The objective of the study was to estimate the prevalence and losses associated with subclinical mastitis (SCM) caused by Staphylococcus aureus in Ethiopian crossbred dairy cows. A split-udder trial was carried out to determine milk yield losses in udder quarter with S. aureus-caused SCM. Each quarter of the study cows was examined using the California Mastitis Test (CMT) and quarter milk production was measured over a period of 8 days. Milk yield losses for CMT positive quarters were estimated by comparing production of quarters with CMT score 0. Mean milk yield for uninfected healthy quarters was 1.66 kg per milking (95% CI, 1.66–1.55 kg per milking), and the rate of milk reduction for quarters with CMT scores of 1+, 2+, and 3+ was 25%, 33%, and 48%, respectively. Economic losses at different farm-size levels were calculated by multiplying the prevalence of CMT scores with milk yield losses associated with respective CMT scores. In Debre Ziet dairy herds, a quarter with SCM due to S. aureus lost an average of 34.5% of its potential milk production while the total milk yield loss per cow was estimated at 6.8%. Losses were highest in large-scale (13%) farms and lowest (3.7%) in small-scale. Based on the prevalence, the overall financial loss for each cow per lactation was 984.64 Eth Birr (US78.65) and losses in large farms (1,882.40 Eth Birr or US78.65) and losses in large farms (1,882.40 Eth Birr or US150.35) were over 3.5 times the loss in small-size farms. These figures possibly underestimate the potential benefits of mastitis control program as they do not include other direct and indirect costs.  相似文献

Following the rapidly expanding dairy enterprise, mastitis has remained the most economically damaging disease. The objective of this study was mainly to investigate the in vitro antibacterial activities of ethanol extracts of Combretum molle (R.Br.Ex.G.Don) Engl & Diels (Combretaceae) against antibiotic-resistant and susceptible Staphylococcus aureus and Streptococcus agalactiae isolated from clinical cases of bovine mastitis using agar disc diffusion method. The leaf and bark extracts showed antibacterial activity against S. aureus at concentrations of 3 mg/ml while the stem and seed extract did not show any bioactivity. Although both leaf and bark extracts were handled in the same manner, the antibacterial activity of the bark extract against the bacterial strains had declined gradually to a lower level as time advanced after extraction. The leaf extract had sustained bioactivity for longer duration. The susceptibility of the bacteria to the leaf extract is not obviously different between S. aureus and S. agalactiae. Also, there was no difference in susceptibility to the leaf extract between the antibiotic-resistant and antibiotic-sensitive bacteria. Further phytochemical and in vivo efficacy and safety studies are required to evaluate the therapeutic value of the plant against bovine mastitis.  相似文献

Direct immunofluorescence and PCR detection methods were compared for sensitivity in evaluating the rabies status of archival specimens of Carnoy-fixed, paraffin-embedded brain tissue. The material consisted of 23 samples obtained during a rabies outbreak in Finland in 1988, and one sample isolated from a bat researcher who died of rabies in Finland in 1985. These results were compared with the original diagnoses performed on the fresh tissues. The immunofluorescence assay detected 100% (12/12) of the rabies-positive archival cases. A PCR assay designed to detect a 139-bp target near the 5' end of the rabies nucleoprotein gene also detected 100% (12/12) of the samples identified as positive in the fresh tissue specimens. A PCR assay designed to detect a 304-bp target spanning the 139-bp target of the first assay detected only 67% (8/12) of the original cases. No false positives were recorded. Both immunofluorescence detection of antigen and PCR detection of a short region of the nucleoprotein gene are useful in determining the rabies status of fixed, paraffin embedded (archival) material.  相似文献

