Aquaculture research has focused on polysaccharides as they are among the most promising new-generation immunostimulant used to control aquatic diseases. The aim of this study was to investigate the immunomodulatory effects of fermented wheat bran polysaccharides (FWBPs) on juvenile common carp (Cyprinus carpio). Carps were fed different FWBP amounts (0%, 0.1%, 0.2%, and 0.4%) for 8 weeks and then their skin mucus and serum immune parameters, hepatopancreas antioxidant indicators, and immune-related gene expression in the intestines, kidneys, and spleen were measured. The skin mucus IgM levels significantly increased in 0.1% FWBP group, but decreased in 0.4% FWBP group. The skin mucus protease and the serum alkaline phosphatase activities increased significantly in the 0.2%, 0.1%, and 0.4% FWBP groups, respectively. The serum total Ig levels increased noticeably in the 0.1% and 0.2% FWBP groups. The highest and lowest serum lysozyme activities were observed in the 0.1% and 0.4% FWBP groups, respectively. The hepatopancreatic total superoxide dismutase activity was higher in the 0.1% FWBP group than in the control. The malondialdehyde levels decreased significantly in the 0.2% and 0.4% groups. The intestinal mRNA levels of the LZM-C and IL-10 genes were significantly higher in the 0.2% than in the 0.4% FWBP group; TNF-α was significantly upregulated in the 0.1% group. The gene expression in the kidneys did not differ significantly among the treatments, except for a significant increase in the IL-10 expression in the 0.1% treatment. Significantly elevated expression of LZM-C in 0.2% group and IL-10 in 0.1% group was observed in the spleens. TNF-α and IL-1β were significantly downregulated in the 0.4% group. These results suggest that FWBPs could be used as immunostimulant feed additives in common carp cultures.

In order to investigate the relationship between the PD-1 pathway and impairment of immune responses with the CSFV infection, the mRNA expression of PD-1 and its ligands were evaluated by quantitative polymerase chain reaction (qPCR) during artificial CSFV infection. Simultaneously, expression of IL-2 and IL-10 mRNA were detected. The T cell proliferation and CSFV load in plasma were also measured. Results showed that the expression of PD-1 and its ligands mRNA were significantly increased (p < 0.01) in PBMC from 3 to 7 days post infection (dpi). Meanwhile the level of IL-10 was up-regulated (p < 0.01). The IL-2 mRNA was not obviously changed but it is significantly increased from 14 dpi. The T cell proliferation was notably decreased at 7 dpi. The CSFV load was also increased in plasma. Overall, our results suggest that the expression of PD-1 and its ligands were up-regulated and probably correlated with immune inhibition during acute CSFV infection. 相似文献
Riemerella anatipestifer causes epizootic infectious disease in poultry and serious economic losses especially to the duck industry. However, little is known regarding the molecular basis of its pathogenesis. The ability to acquire iron under low-iron conditions is related to the virulence of a variety of bacterial pathogens. In this study, a sip (Riean_1281) deletion mutant CH3Δsip was constructed and characterized for iron-limited growth, biofilm formation, and pathogenicity to ducklings. Results showed that siderophore-interacting protein (SIP) was involved in iron utilization and the sip deletion significantly reduced biofilm formation and adherence to and invasion of Vero cells. In addition, the sip gene was absent in 1 of 24 (4.17%) virulent strains and 2 of 3 (66.7%) avirulent strains of R. anatipestifer, and the sip gene from six R. anatipestifer strains, which belong to serotypes 1, 2, and 10, respectively, shared 100% amino acid identities to those of R. anatipestifer strains DSM15868 and RA-GD. These results suggested that siderophore-mediated iron acquisition may be an important iron-uptake pathway in R. anatipestifer. Animal experiments indicated that the median lethal dose of the CH3Δsip mutant in ducklings was about 35-fold higher than that of the wild-type CH3 strain. Thus, our results demonstrated that R. anatipestifer SIP was involved in iron acquisition and necessary for its optimal virulence. 相似文献
