Summary Random amplified polymorphic DNA (RAPD) analysis was carried out to evaluate polymorphism and genetic similarity between 39 Mediterranean mandarin genotypes. One hundred eleven amplification products were identified using 21 random primers. An average of 2.2 RAPD markers was obtained for each primer, corresponding to 42% of the amplification products. Genotype-specific RAPD markers were also found, mainly in known hybrids. UPGMA cluster analysis revealed the low level of genetic variation between accessions of Mediterranean mandarins, whereas their hybrids with other Citrus species showed greater genetic dissimilarity. Twenty accessions yielded very similar patterns, suggesting either that they could be a single clone, or that the technique was not able to detect genomic variation. However, for the other specimens genetic polymorphism can easily be detected by RAPD, although the genetic variation between accessions was quite low. The large number of hybrids and the low polymorphism between accessions support the hypothesis that Mediterranean mandarins are all true hybrid of Common mandarins (Citrus reticulata Blanco).  相似文献

This work was performed in the Rio de Janeiro State, Brazil, with the objective to evaluate the nutritional status of yellow passion fruit plants along different phenological stages, using the DRIS method. Fifty-four passion fruit cultivated areas with an annual yield productivity ranging from 6.95 to 33.8 t ha?1 year?1 and average productivity of 16.9 t ha?1 year?1 were selected in the region. The contents of nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), sulfur (S), chlorine (Cl), iron (Fe), zinc (Zn), manganese (Mn), and boron (B) were evaluated. The reference standards were established (mean and variation coefficient) for the values of the nutrient concentration ratios, two by two, on samples from high yield productivity plantations and analyzed by the DRIS index of nutrients for the medium and low yield productivity areas. The established standards differed according with the phenological stage of the culture. In a general way, the mean content of the evaluated nutrients did not differed between the two productivity levels into each phonological stage. There was a difference for the Nutritional Limitation Order between different phenological stages of yellow passion fruit plants. The most negative DRIS indexes and the highest absolute values for the Average Nutritional Balance on yellow passion fruit plants in the region, were found for potassium in May, phosphorus in October and iron in January.  相似文献

Carpotroche brasiliensis is a dioecious tree species native of the Brazilian Atlantic rainforest. Due to medical and industrial use of the oil extracted from its seeds, C. brasiliensis has a great potential for cultivation as non-timber forest product in agroforestry systems. This study was conducted with the objective to analyze the leaf dimensions of male and female adult trees and seedlings of C. brasiliensis. Two hypotheses were tested: (a) leaf dimensions do not differ between male and female adult genotypes; and (b) it is possible to develop single regression models for predicting leaf area (LA) from dimensional variables encompassing male and female adult genotypes and seedlings. LA, leaf length (L) and maximum leaf width (W) were measured in leaves collected from seven male and seven female adult genotypes and three seedling lots. The feasibility of using a single model for leaves of males and females, and seedlings and adults, was tested by analysis of covariance (ANCOVA). The prediction errors (PE) for each of the regression models were calculated from the cross-validation method. The average values of L, W and LA were, respectively, 136, 142 and 457 % higher in adults than in seedlings, and the average values of leaf shapes (L:W) of seedlings were intermediate between the average values of L:W of adult males and females. The average values of L did not differ between adult males and females, but significant differences were observed between males and females for W, LA and L:W (both p < 0.01, nested ANOVA). The mean L:W values of adult males and females, and seedlings, indicate that leaf shape should be used as a criterion for sex differentiation in this species. It was not possible to develop single models encompassing adult males and females, and seedlings; but high accurate predictive models of LA from L × W measurements were developed for adult males (R2 = 0.98, PE = 0.69, n = 350), adult females (R2 = 0.98, PE = 0.01, n = 350), and seedlings (R2 = 0.99, PE = 6.80, n = 150).  相似文献

Since the myrtle rust pathogen (Austropuccinia psidii) was first reported (as Puccinia psidii) in Brazil on guava (Psidium guajava) in 1884, it has been found infecting diverse myrtaceous species. Because Apsidii has recently spread rapidly worldwide with an extensive host range, genetic and genotypic diversities were evaluated within and among Apsidii populations in its putative native range and other areas of myrtle rust emergence in the Americas and Hawaii. Microsatellite markers revealed several unique multilocus genotypes (MLGs), which grouped isolates into nine distinct genetic clusters [C1–C9 comprising C1: from diverse hosts from Costa Rica, Jamaica, Mexico, Puerto Rico, and USA‐Hawaii, and USA‐California; C2: from eucalypts (Eucalyptus spp.) in Brazil/Uruguay and rose apple (Syzygium jambos) in Brazil; C3: from eucalypts in Brazil; C4: from diverse hosts in USA‐Florida; C5: from Java plum (Syzygium cumini) in Brazil; C6: from guava and Brazilian guava (Psidium guineense) in Brazil; C7: from pitanga (Eugenia uniflora) in Brazil; C8: from allspice (Pimenta dioica) in Jamaica and sweet flower (Myrrhinium atropurpureum) in Uruguay; C9: from jabuticaba (Myrciaria cauliflora) in Brazil]. The C1 cluster, which included a single MLG infecting diverse host in many geographic regions, and the closely related C4 cluster are considered as a “Pandemic biotype,” associated with myrtle rust emergence in Central America, the Caribbean, USA‐Florida, USA‐Hawaii, Australia, China‐Hainan, New Caledonia, Indonesia and Colombia. Based on 19 bioclimatic variables and documented occurrences of Apsidii contrasted with reduced sets of specific genetic clusters (subnetworks, considered as biotypes), maximum entropy bioclimatic modelling was used to predict geographic locations with suitable climate for A. psidii which are at risk from invasion. The genetic diversity of Apsidii throughout the Americas and Hawaii demonstrates the importance of recognizing biotypes when assessing the invasive threats posed by Apsidii around the globe.  相似文献

