of ryegrass when grown with Aurora but not when grown with Melle, and the SLAs of both species increased during the year. By October the SLAs of both grass varieties were less than those of their companion clovers.
The results are discussed in relation to their implications for the species composition of the swards.  相似文献

AIMS: To determine efficacy of a minimally invasive (MI) surgical approach using a human MI lumbar retractor for canine lumbosacral dorsal laminectomy and partial discectomy and to compare this technique to the standard open surgical (OS) approach.

METHODS: Lumbosacral dorsal laminectomy and partial discectomy was performed on 16 large-breed canine cadavers using either a standard OS (n=8) or MI (n=8) approach. Skin and fascial incision length, procedure time, and intraoperative complications were recorded. Postoperatively specimens were evaluated for laminectomy and discectomy dimensions, and visible damage to the cauda equina and exiting nerve roots.

RESULTS: Median length of skin and fascial incisions in the OS group were longer than in the MI group (p<0.001). Median laminectomy length was similar between both approaches (p=0.234) but width was greater for the MI than OS approach (p=0.002). Both approaches achieved similar partial discectomy width (p=0.279). Overall surgical time was longer for MI approaches compared to OS, with a median of 18.5 (min 15.5, max 21.8) minutes for MI compared to 14.6 (min 13.1, max 16.9) minutes for OS (p=0.001).

CONCLUSIONS: The MI approach reduced incision lengths while retaining comparable laminectomy and discectomy dimensions. For this in vitro model the MI approach required more time to complete, but this difference may not be relevant in clinical cases.

CLINICAL RELEVANCE: Dogs undergoing lumbosacral dorsal laminectomy are commonly large-breed dogs. The traditional open approach requires a large skin incision and soft tissue dissection, especially in overweight animals. A MI approach accomplishing the same surgical result while minimising soft tissue trauma could reduce post-operative pain and recovery time, and may lower wound-related complications. Clinical studies are needed to confirm postoperative benefit and assess operating times in vivo.  相似文献

Two contrasting decision rules for adjusting stock density to control the sward height of continuously grazed grass swards were tested and developed. One rule calculated the adjustment as a percentage of the existing stock density (Percentage Rule); the other related the adjustment to estimates of the bulk density of the grazed horizon and individual animal intake (Absolute Rule). Both decision rules related the adjustments to the change in sward height over the previous week and to the deviation from the target value. The decision rules were tested by continuously grazing lactating ewes and then dry ewes after weaning with a target sward height of 4 cm on 0·33-ha plots with stock density adjusted by varying animal numbers. The Percentage Rule failed to increase the stock density sufficiently rapidly when grass growth was accelerating, leading to increasing deviation in sward height from a 4 cm target value. The Absolute Rule succeeded in controlling sward height once the bulk density term had been correctly adjusted. Pre-emptive adjustment of stock density in anticipation of a change in grass growth rate marginally improved the control that was achieved by the Absolute Rule.  相似文献

A series of twenty-four swards containing different proportions of white clover (0·20-0·25) and perennial ryegrass were created by using different seed mixtures, herbicide applications and previous cutting Frequencies. These swards were used to study the diet of oesophageally-fistulated wether sheep which grazed the various swards for a 30-min period after 1, 2 and 3 weeks of regrowth.
The proportion of white clover in the diet was generally greater than that in the sward. Fifty-seven percent of the variation in the proportion of white clover in the diet could be attributed to the proportion of white clover in the sward. White clover and perennial ryegrass leaf and stem were grazed to the same height and the proportion of white clover in the grazed horizon of the sward explained 83% of the variation in the proportion of white clover in the diet. The proportion of white clover in the diet was greater than the proportion in the grazed horizon of the sward in week 3 of regrowth, but not in weeks 1 and 2, and greater when the proportion of white clover in the grazed horizon was lower than 0·20. Both these observations were interpreted as indicating selection for white clover by the sheep within the grazed horizon.
There was a positive and linear relationship between the depth of the grazed horizon and sward height which, together with the relationship between the proportion of white clover in the grazed horizon and in the diet, would allow the prediction of the proportion of white clover of the diet from the height and the white clover content of the grazed horizon of the sward.  相似文献

The effects of two periods of rest from grazing by sheep (either April-May or May-June) on the composition of swards of both an early-flowering (Aurora) and a late-flowering (Melle) perennial ryegrass variety grown with cither a small-leaved (Kent) or a medium-leaved (Milkanova) white clover variety are described. Compared with a continuously stocked treatment, the early rest period did not affect the mass of clover in the swards (expressed as a proportion of the total) but resulted in a subsequent decrease in its population density. The late rest period doubled clover mass in the swards at the time of cutting but did not affect the subsequent population density of clover. The magnitude of the change in mass during the late rest period depended on ryegrass variety, but not on clover variety. The adverse effect of the early rest period on the subsequent population density of clover was associated with the degree of shading experienced by clover laminae at the end of the rest period. Temperature may also have had an effect here as mean temperatures were cooler during the early rest period than during the late rest period. It is concluded that the effects of the two rest periods on the clover content of the swards were primarily due to timing, and less to inherent differences between ryegrass varieties.  相似文献
