基于合成法的定位误差的分析与计算

为保证机床夹具设计的可靠性,定位误差的计算是机床夹具设计过程中必不可少的重要环节之一。为此从定位误差的产生机理出发,介绍了定位误差的概念、产生原因,并给出计算公式,有助于快速实现定位误差的求解。     

Sublytic C5b-9 induces protective autophagy in cultured podocytes

AIM: In podocytes, autophagy occurs at a high basal level and dysregulated autophagy is associa-ted with a variety of podocytopathies. This paper is to investigate the role of autophagy in sublytic C5b-9-induced podocyte injury. METHODS: Sublytic complement C5b-9 stimulation was used as an in vitro model. Autophagosomes were confirmed using monodansylcadaverine (MDC) staining. Immunoblotting was used to measure the change of autophagy-related markers. Cellular morphological changes were observed by Wright-Giemsa staining. Immunofluorescence staining and confocal microscopy were used to detect the expression and distribution of nephrin. The cell viability was assessed by methylthiazol tetrazolium (MTT) assay. The cell apoptosis was assessed by Annexin V-fluorescein isothiocyanate/PI staining. RESULTS: For ensuring sublytic complement injury, the maximal amounts of anti-podocyte antiserum and 160×-diluted normal human serum were used without inducing cell lysis (defined as >5% LDH release). Sublytic C5b-9 promoted autophagy of podocytes in vitro. The proautophagic effect of sublytic C5b-9 manifested in the form of accumulated MDC-labeled vesicles and enhanced the expression of LC3-Ⅱ. Autophagy inhibitor 3-methyladenosine (3-MA) promoted sublytic C5b-9-induced podocyte morphological abnormalities. Compared with the sublytic C5b-9-injured podocytes, 3-MA exposure further decreased the expression of nephrin. 3-MA enhanced sublytic C5b-9-induced podocyte apoptosis. CONCLUSION: Sublytic C5b-9 attack induces autophagy, which may play a protective role against complement-mediated podocyte injury.

财产保险公司绝大部分业务来自于车险业务，因而车险业务的绩效关系到财险公司的经营业绩，甚至整个保险业的稳定和健康发展。从承保、理赔、续保及财务四个方面选取17个具体指标构建车险业务绩效评价指标体系进行有效性检验，计算某财险公司湖南分公司车险业务绩效水平指数，评价其车险业务经营状况。结果显示：该公司财务指标对车险业务影响的权重最大，综合车险指数呈上升趋势，表现出较好的经营状况。     

Cloning and Expression of Bile Salt Hydrolase Gene from Lactobacillus plantarum M1-UVS29

We cloned and expressed bile salt hydrolase gene of Lactobacillus plantarum M1-UVS29 in Lactococcus lactis NZ9000 successfully. Gene-specific primers for amplification of L. plantarum bsh were designed by using sequence which availabled from Gen Bank. The production of PCR amplicon was confirmed by sequencing and cloned into pMD18-T vector, and then recombined into expression vector p NZ8148 and yielding vector p NZ8148-BSH. p NZ8148-BSH was transferred into Lactococcus lactis NZ9000. Sequencing indicated that the cloned bsh fragment contained 995 nucleotides, and shared 99.3% sequence homology with bsh gene from L. plantarum MBUL10. Cloned bsh fragment was successfully transduced into NICE expression system and confirmed by PCR and restriction digest. Recombinant BSH protein was analyzed by SDS-PAGE. The molecular weight of BSH protein was approximately 37 ku. Activity of the expressed protein was 0.77 μmol · min-1. The successfully expressed proteins by genetic engineering technology made the function of lactic acid bacteria be abundant and laid the foundation for further researches into cholesterol-lowering lactic acid bacterium food and probiotics.     

贵州省番茄优势区域发展思考

利用贵州省山区特有的立体和区域性气候特点,针对6月到11月目标市场,发展夏秋反季节番茄产业,能缓解华南、“川渝”、“两湖”及港澳地区夏秋淡季市场.由于市场需求拉动,近年来,贵州省番茄种植面积逐年上升,但基地规模小,集中度不够,供应时间不长,产业化程度低等问题依然存在.为了进一步提升产业潜力,对贵州省近年来番茄种植现状进行深入调研,针对产业发展中存在的问题,结合自身气候资源禀赋和优势,找准目标市场定位,对番茄优势区域产业发展提出了发展措施和建议.     

赤峰市苦荞麦品种比较试验初报

为筛选赤峰市适宜种植的苦荞品种,2011年在赤峰市进行9个苦荞麦品种比较试验.结果表明：西荞1号、黔苦3号、苦荞04-46、川荞1号、黔苦5号、晋荞麦2号、西荞2号、六苦2081产量均比对照CK-10-2高,适宜当地推广种植.     

辽宁省普惠金融发展的问题与对策探讨

振兴东北老工业基地的一条重要路径是促进普惠金融发展。普惠金融的重点服务对象需要有政府的介入与协调以及资金和政策的支持。根据辽宁省普惠金融发展现状的调查,本文通过研究,发现在辽宁普惠金融发展过程中存在一系列的问题,并给出相应的政策建议。     

肉牛源细极链格孢菌的分离鉴定

为鉴定肉牛皮肤病病原及其致病性并筛选其敏感药物,本研究取患皮肤病肉牛病变部位皮屑、毛发和痂皮进行病原菌的分离,并对分离菌株进行了形态学鉴定、ITS序列分析、小鼠致病性试验和药敏试验。结果显示分离到1株形态特征与细极链格孢菌极其相似的菌株(PY2-1-2);经PCR扩增、测序和BLAST序列比对,结果显示分离株(PY2-1-2)的ITS基因序列与细极链格孢菌(MG975630.1)的ITS基因序列的相似性为99%,分离株PY2-1-2的ITS基因序列长度为543 bp,该片段包括ITS1、5.8S rDNA和ITS2的全部基因序列以及18S rDNA和28S rDNA的部分基因序列,提交GenBank(MH656780.1)。根据形态学结合ITS基因序列分析结果确定该分离株为细极链格孢菌。小鼠致病性试验结果显示该菌对小鼠有致病性。药敏试验结果显示:该菌对灰黄霉素、氟康唑、伊曲康唑、酮康唑的最小抑菌浓度(MIC)值分别为0.5μg/mL、4μg/mL、1μg/mL、1μg/mL。本研究为今后进一步探究细极链格孢菌引起的皮肤病诊治提供科学有效的参考。     

“岳苹”苹果在朝阳地区的引种试栽表现

“岳苹”是辽宁省果树研究所选育的晚熟型苹果品种,2013年在朝阳地区引种试栽,6a来对其在本地区物候期、抗寒性、抗病能力以及果实品质进行了观察与测定。结果表明:1)“岳苹”栽植后的第4a开花结实,花期26~27d,果实发育期123~125d,果实10月上旬成熟。2)平均单果重280g,最大单果重505g,果形指数0.86,可溶性固形物含量15.5%,果肉硬度11.2kg/cm^2。3)果皮鲜红色、果肉黄白色,多汁、松脆,品质上等。4)树冠较小、树势中庸、萌芽力强,抗病抗寒能力强,适宜在朝阳地区矮化密植建园。
