Culture media formulation for Arthrospira maxima (formerly designated as Spirulina maxima) production is a constraining factor. Use of synthetic media could be expensive because most of the commercial strains require salinity conditions higher than 20 g/L. In this study, a traditional, pre‐Hispanic mineral resource, known as “tequesquite,” was used as the base for the formulation of an alternative and low‐cost culture medium. Tequesquite is available from outcrop areas located in the basin of the former Lake Texcoco, where A. maxima thrived in pre‐Hispanic times. The effect on the biomass production of different tequesquite‐based formulations, varying N, P, and , was tested and compared with results obtained with Zarrouk's medium (ZM). Growth of biomass in some alternative formulations showed no significant differences compared with ZM. Production in a bubble‐column photobioreactor of the alternative medium reached a maximum of X = 2570.4 mg/L dry weight, compared with X = 2931.4 mg/L dry weight obtained with ZM. The proposed tequesquite‐medium formulation has equivalent N and P concentrations as ZM, but only 50% of NaHCO3, thus reducing the medium's cost. In addition, it avoids the use of seawater or NaCl to provide the salinity condition required for the adequate growth of Arthrospira.  相似文献

Mediterranean wooded grasslands that emerge from silvopastoral activities are multifunctional systems that result in high biodiversity and offer ecosystem services such as forage production and soil carbon sequestration. During 3 years, ten grazed wooded grassland fields were studied in the Berchidda–Monti long-term observatory, located in NE Sardinia, Italy, with the aim of exploring the synergies and trade-offs between biodiversity and selected ecosystem services. Positions below and outside the canopy of three cork oak trees in each field were randomly selected to compare seasonal pasture production, pasture utilization rate by animals, botanical composition, biodiversity indicators (Shannon index and plant species richness) and soil organic carbon. In autumn, dry matter production of pasture was similar in the two positions; in two winters out of three it was greater below the trees than outside, and in spring it was greater outside than below the trees. While plant species richness and Shannon index were not significantly influenced by the position, the overall wooded grassland plant species richness was 31% higher than that outside of the tree crown. The soil organic carbon content in the 0–40-cm soil layer was also higher below the trees. Our findings highlight that if the main purpose of the wooded grasslands is to provide forage for grazing animals rather than conserving and/or enhancing plant diversity and soil fertility, the presence of trees constrains the overall forage productivity, although the greater forage availability in winter under the trees can contribute to improve the seasonal distribution of forage production.  相似文献

A method for the determination of free and total sulfur dioxide in wines, based on the use of an optical sensor that employs a dichlorobis(diphenylphosphino)methane dipalladium I complex [Pd(2)(dppm)(2)Cl(2)] immobilized in a PVC membrane plasticized with o-nitrophenyloctylether (o-NPOE) is described. A sensing membrane [4.2% Pd(2)(dppm)(2)Cl(2), 20.8% PVC, and 75% o-NPOE] was adapted to the tip of a bifurcated optical fiber bundle to perform reflectance measurements at 550 nm. The detection system consisted of two cells (40 mL), which hold the sample solution (plus reagents) and the optical sensor, respectively. For the determination of free SO(2), a wine sample was mixed with H(2)SO(4) solution in the sample cell, into which N(2) was bubbled, providing mixing of the solutions and conducting the SO(2) formed toward the detection cell. For determination of total SO(2), a KOH solution was mixed with the wine in the sample cell. Afterward, an H(2)SO(4) solution was added to the cell, and then N(2) was bubbled to conclude the measurement. Linear responses up to 50 and 150 mg L(-1) were obtained for free and total SO(2), with detection limits of 0.37 and 0.70 mg L(-1), respectively. The repeatability of the method was evaluated by carrying out 10 measurements using a single wine sample, providing relative standard deviation values of 2.2 and 2.5% for free and total SO(2), respectively. The sensing membrane prepared from 10 muL of the cocktail solution lasted for 80 measurements, whereas those prepared from 200 muL can be used for 250 measurements. The method was applied to free and total SO(2) determination in wines, and the results did not show significant difference from those obtained with the Ripper reference method at a confidence level of 95%.  相似文献

