Immunoregulatory feedback between interleukin-1 and glucocorticoid hormones   总被引:56,自引:0,他引:56  
The production and action of immunoregulatory cytokines, including interleukin-1 (IL-1), are inhibited by glucocorticoid hormones in vivo and in vitro. Conversely, glucocorticoid blood levels were increased by factors released by human leukocytes exposed to Newcastle disease virus preparations. This activity was neutralized by an antibody to IL-1. Therefore the capacity of IL-1 to stimulate the pituitary-adrenal axis was tested. Administration of subpyrogenic doses of homogeneous human monocyte-derived IL-1 or the pI 7 form of human recombinant IL-1 to mice and rats increased blood levels of adrenocorticotropic hormone (ACTH) and glucocorticoids. Another monokine, tumor necrosis factor, and the lymphokines IL-2 and gamma-interferon had no such effects when administered in doses equivalent to or higher than those of IL-1. The stimulatory effect of IL-1 on the pituitary-adrenal axis seemed not to be mediated by the secondary release of products from mature T lymphocytes since IL-1 was endocrinologically active when injected into athymic nude mice. These results strongly support the existence of an immunoregulatory feedback circuit in which IL-1 acts as an afferent and glucocorticoid as an efferent hormonal signal.  相似文献

Most people hold beliefs about personality characteristics typical of members of their own and others' cultures. These perceptions of national character may be generalizations from personal experience, stereotypes with a "kernel of truth," or inaccurate stereotypes. We obtained national character ratings of 3989 people from 49 cultures and compared them with the average personality scores of culture members assessed by observer ratings and self-reports. National character ratings were reliable but did not converge with assessed traits. Perceptions of national character thus appear to be unfounded stereotypes that may serve the function of maintaining a national identity.  相似文献

Radio waves from the sun were detected 50 years ago, but the microwave detection of other single solar-type stars has remained a challenge. Here, the discovery of four solar-type radio stars is reported. These "solar twin" G stars are radio sources up to 3000 times stronger than the quiet sun. The microwaves most likely originate from a large number of relativistic electrons, possibly produced along with coronal heating, a process that is not understood. Two of the stars are younger than the sun and rotate more rapidly; the dynamo process in the stellar interior is therefore presumably more vigorous, resulting in enhanced coronal activity. One of the detections, however, is an old, metal-deficient G dwarf.  相似文献

To act as guides in the RNA interference (RNAi) pathway, small interfering RNAs (siRNAs) must be unwound into their component strands, then assembled with proteins to form the RNA-induced silencing complex (RISC), which catalyzes target messenger RNA cleavage. Thermodynamic differences in the base-pairing stabilities of the 5' ends of the two approximately 21-nucleotide siRNA strands determine which siRNA strand is assembled into the RISC. We show that in Drosophila, the orientation of the Dicer-2/R2D2 protein heterodimer on the siRNA duplex determines which siRNA strand associates with the core RISC protein Argonaute 2. R2D2 binds the siRNA end with the greatest double-stranded character, thereby orienting the heterodimer on the siRNA duplex. Strong R2D2 binding requires a 5'-phosphate on the siRNA strand that is excluded from the RISC. Thus, R2D2 is both a protein sensor for siRNA thermodynamic asymmetry and a licensing factor for entry of authentic siRNAs into the RNAi pathway.  相似文献

In the present work, the aroma profiles of wines elaborated from sound and sour rot-infected grapes as raw material have been studied by sensory analysis, gas chromatography-olfactometry (GC-O), and gas chromatography-mass spectrometry (GC-MS), with the aim of determining the odor volatiles most likely associated with this disease. The effect of sour rot was tested in monovarietal wines produced with the Portuguese red grape variety Trincadeira and in blends of Cabernet Sauvignon and sour rotten Trincadeira grapes. Wines produced from damaged berries exhibited clear honey-like notes not evoked by healthy samples. Ethyl phenylacetate (EPhA) and phenylacetic acid (PAA), both exhibiting sweet honey-like aromas, emerged as key aroma compounds of sour rotten wines. Their levels were 1 order of magnitude above those found in controls and reached 304 and 1668 μg L(-1) of EPhA and PAA, respectively, well above the corresponding odor thresholds. Levels of γ-nonalactone also increased by a factor 3 in sour rot samples. Results also suggest that sour rot exerts a great effect on the secondary metabolism of yeast, decreasing the levels of volatiles related to fatty acids and amino acid synthesis. The highest levels of γ-decalactone of up to 405 μg L(-1) were also found in all of the samples, suggesting that this could be a relevant aroma compound in Trincadeira wine aroma.  相似文献

The Siberian moth, Dendrolimus sibiricus, Tschtv. is the most harmful defoliator of coniferous forests in North Asia. The pest has already spread over the Urals and continues moving westwards. Recently, it has been recommended for quarantine in member countries by European and Mediterranean Plant Protection Organization (EPPO). The performances of the pest on coniferous species planted in Europe were assessed on a range of potted trees corresponding to the spectrum of economically important conifers in the EU: European larch Larix decidua, Norway spruce Picea abies, Scots pine Pinus sylvestris, European black pine Pinus nigra, and the North American species: Douglas fir Pseudotsuga menziesii and grand fir Abies grandis. Larvae showed a potential to survive and complete the development on all these host tree species. Favorable hosts were grand fir, European larch, and Douglas fir that allowed higher survival, better larval development, and as a result, yielded heavier pupae and adult moths with higher longevity. Black pine was a poor host but, however, could still support larval and pupal development. Norway spruce and Scots pine had an intermediate behavior. If accidentally introduced to Europe, the Siberian moth may become especially damaging in forest stands predominated by European larch and by the North American firs. Norway spruce and especially the two-needle pines will be less prone to intensive defoliation by this species. The fact that the pest may damage the range of economically important coniferous species should be taken into account in the pest risk assessment for Europe and also for North America where the Siberian moth occurrence is considered likely.  相似文献

