The effects of the severity and timing of leaf removal(LR) on the amino acids of Sauvignon Blanc grapes and wines were studied during the 2017 growing season. High-performance liquid chromatography(HPLC) was used to analyze the amino acids profiles of grape berries and wines. The basal leaves were removed at three time points(40, 56 and 72 days after flowering, named LR40, LR56 and LR72, respectively) at two severity levels(one at which the first, third, and fifth basal leaves of each shoot were removed(50% level); and another at which the first six basal leaves were removed(100% level)). The results showed that leaf removal had little impact on total soluble solids(°Brix), titratable acidity, pH or berry weight. The LR72-50% treated grapes had higher berry weight, titratable acidity and °Brix than those of the other treatments. The highest concentrations of total amino acids and of total amino acids except proline were detected in LR72-50% treated grapes(2 952.58 and 2 764.36 mg L~(-1), respectively); the lowest were detected in LR72-100% treated grapes(2 172.82 and 2 038.71 mg L~(-1), respectively). LR72-50% treatment significantly promoted the synthesis of aspartic acid, serine, arginine, alanine, aminobutyric acid and proline at both severity levels for grapes, the concentrations of all of these amino acids were increased relative to the control concentrations. The LR72-50%, LR40-100% and LR72-100% treated wines had higher total amino acids concentrations and higher concentrations of some individual amino acids, such as arginine, alanine and serine, than did the control wines. Of all the amino acids studied, glycine, tyrosine, cysteine, methionine and lysine were not significantly influenced by the timing or severity basal defoliation in grapes and wines. The assay was aimed to study the effect of Hedyotis diffusa Willd superfines powder on immune function. Three groups of mice were gavaged with the drugs at doses of 1.75,1.25 and 0.75 g/kg once a day for 7 consecutive days, repectively. The mice of control group were administered with the same volume of saline.At the last time, mice were injected into the muscle with Newcastle disease vaccine,and get peripheral blood on days 7, 14 and 21 after the medicine administration. The enhancement of innate immune responses were evaluated by using CCK-8 method, hemagglutination inhibition test, macrophage phagocytic function test and organ coefficient method. The levels of lymphocytes proliferative capacity, serum antibody, peritoneal macrophage phagocytosis and the indexes of immune organs in mice were extremely significantly enhanced (P<0.01). 试验旨在分析绵羊初乳和常乳乳蛋白质组的差异,揭示绵羊不同泌乳阶段的乳蛋白质组特征。选用6只胎次相同、预产期相近的湖羊母羊,分别采集分娩后第1、3、7天的初乳和第14、28、56天的常乳,每个时间点采集6份奶样并等体积混合,离心弃去乳脂肪,采用双向电泳(2-DE)技术对初乳和常乳的脱脂乳蛋白进行比较分析。结果发现,初乳中乳蛋白含量较高,且随着泌乳期的延伸,乳蛋白含量降低。以第1天乳蛋白2-DE图谱为参照,第3天检测到38个差异蛋白点,其中有20个蛋白点仅存在于第1天图谱中,有1个蛋白点仅存在于第3天图谱中,其余17个蛋白点呈现表达量的差异;第7天检测到35个差异蛋白点,其中有19个蛋白点仅存在于第1天图谱中,有1个蛋白点仅存在于第7天图谱中,其余15个蛋白点呈现表达量的差异;第14天检测到34个差异蛋白点,其中有11个蛋白点仅存在于第1天图谱中,有1个蛋白点仅存在于第14天图谱中,其余22个蛋白点呈现表达量的差异;第28天检测到38个差异蛋白点,其中有28个蛋白点仅存在于第1天图谱中,有1个蛋白点仅存在于第28天图谱中,其余9个蛋白点呈现表达量的差异;第56天检测到36个差异蛋白点,其中有26个蛋白点仅存在于第1天图谱中,有1个蛋白点仅存在于第56天图谱中,其余9个蛋白点呈现表达量的差异。综上,共检测出44个差异表达的蛋白点,其中有8个蛋白点仅存在于第1天图谱中,而这些蛋白主要涉及免疫活性功能。 相似文献

温度在气体污染物扩散的过程中起一定的作用。在现实生活中,由于日晒和地表辐射的存在,使得地面、建筑物外表面都与空气存在一定的温差。温差的存在会改变地表及壁面附近的流场,从而对住宅区污染物的扩散产生影响。利用理论推导对热力因素及扩散因素的关系进行了研究,并且与数值模拟的试验结果进行比较。结果发现理论推导与试验结果整体趋势相一致。这表明在与无温差情况相比较,温差存在时,地表及壁面附近流场更加紊乱;温差与气体污染物的扩散具有正相关性,对住宅区污染物的稀释起积极作用。 相似文献

鸭甲型肝炎(duck hepatitis A,DHA)是危害养鸭业的主要疾病之一,是一种雏鸭的急性、高度致死性传染病。本研究旨在研制一种对我国流行的鸭甲型肝炎血清1型(DHAV-1)和血清3型(DHAV-3)病毒具有交叉中和作用的单克隆抗体(monoclonal antibody,McAb)。人工合成12条DHAV-1和DHAV-3的VP1蛋白共有的抗原表位肽,分别与钥孔血蓝蛋白(keyhole limpet hemocyanin,KLH)载体偶联作为免疫抗原,免疫BALB/c小鼠制备McAb。通过检测McAb与DHAV-1和DHAV-3的交叉反应性,测定McAb对病毒增殖的抑制效率、中和效价以及攻毒保护率等来筛选McAb。本研究共获得了12株稳定分泌McAb的杂交瘤细胞株,其中6株(C1、C4、C7、C12、C13、C16)分泌的McAb同时与DHAV-1和DHAV-3发生特异性交叉反应; C1、C4、C7、C16可以抑制DHAV-1和DHAV-3在鸭胚中的增殖,抑制率在75.34%~100.00%不等;对DHAV-1和DHAV-3的中和效价:C1(1:3&1:5)、C4(1:3&1:3)、C7(1:10&1:11)和C16(1:9&1:9);C7和C16对DHAV-1和DHAV-3攻毒雏鸭的保护率较高,分别为“70%、80%”和“100%、60%”。本研究成功研制出对DHAV-1和DHAV-3具有交叉中和活性的McAb 4株,其中2株对病毒的感染具有良好的预防效果,为DHA的防控提供了新材料和新思路。 相似文献

The aim of this study was to investigate the differential expression genes induced by ApoCⅢ,and study the function of ApoCⅢ.Porcine aortic vascular endothelial cells were successfully isolated using enzyme digestion,and then screened the differential expression genes induced by ApoCⅢ using the Solexa high-throughput sequencing technology.The results showed 647 differential expression genes,including 390 up-regulated genes and 257 down-regulated genes.The qRT-PCR results verified that the gene expression results from Solexa sequencing data were reliable.GO and Pathway analysis showed that the function of differential expression genes were related to immune response,cell apoptosis and death.These findings suggested that ApoCⅢ affected the physiological function of porcine aortic endothelial cells by the molecular pathways of inflammation,cell adhesion and apoptosis,which provided a theoretical basis for further understanding the molecular mechanisms of atherosclerosis caused by ApoCⅢ. 相似文献