ABSTRACT

A greenhouse experiment was performed to evaluate macronutrients and boron deficiencies on vitexin bioproduction by sweet passion fruit leaves. Sand irrigated with nutrient solution was used as substrate in a complete randomized design, with eight treatments: 1) complete, 2) nitrogen-deficient(-N), 3) phosphorus-deficient(-P), 4) potassium-deficient(-K), 5) calcium-deficient(-Ca), 6) magnesium-deficient(-Mg), 7) sulfur-deficient(-S), and 8) boron-deficient(-B). After thirty days, the fourth fully expanded leaves were harvested. Under deficiency treatments, leaf dry matter concentrations of N, P, K, Ca, Mg, S, and B were 52, 53, 62, 76, 69, 31, and 80% lower than in complete treatment, respectively. Nitrogen, P, and K deficiency increased vitexin leaf concentration by 46, 16, and 18%, although Ca and B deficiencies decreased vitexin concentration by 22 and 33%, respectively, when compared to complete treatment. Magnesium and S deficiencies had no significant effect on vitexin concentration. In complete treatment, the concentration of nutrients and vitexin, in leaf dry matter were: 43.4 g kg? 1 of N, 2.47 g kg? 1 of P, 27.4 g kg? 1 of K, 15.6 g kg? 1 of Ca, 3.8 g kg? 1 of Mg, 5.28 g kg? 1 of S, 64 mg kg? 1 of B, and 5.57 mg kg? 1 of vitexin.  相似文献

Milk production in Malawi is stagnating while steadily increasing in neighboring countries. We performed a literature review to identify factors negatively affecting milk production in Malawi and propose potential solutions to overcome the particular constraints. We structured the analysis along the value chain with three major components: upstream providers, on-farm production, and downstream providers. It is shown that the literature strongly focuses on on-farm constraints and most prominently on animal health, breed choice, and milk yield. The literature research did not reveal any reasons specific to Malawi that would hinder an increase in milk production as experienced in neighboring countries. We suggest a focus on crossbreeds accompanied by local breeding programs, which would reduce dependence on imported semen and strengthen the currently unreliable artificial insemination services. In addition, crossbreeds may be better suited than pure breeds for current feeding practices and climate conditions and thus come close to reaching their genetic potential. We conclude that cost advantages of crossbreeds clearly compensate for the lower revenues compared with exotic breeds such as Holstein-Friesian or Jersey.  相似文献

The quality of the microalgae provided on Paracentrotus lividus larvae rearing is a primordial factor having a direct (nutritional properties) and indirect (water quality) impact on growth, competence and survival. Skeletonema costatum is a diatom commonly used in the bivalve cultivation. However, the use of this diatom in P. lividus larval cultivations is poorly known. The Rhodomonas spp. is a microalgae commonly used in sea urchin larvae culture. Three different diets were tested on P. lividus larvae and post‐larvae cultivation (D1—Rhodomonas marina, D2—S. costatum, D3—mixture of both algae). Larvae fed with the D2 diet (55.8%) and D3 (39.9%) had a survival at 15 DAH higher than D1 (5.5%). The low survival in D1 could be due to the higher microbiological load on microalgae (Vibrio alginolyticus and V. pectenicide). Larvae fed with S. costatum (D2) showed a lower development than other diets. The competency index was lower for larvae fed with the D2. These results show that microalgae diversified diets contribute to a better development of P. lividus larvae. During the settlement and post‐settlement phase, there was also a lower growth of the sea urchin fed with the D2 and a higher survival for D3.  相似文献

ABSTRACT: In dogs Helicobacter spp. are found in all gastric regions usually localized in the surface mucus, gastric glands and parietal cells. The aim of this study was to detail the distribution of Helicobacter spp. in the fundic mucosa of asymptomatic Beagle dogs and their intracellular localization within parietal cells, in order to evaluate species-specific pathogenetic effects on gastric cells. The presence of Helicobacter spp. was investigated by immunohistochemistry, TEM, and PCR in the fundic mucosa of six Beagle dogs. Helicobacter spp. were found in all dogs examined, and H. bizzozeronii and H. felis were identified by PCR and confirmed by TEM. In the lumen of the fundic glands, co-localization was common. H. bizzozeronii was present in larger numbers than H. felis in both intraluminal and intraparietal localization. The amounts of H. bizzozeronii were similar in superficial and basal portions of the glands. H. felis was predominantly localized in the superficial portions of gastric glands but almost absent from the base. Within parietal cells, most Helicobacter organisms were intracanalicular, but intact and degenerate Helicobacter organisms were also visualized free in the cytoplasm or in secondary lysosomes. No specific degenerative lesions were found in infected parietal cells. Helicobacter organisms were also observed within macrophages in the lamina propria. In conclusion, there is a differential distribution of H. bizzozeronii and H. felis in the fundic mucosa of Beagle dogs, and their intracellular localization in parietal cells and macrophages suggests novel pathogenic scenarios for the development of immune response and maintenance of chronic gastritis in dogs.  相似文献