About 90% of the wildland fires occurred in Southern Europe are caused by human activities. In spite of these figures, the human factor hardly ever appears in the definition of operational fire risk systems due to the difficulty of characterising it. This paper describes two spatially explicit models that predict the probability of fire occurrence due to human causes for their integration into a comprehensive fire risk–mapping methodology. A logistic regression technique at 1 × 1 km grid resolution has been used to obtain these models in the region of Madrid, a highly populated area in the centre of Spain. Socio-economic data were used as predictive variables to spatially represent anthropogenic factors related to fire risk. Historical fire occurrence from 2000 to 2005 was used as the response variable. In order to analyse the effects of the spatial accuracy of the response variable on the model performance (significant variables and classification accuracy), two different models were defined. In the first model, fire ignition points (x, y coordinates) were used as response variable. This model was compared with another one (Kernel model) where the response variable was the density of ignition points and was obtained through a kernel density interpolation technique from fire ignition points randomly located within a 10 × 10 km grid, which is the standard spatial reference unit established by the Spanish Ministry of Environment, Rural and Marine Affairs to report fire location in the national official statistics. Validation of both models was accomplished using an independent set of fire ignition points (years 2006–2007). For the validation, we used the area under the curve (AUC) obtained by a receiver-operating system. The first model performs slightly better with a value of AUC of 0.70 as opposed to 0.67 for the Kernel model. Wildland–urban interface was selected by both models with high relative importance.  相似文献

Our main goal was to investigate if robust chemical fingerprints could be developed for three Argentinean red wines based on organic, inorganic, and isotopic patterns, in relation to the regional soil composition. Soils and wines from three regions (Mendoza, San Juan, and Co?rdoba) and three varieties (Cabernet Sauvignon, Malbec, and Syrah) were collected. The phenolic profile was determined by HPLC-MS/MS and multielemental composition by ICP-MS; (87)Sr/(86)Sr and δ(13)C were determined by TIMS and IRMS, respectively. Chemometrics allowed robust differentiation between regions, wine varieties, and the same variety from different regions. Among phenolic compounds, resveratrol concentration was the most useful marker for wine differentiation, whereas Mg, K/Rb, Ca/Sr, and (87)Sr/(86)Sr were the main inorganic and isotopic parameters selected. Generalized Procrustes analysis (GPA) using two studied matrices (wine and soil) shows consensus between them and clear differences between studied areas. Finally, we applied a canonical correlation analysis, demonstrating significant correlation (r = 0.99; p < 0.001) between soil and wine composition. To our knowledge this is the first report combining independent variables, constructing a fingerprint including elemental composition, isotopic, and polyphenol patterns to differentiate wines, matching part of this fingerprint with the soil provenance.  相似文献

Isolation and identification of the inhibitors of butyrylcholinesterase (BChE), obtained from the extracts of roots and fruits of Angelica archangelica L., are reported. Our results confirmed the weak inhibitory effect of Angelica roots on acetylcholinesterase activity. BChE inhibition was much more pronounced at a concentration of 100 μg/mL for hexane extracts and attained a higher rate than 50%. The TLC bioautography guided fractionation and spectroscopic analysis led to the isolation and identification of imperatorin from the fruit's hexane extract and of heraclenol-2'-O-angelate from the root's hexane extract. Both compounds showed significant BChE inhibition activity with IC(50) = 14.4 ± 3.2 μM and IC(50) = 7.5 ± 1.8 μM, respectively. Only C8-substituted and C5-unsubstituted furanocoumarins were active, which could supply information about the initial structures of specific BChE inhibitors.  相似文献

The influence of vegetation cover on soil hydrological properties and its response to the impact of different fire intensities, in a Mediterranean forest environment, has been evaluated. The study was carried out in the Permanent Experimental Field Station of La Concordia (Llíria–Valencia, Spain), on a set of nine erosion plots (4 × 20 m2). The Station is located on a calcareous hillside S–SE oriented, with soils of Rendzic Leptosol type and supporting Mediterranean shrubland vegetation. All runoff generated and sediment produced in every rain event was collected from each plot. The set up includes a system of sensors for the continuous monitoring of climatic parameters (air temperature and humidity, rain volume, intensity, etc.).In June 1995, a set of experimental fires was carried out to the Station. Three of the plots were burned with high intensity fire, three with moderate intensity and the remaining were left unaltered. Soil water content and water retention capacity (WRC) were measured in the different plots and in two different vegetation covers: under canopy (UC) and in bare soil (BS). The pF curves were also obtained for each fire treatment.A year after the fires (June 1995–June 1996), great differences, reaching 77.15%, in runoff generation between fire treatments and the control plots were observed.No significant differences were detected on water retention capacity between soils UC and BS in the burned plots. However, these differences appeared in the control plots, giving UC and BS values of 13% and 18%, respectively. Plots corresponding to the high intensity fire treatment showed values of WRC significantly higher than those of the moderate intensity and of the control treatments.The pF curves show that the values of water volume, at the different pressure points studied, were slightly greater on UC soil. Values obtained for BS samples are higher in the fire treatments, showing significant differences in respect to the control plots at pF 1 and 2. These differences were also observed for UC soil, but in this case at pF 2, 2.5 and 4.2.  相似文献
