Trisomy 21 usually results from nondisjunction during meiosis I. In order to determine whether nondisjunction results from failure of normal chromosome pairing or premature unpairing, recombination frequencies were estimated between DNA polymorphic markers on the long arm of chromosome 21 in families containing one individual with trisomy 21. The recombination frequencies on chromosomes 21 that had undergone nondisjunction were then compared to those on chromosomes 21 that had disjoined normally. The data indicate that recombination is reduced between DNA markers on nondisjoined chromosomes 21. These results are consistent with the hypothesis that reduced chiasma formation predisposes to nondisjunction, resulting in trisomy 21 in humans. 相似文献

Three in vitro experiments were conducted to determine the effects of pH on ruminal conversion of L-tryptophan (TRP) to 3-methylindole (3-MI) and indole (IND). Experiment 1 involved 2 closed-system incubations, each with triplicate replications of buffered ruminal fluid at initial pH of 4.5, 5.0, 5.5, 6.0, 6.5, 7.0, 7.5, and 8.0. Highest molar conversions of TRP to 3-MI were 61.5 +/- 2.7% and 64.3 +/- 0.8% at initial pH treatments of 7.5 and 7.0 (final pH 7.3 and 6.9) for the 2 incubations, respectively. Experiment 2 used duplicate continuous cultures at each of 4 pH treatments. Following a common 5-day pretreatment period, effluent pH averaged 5.6, 6.1, 6.5, and 6.9 for the respective treatments. Data for 4 subsequent 5-day time periods showed production of 3-MI was affected by pH treatment (P less than 0.01), time (P greater than 0.01), and treatment X time (P less than 0.01); conversion of TRP to 3-MI reached 78% at pH 6.9 and decreased to less than 1% at pH 5.6. Production of IND was not related to pH treatment (P greater than 0.10). Total moles of volatile fatty acid (VFA) carbon produced showed a pH treatment X time interaction (P less than 0.01) which reflected a trend toward decreasing VFA production at lower pH and increasing VFA production at higher pH. 为充分利用林区的自然资源,1986—1987年先后在帽儿山实验林场和依兰县先锋林场进行了养鱼试验,经过120天的饲养期,鱼产量在帽儿山为3094.5kg/ha,在先锋为1882.5kg/ha。 林区水资源丰富,水质稳定;水的pH值偏低,多为软水,水中营养盐类贫乏。用石灰清塘可使pH值由6.5升至8.3。施肥可促进浮游生物的增殖,藻类的优势种为隐藻,浮游动物为近邻剑水蚤。灯光诱虫既能使鲤鱼生长明显加快,又能除去部分森林害虫。林区水体污染较轻,故家养鱼类病害也较少。 在帽儿山、鱼的增重倍数为5.33,饵料系数为2.14;在先锋则分别为7.68和1.35。由于鲤鱼适应低温环境,所以长势较鲢鳙鱼为好。在帽儿山鲤鱼平均重0.31kg,在先锋则为0.40kg。 林区宜主养鲤鱼,也应放养草食和腐食性的鱼类,以充分利用水体中的生态位。 相似文献

In classical enzymology, intermediates and transition states in a catalytic mechanism are usually inferred from a series of biochemical experiments. Here, we derive an enzyme mechanism from true atomic-resolution x-ray structures of reaction intermediates. Two ultra-high resolution structures of wild-type and mutant d-2-deoxyribose-5-phosphate (DRP) aldolase complexes with DRP at 1.05 and 1.10 angstroms unambiguously identify the postulated covalent carbinolamine and Schiff base intermediates in the aldolase mechanism. In combination with site-directed mutagenesis and (1)H nuclear magnetic resonance, we can now propose how the heretofore elusive C-2 proton abstraction step and the overall stereochemical course are accomplished. A proton relay system appears to activate a conserved active-site water that functions as the critical mediator for proton transfer. 相似文献

X-ray absorption spectroscopy was used to probe the chemical and structural environment of vanadium in coal. It was found that vanadium exists in at least two environments, in both of which it was coordinated to oxygens. There was no evidence of vanadium in nitrogen (porphyrin) or sulfide environments. It was also found that the vanadium environments in the raw coal did not survive unchanged in a liquefaction process. These findings have implications for coal cleaning processes and for trace element release into the liquefaction process stream. 相似文献

Nitric oxide synthase (NOS) is an enzyme that catalyzes the formation of nitric oxide (NO), an important biological messenger from L-arginine. There are considerable evidence showing the expression of NOS in mammalian tissues. Information on distribution of NOS activities in various organs and tissues of fish is rare. Non-functional NOS activities were documented in fish semi-quantitatively either by an indirect nicotine-adenine-dinucleotide-phosphate diaphorase (NADPH-d) activity histochemical staining method or by an immunohistochemical method using a cross-reacting antibody to brain NOS. Report on the functional levels of NOS activities in fish is lacking. This report represent the first attempt to document the functional NOS levels in various fish organs and tissues. Constitutive NOS (cNOS) activities in various organs of big-head carp (Aristichthys nobilis) was measured by a chemiluminescence method with a detection limit as low as 10 mol of NO produced. It was found that constitutive NOS activity was highest in the brain, followed by the intestine, stomach, retina, olfactory lobe, swim bladder, skeletal muscle, heart, kidney, ovary and liver. NOS activity could not be detected in the gill filaments. Omission of NADPH in the reaction mixture caused a 57–100% decrease in cNOS activities. However, omission of arginine in the mixture only caused a 56–87% drop in cNOS activities. When compared with cNOS activities documented from other species, a similar pattern of cNOS activities in the various organs and tissues of big-head carp could be seen. 相似文献

Posttranslational modifications of proteins regulate many biological processes, including metabolism, signal transduction, and gene expression. The synthetic challenges associated with generating homogeneous populations of selectively modified proteins, however, have hindered detailed studies of the effects of these modifications on protein structure and function. Here, we report an approach to the cotranslational synthesis of selectively glycosylated proteins in which the modified amino acid is genetically encoded. We show that myoglobin containing beta-N-acetylglucosamine (GlcNAc)-serine at a defined position can be expressed in Escherichia coli in good yield and with high fidelity. The beta-GlcNAc moiety can be recognized by a saccharide-binding protein, or subsequently modified with a galactosyltransferase to build more complex carbohydrates. This approach should be generally applicable to other posttranslational modifications such as protein phosphorylation, acetylation, and methylation. 相似文献

ABSTRACT From a transaction cost perspective, this paper shows how the tradition of consuming marine fish by the majority of Hong Kong's Chinese citizens has shaped the means and modes of marketing seafood in Hong Kong. It is argued that consumer preferences stimulate aquaculture as a non-open access measure to bypass state fish marketing regulations. This has resulted in two outcomes. First, the transaction cost savings on metering output quantity and quality under private property have led not only to the collapse of the state monopoly on marine fish marketing, but also to the emergence of the (then new but) currently popular kind of Chinese seafood restaurant in Hong Kong. Second, consumer preferences for variety have shaped the form of coordination between the producer and the consumer. The advantage of market coordination has led to the emergence of a system of wholesaling and retailing rather than vertical integration. Issues concerning the emergence of Hong Kong as a regional live marine fish production and trading centre are discussed in terms of the contribution of local mariculture to sustainable development. 相似文献