Bacterial leaf blight of eucalypt, caused by Xanthomonas axonopodis, occurs mainly in forest nurseries, but also affects young plants in the field. Avoidance of leaf wetness for long periods of time over the whole nursery and using drip irrigation for clonal hedges are, currently, the most effective measures for controlling bacterial leaf blight on eucalypt. However, as these techniques are very costly, alternative management measures are required. We hypothesized that the management of mineral nutrients could reduce disease severity on eucalypts. To examine this hypothesis, rooted cuttings of Eucalyptus urophylla × E. grandis were irrigated with nutrient solutions containing different concentrations of copper (Cu), boron (Bo), potassium (K), calcium (Ca) and nitrogen (N) before inoculation with X. axonopodis 30 days after starting the nutrient solution treatments. Concentration of K higher than that recommended, commonly used in miniclonal hedges, reduced the severity of bacterial leaf blight. However, disease severity increased with an increasing of N/K ratio. The management of mineral nutrition solution, especially K, has great potential for integrated disease management of X. axonopodis in eucalypt nurseries.  相似文献

Methods based on RGB (Red, Green, Blue) image segmentation may emerge as a new and low-cost method for estimation the quality of tree seedlings. However, the vast number of indexes based on the use of the RGB image segmentation and the lack of references in the literature still hinder the widespread use of this technology. Thus, we conducted a study aiming to test the relationships between methods based on absorbance and reflectance, widely used for the estimation of chlorophyll contents and physiological status of trees, and ten indexes based on RGB component analysis. We used leaves of five tropical tree species, belonging to different botanical families. Leaf absorbance was measured using the handheld chlorophyll meter SPAD-502, reflectance was measured using a spectrometer and the RGB indices were obtained from digitalized images of the leaves using a flatbed scanner. Modified linear regression models including all five species were used to relate RGB indices to absorbance and reflectance indices. Data collected from leaves of seedlings of five tropical tree species indicated that digital image processing technology can be a useful and rapid nondestructive method for assessment of physiological status of tree seedlings at nursery stage. Among the RGB indexes tested in this study the R, 2R*(G???B)/(G?+?B) and 2G*(G???B)/(G?+?B) are the most promising for analysis the tropical seedlings physiological status and quality.

This study evaluated the efficiency of taper functions and the application of mixed-effect modelling for diameter estimation along the stems of Tectona grandis. We sampled 266 trees of Tectona grandis, measuring the diameter at relative heights for volume determination, grouping the data according to three form-factor classes. Six taper functions were fitted, selecting the function with better fit performance. Six taper functions were fitted, selecting the function with better fit performance. The selected function was fitted in its basic formulation, and with the mixed non-linear modelling technique in different scenarios, and for the stem stratified in three portions of the total height. The precision and selection of the adjusted models were evaluated regarding the coefficient of determination, standard error of estimate, the Akaike information criterion, bias, quadratic error and absolute bias. According to the statistical criteria used, the model of Kozak was selected for the adjustments. For diameter estimation, the scenario with two coefficients as random effects provided an accuracy increase of 11.91%, and the mixed non-linear modelling better estimated the stem diameter for the stratified stems. In conclusion, the model of Kozak can be used to describe the stem shape of Tectona grandis, and the mixed-effect non-linear model approach was the best technique to estimate diameter along the stem of Tectona grandis.  相似文献

Three diagrammatic grading keys were designed for the assessment of the severity of late blight (caused by Phytophthora infestans ) in tomato leaves. Simplified and broad keys considered, respectively, six (3, 12, 22, 40, 60 and 77%) and eight (3, 6, 12, 22, 40, 60, 77 and 90%) levels of disease severity, whilst a modified key based on a previous proposal for potato late blight considered six levels (1, 5, 10, 16, 32 and 50%). The keys were validated by 24 evaluators who assessed digital images of tomato leaves exhibiting different areas with lesions. Evaluator errors were compared using a mixed model in which evaluators were considered as random effects and the keys and evaluations as fixed effects. The accuracy and precision of the evaluators were compared by simple linear regression between the estimated and actual values of disease severity. The repeatability of evaluators was assessed using Pearson's correlation coefficient. There was significant ( P  ≤   0·001) variability amongst the errors made by evaluators, although the precision of each of the three keys was high with a coefficient of determination (R2) of 0·96, 0·93 and 0·83 for the simplified, broad and modified key, respectively. Repeatability of estimations amongst the evaluators was adequate (correlation coefficients of 0·91, 0·91 and 0·90 for the three keys, respectively). The simplified and broad keys resulted in higher precision and accuracy for the estimation of severity than did the modified key. Since the simplified key considers a smaller number of disease severity levels, its use is recommended in the assessment of late blight in tomato leaves.  相似文献