Up to 100 single plant derived lines of 18 barley landraces, collected from 18 localities of six barley growing regions of Ethiopia were studied for two years at Holetta, Ethiopia for variation in five quantitative traits; scald severity, earliness, plant height, 1000 grain weight, and leafiness. The relative latent period in the adult plant stage to barley leaf rust, a good measure for partial resistance, was assessed on the landrace lines at Wageningen, The Netherlands. The observed variation between and within landraces was very large for all traits. The magnitude of variation was so large that most, if not all, plants within a landrace had a different genotype. The landraces also varied in the degree of variation. Some landraces, 1726 and 3288 for instance, were more variable for most or all traits than other landraces such as 208925 and 212938. Days to heading and scald severity were significantly (P = 0.01) correlated with altitude, r being 0.66 and - 0.65, respectively. Resistance to scald and leafiness also increased with altitude. On average landraces became later, more resistant to scald and more leafy the higher the altitude of collection. This associated complex of traits could be an adaptation to the wetter and cooler conditions at higher altitudes. In Tigray (Northern Ethiopia), soil moisture has been identified as the most limiting factor in agricultural production; on the other hand, loss of rain water through runoff as well as the induced soil loss has been determined as a critical problem in the region in the last two to three decades. To alleviate the above paradox, the government has mobilized communities and resources for the construction of physical soil and water conservation structures (stone bunds, terraces) in almost all land uses. However, yield improvement was mainly concentrated within the vicinity of the structures and runoff continued to overtop the structures, as no measures for in situ soil conservation were taken. The terwah system, consisting of traditional ploughing followed by making every 1.5–2 m contour furrows, and permanent raised beds with contour furrows at 60–70 cm interval treatments, were considered and evaluated as practices that could increase the efficiency of in situ water utilization and soil conservation. An experiment was started in Gum Selasa, which is one of the drought prone areas in Tigray, whereby runoff volume and sediment load were measured after every rain event. Permanent raised beds with contour furrows at 60–70 cm interval significantly (P < 0.05) reduced runoff volume, runoff coefficient and soil loss as compared to traditional ploughing: 255, 381 and 653 m3 ha−1 runoff was recorded from permanent bed, terwah and traditional ploughing, respectively during the whole cropping season. The above runoff induced 4.7 t ha−1 soil loss from permanent bed, 7.6 t ha−1 from terwah and 19.5 t ha−1 from traditional ploughing. Overall, contour furrows and permanent raised beds can be part of the ongoing intensification process which includes physical soil and water conservation, slope reforestation, irrigation development and agro forestry in crop lands. Moreover, the use of permanent raised beds if combined with crop mulching and crop diversification is an important component for the development of sustainable conservation agriculture practices in the region.  相似文献

Bactrocera invadens, the Asian fruit fly, was first reported in Kenya in 2003, and it spread fast to most tropical countries in Africa. To our knowledge, there is no detailed data on the fruit damage and status of fruit flies in Arba Minch and elsewhere in Ethiopia. Hence, information on the species composition and pest status of the fruit fly species is urgent to plan management strategies in the area. Fruit flies were captured using male parapheromone-baited traps. Matured mango (Mangifera indica) fruits were collected from randomly selected mango trees and incubated individually in cages (15 by 15 by 15 cm) with sandy soil. B. invadens was the predominant (96%; 952 of 992) captured species and the only fruit fly species emerging from mango fruits incubated in the laboratory. The mean number of adult B. invadens emerging per mango fruit was 35.25, indicating that the species is the most devastating mango fruit fly in the area. The loss due to this species would be serious if no management strategies are implemented.  相似文献

A study was carried out on 59 clinically healthy Zebu x Friesian crossbred cows to determine the prevalence of subclinical endometritis, associated factors and its effect on the reproductive performance. Subclinical endometritis was diagnosed by endometrial cytology using uterine lavege technique and subclinical mastitis using CMT. The clinical, management and reproductive data were obtained from a weekly follow up visit of each cow for a period of 6 months after calving. The prevalence of subclinical endometritis, with ≥5% neutrophil count, was 47.5% and 30.5% at week 4 and 8, significantly decreasing (P = 0.002) as postpartum period advanced. Body condition score at week 4 postpartum (OR = 4.5, P = 0.017) and regular cow exercise (OR = 4.8, P = 0.026) were the significant risk factors while post-calving hygiene (P = 0.06) was poorly associated. Subclinical endometritis was also directly associated with subclinical mastitis at both week 4 (OR = 4.5, P = 0.012) and 8 (OR = 3.6, P = 0.031) postpartum. The risk of first service pregnancy (OR = 5.1, P = 0.004) was higher in cows negative for subclinical endometritis at week 8 postpartum and the proportion of cows that required more than 3 services was higher in cows with uterine inflammation. Also the proportion of cows diagnosed pregnant within 180 DIM was higher in cows with normal uterus at both week 4 (OR = 10.3, P = 0.001) and week 8(OR = 21.8, P = 0.001). These results indicated that subclinical endometritis was directly associated with poor body condition and subclinical mastitis and that it had negative effect on reproductive traits of dairy cows. This association may also reflect the possibility of translocation of bacteria/bacterial products from the uterus to the udder or vic-versa or else the presence of common cause for both endometritis and mastitis.  相似文献