Many traditional Chinese medicine (TCM) decoctions are proven to have multiple functions in animal production. These decoctions are seldom recognized by the international scientific community because the mechanisms of action are not clearly elucidated. According to TCM theory, Cortex Phellodendri (COP), Rhizoma Atractylodes (RA), Agastache Rugosa (AR), and Gypsum Fibrosum (GF) can be used to formulate a medicinal compound that prevents or cures animal disease caused by heat stress. The aim of this research was to study the regulatory functions of the active components of TCM and to elucidate the effects of different TCM decoctions on antioxidant activity and lipid peroxide content, using in vitro and in vivo models of heat stress. For in vitro experiments, intestinal crypt-like epithelial cell line-6 (IEC-6) cells were employed to evaluate the effects of the active components of COP, RA, AR, and GF. For in vivo experiments, forty-eight 2-mo-old Chinese experimental mini-pigs (7.20 ± 0.02 kg) were randomly assigned to 4 groups: a normal-temperature group (NTG); a high-temperature group (HTG); HTG treated with COP, RA, AR, and GF (1:1:1:1, TCM1); and HTG treated with COP, RA, AR, and GF (1:1:1:0.5, TCM2). Results showed that the active components of the COP, RA, AR, and GF increased (P < 0.05) the proliferation and viability of heat-stressed IEC-6 cells and that the most effective treatment doses of COP alkaloid, RA Aetherolea, Herba Agastachis Aetherolea, and GF water extract were 200, 100, 100, and 200 μg/mL, respectively. All 4 active components increased (P < 0.05) superoxide dismutase, glutathione peroxidase activities, and glutathione content, and decreased (P < 0.05) malondialdehyde content with respect to the heat-stressed group to concentrations similar to those seen in NTG. In vivo experiments demonstrated that TCM1 and TCM2 improved (P < 0.05) the poor growth performance seen in HTG pigs. The superoxide dismutase, glutathione peroxidase activities, and malondialdehyde content in porcine jejunum treated with TCM1 and TCM2 were not different (P > 0.05) from those seen in the NTG and were better (P < 0.05) than results seen in the HTG. Overall, it appeared that TCM2 was more effective than TCM1 in ameliorating the effects of heat stress in pigs. In conclusion, this study revealed that the active components of common TCM decoctions have antioxidant functions.  相似文献

Relatively few studies have documented the impacts of afforestation, particularly production forestry, on belowground carbon dioxide (CO2) effluxes to the atmosphere. We evaluated the changes in the soil CO2 efflux—a proxy for soil respiration (Rs)—for three years following a native grassland conversion to eucalypt plantations in southern Brazil where minimum tillage during site preparation created two distinct soil zones, within planting row (W) and between-row (B). We used root-exclusion and carbon (C)- isotopic approaches to distinguish Rs components (heterotrophic-Rh and autotrophic-Ra respirations), and a CO2 profile tube (1-m deep) to determine the concentration ([CO2]) and isotopic C signature of soil CO213[CO2]). The soil CO2 efflux in the afforested site averaged 0.37 g CO2 m?2 h?1, which was 56% lower than the soil CO2 efflux in the grassland. The δ13CO2 in the afforested site ranged from ? 14.1‰ to ? 29.4‰, indicating a greater contribution of eucalypt-derived respiration (both Rh and Ra) over time. Higher soil CO2 efflux and lower [CO2] were observed in W than B, indicating that soil preparation creates two distinct soil functional zones with respect to C cycling. The [CO2] and δ13[CO2] decreased in both zonal positions with eucalypt stand development. Although the equilibrium in C fluxes and pools across multiple rotations is needed to fully account for the feedback of eucalypt planted forests to climate change, we provide quantitative information on soil CO2 dynamics after afforestation and show how soil preparation can leverage the feedback of planted forests to climate change.
