AIM:To investigate the effects of fibronectin (FN) on the proliferation and collagen synthesis in cultured rat cardiac fibroblasts (CFb) derived from SHR (CFbSHR) and WKY (CFbWKY). METHODS:CFb derived from 12-week-old spontaneously hypertensive rat (SHR) and WKY was cultured by outgrowth of tissue block. Cell proliferation of CFb was measured by cell number counting and[3H]-TdR incorporation using 24-well plates pre-coated with 5 μg/cm2 of FN. Collagen synthesis was determined by [3H]-proline incorporation. RESULTS:As compared with control, the cell number of fibroblasts derived from SHR and WKY were significantly increased to 163.75% and 170.42% respectively after 72 h incubation with FN in the presence of 0.4% FCS from a intial cell density of 1×104 cells/mL. DNA synthesis of CFb was markedly promoted by FN. FN induced an increased in [3H]-proline incorporation in both CFbSHR and CFbWKY. CONCLUSION:FN is able to promote cell proliferation and collagen synthesis of CFb derived both from SHR and WKY. 相似文献

Variation in the development of secondary dormancy in oilseed rape genotypes under conditions of stress 总被引:6,自引:0,他引:6
Summary A substantial amount of seed is left in the fields before and during harvest of oilseed rape. Although this crop exhibits little or no primary dormancy, the absence of certain environmental cues that promote germination of imbibed seeds induces secondary dormancy. The work reported investigated the extent to which environmental stress conditions, including osmotic stress, low oxygen stress and anaerobiosis, induce secondary dormancy in oilseed rape, and examined the variation in development of secondary dormancy between and within genotypes. Osmotic stress was most effective in inducing dormancy. Anaerobic treatment produced very few dormant seeds, as did an atmosphere low in oxygen and high in nitrogen. The development of secondary dormancy under osmotic stress varied considerably between and within genotypes. Dormancy ranged from almost zero to about 60% for winter genotypes and about 85% for spring types. Within genotypes, variations occurred between seed lots and years of harvest. Temperature variations affected the percentage of dormant seeds. More dormant seeds were likely to be produced with incubation under water stress at 20 °C than at 12 °C. In winter genotypes, fewer dormant seeds were produced when incubation temperature and germination test temperatures differed. Thus, incubating at 20 °C and 12 °C, followed by germination tests at 20 °C and 12 °C, respectively, produced most dormant seeds. Also, in the winter genotypes, the potential development of secondary dormancy was positively correlated with the pattern and speed of germination of untreated seeds. 相似文献

东北西部沙地近20年地下水变化动态及其成因分析 总被引:6,自引:4,他引:6
本文以奈曼旗为例,分析研究了我国东北西部沙地1979-1997年地下水位时空变化规律及其与降水量和沙漠化的关系。结果表明:(1)沙地多年平均地下水位为2.19m,其中农区为2.85m,牧区1.16m;(2)地下水位是1981-1983年最低为2.49-2.56m,1991-1992年最高为1.90-1.92m,1997年为2.22m,高于1979年的2.41m;(3)该区季节最高水位主要发生于春季(3-5月),发生频率占50.1% ,最低水位主要发生于冬季(12-2月),发生频率为41.2%,平均4月最高为2.07m,10月最低为2.23m;(4)地下水位的年际变化动态和降水量密切相关,但有滞后1-2年的现象;(5)地下水位空间分布上的差异与土地沙漠化及土地利用方式密切相关,沙漠化土地主要用于放牧,由于人为利用少和蒸散少,有利于地下水抬升;(6)由于耕地面积占总土地面积比例低,水浇地和粮食产量的增加尚未对地下水位造成严重影响。 相似文献

采用杂种指数和形象化散点图法, 探明了变叶海棠以兼性无融合生殖方式与陇东海棠和花叶海棠产生渗入杂交变异, 又以无融合生殖方式保持新产生变异的遗传, 经过数千万年的世代繁衍,形成现今这样极其复杂多样的渗入杂交群体, 该群体为3 个种的杂交复合体。由于杂种的全部或部分能育子代中发生的基因重组和增加基因的突变, 丰富了物种的基因库, 从而深入的揭示了变叶海棠具有多种高抗性的遗传机理。在变叶海棠居群内保护或栽植适量的陇东海棠和花叶海棠, 对于保持和促进变叶海棠多样性的形成与分化具有重要意义。 相似文献

AIM:To investigate the effect of the heat shock response on the reperfusion arrhythmias(RAs) and the possible mechanism involved. METHODS:Fifty-five Sprague Dawley rats were randomly divided into 2 groups: the heat shock group (group H,n=29) and the control group (group C,n=26). The rats in group H were preconditioned with heat shock 24 hours before, and that in group C were not. The hearts of 16 rats in group H and 16 in group C were exercised and mounted on a non-circulating Langendorff perfusion apparatus and perfused retrogradely with modified K-H buffer and mimic ischemia/reperfusion was applied. Additionally, conventional intracellular microelectrode techniques were used for recording such electrophysiological parameters as resting potential(RP), action potential amplitude(APA), over shot(OS), maximum depolarization velocity(Vmax) of the hearts of other 13 rats in group H and 10 in group C. RESULTS:①Prior heat stress significantly decreased reperfusion arrhythmia. ②The amount of CK release in the effluent in group H was much less than that in group C. ③Myocardial HSP70 content was elevated significantly in group H. ④Heat stress significantly increased myocardial anti-oxydases activity and decreased lipid peroxydative products. Additionally, heat stress significantly reduced the Vmax of action potential. It indicated that rapid Na+ channel of papillary muscles may be inhibited by heat shock. The degree of change of Vmax after ischemia in H group was significantly less than that in group C. And the time of reperfusoin with Tyrode's solution till the action potential appeared as large as that before perfusion with mimic ischemic solution is shorter in group H than in group C. CONCLUSION:Heat shock pretreatment markedly reduces ischemia/reperfusion-induced injury of heart and ventricular arrhythmias in rats and this effect may be associated with the inhibition of rapid Na+ channel of papillary muscles by heat shock and the increase in myocardial HSP70 and anti-oxydase activity. 相似文献