We conducted a study in Komto Forest in East Wollega Zone, Oromia National Regional State, West Ethiopia for determining vegetation structure and regeneration status in this forest. We systematically sampled 53 quadrats (20 m × 20 m) along line transects radiating from the peak of Komto Mountain in eight directions. Vegetation parameters such as DBH, height, seedling and sapling density of woody species, and location and altitude of each quadrat were recorded. In total, 103 woody plant species of 87 genera and 45 families were identified. Analysis of selected tree species revealed different population structures. Generally, the forest was dominated by small trees and shrubs characteristic of secondary regeneration. Observations on the regeneration of the forest indicated that there are woody species that require urgent conservation measures. Based on the results of this study, we recommend detailed ecological studies of various environmental factors such as soil type and properties, and ethnobotanical studies to explore indigenous knowledge on uses of plants.  相似文献

Hot pepper is the most important worldwide grown and consumed spice and vegetable crop. Though hybrid breeding has been proposed for genetic improvement in the crop, but there is lack of information on heterosis in crosses among crop genotypes in Ethiopia. Twelve genotypes (nine Asian and three Ethiopian parents) of hot pepper were crossed in 2003 cropping season in a half-diallel fashion to fit Griffing’s fixed effect model analysis. An open field experiment was conducted in 2004/2005 to investigate heterosis for fourteen traits in 66 F1 hybrids grown together with their 12 selfed parents. Highly significant genotypic differences were observed for all the traits except for leaf area. Variance component due to specific combining ability (dominance) were larger than that due to general combining ability (additive) for each of the studied traits with few exceptions. Broad sense heritability (H b2) for fruit traits were more than 60% and with wide gap from narrow sense heritability (h n2) for most of the important traits like number of fruit per plant (H b2 = 88.3% and h n2 = 46.0%), days to maturity (H b2 = 87.2% and h n2 = 23.1%) and dry fruit yield per plant (H b2 = 72.6% and h n2 = 14.6%). Maximum heterosis over mid-parent and better-parent, and economic superiority of hybrid over standard check were recorded, respectively for dry fruit yield per plant (163.8, 161.8 and 92.1%), number of fruits per plant (104.4, 79.6 and 136.4%) and days to maturity (−29.8, −31.5 and −23.6%). These observations suggested a possibility of utilizing dominance genetic potentiality available in diverse genotypes of the crop by heterosis breeding for improving hot pepper to the extent of better economic return compared to the current commercial cultivar under production in the country. Low narrow sense versus very high broad sense heritability for days to maturity and dry fruit yield per plant could be a sign for achievability of earliness and high fruit yield using heterosis in hot pepper. The maximum heterobeltiosis were recorded either from F1s obtained from Ethiopian and Asian crosses or from within Asian crosses, suggesting the possibility of maximizing heterosis by considering genetically diverse parental genotypes. The manifestation of highest heterosis in hybrids from among Asian lines indicated existence of genetic diversity among Asian genotypes and the potentiality for improvement of hot pepper using genotypes from different regions of the world along with elite inbred lines from local cultivars.  相似文献

African nightshades are becoming more important as leafy vegetables in sub-Saharan Africa. Previously considered as food for the poor, their cultivation is now being promoted, and some cultivars are commercialised; however, most farmers use self-produced seeds, leading to low and varying yields. Improvement through conventional breeding depends on the available genetic diversity, the possible breeding systems, and the nutritional value of the accessions. Therefore, we review the information on these topics with the following main outcomes: the most commonly discussed species, S. nigrum, S. scabrum, S. villosum, and S. americanum, could be differentiated using molecular markers, but further sub-clustering was rarely possible, and statistical support often missing. S. nigrum and S. scabrum seem to be most closely related to each other. The mainly self-pollinating African nightshades form a polyploidy series with diploid (2n = 2x = 24) to hexaploid taxa. Interploidy hybridisations between diploids and tetraploids are possible, whereas the hexaploid S. nigrum and S. scabrum could not be crossed to genotypes of lower ploidies. Solanine, solamargine, solasonine, and chaconine are the major steroidal alkaloid glucosides in African nightshades. Amounts are age and environment dependant. Mineral and vitamin contents in leaves are at least as high as in Brassica oleracea or Spinacia oleracea, underlining their relevance as local vegetables.  相似文献
