Effect of immunosuppression on pathogenicity of herpesvirus of turkeys (HVT) in chickens was studied. In three experiments, newly hatched chickens of two genetic strains were surgically thymectmized and gamma-irradiated and then exposed to HVT. Lymphoid cells of thymectomized-irradiated chickens had reduced in vitro mitogenic response; this result indicated that the immunosuppressive regimen caused suppression of T cell function. The immunosuppressed chickens had more circulating HVT and had a slightly higher frequency of microsocpic lesions in peripheral nerves than the intact control chickens, although the intensity of lymphoproliferation remained very mild and no gross lesions or clinical disease was noted during the 5-week observation period. The absence of significant lesion development in an immunosuppressed host indicated that pathogenic potentials of HVT may be limited.  相似文献

Angiogenesis, which is essential for malignancies to progress, depends on various signalling proteins including vascular endothelial growth factor (VEGF) and vascular endothelial growth factor receptors 1 and 2 (VEGFR-1 and VEGFR-2). Microvessel density (MVD) is frequently used to evaluate angiogenesis. This study assessed the relationship between expression of VEGF, VEGFR-1 and VEGFR-2, MVD and the survival time in dogs with lymphoma. VEGF, VEGFR-1 and VEGFR-2 expression was evaluated immunohistochemically and microvessel profiles were counted in 34 lymphoma samples. Seventy-nine percent of the samples showed high VEGF expression and 62% were highly positive for VEGFR-1; VEGFR-2 immunoreactivity was mostly negative. Dogs treated with chemotherapy had a median survival time of 266days, but no significant relationships were found between overall survival time, MVD and expression of VEGF, VEGFR-1 or VEGFR-2. In this study, VEGF its receptors and the MVD were no prognostic factors in dogs with lymphoma.  相似文献

SUMMARY A three-year-old neutered female Border Collie was presented with in-appetence, vomiting and diarrhoea. Abdominal radiographs revealed an obstructive pattern but no physical obstruction was evident at laparotomy. A diagnosis of chronic intestinal pseudo-obstruction was made based on histopathological changes in intestinal biopsies. Treatment was unsuccessful and the dog deteriorated progressively until euthanased five weeks after presentation.  相似文献

This study compared the sensitivity of acetate tape impression and skin squeezing with that of deep skin scraping for the diagnosis of demodicosis in dogs. Demodex canis was detected in 100% of acetate tape impressions obtained after skin squeezing and in 90% of deep skin scrapings. There was a significant difference (P < 0.001) between the techniques in the total number of mites detected. Acetate tape impression with skin squeezing was found to be more sensitive than deep skin scraping and is an alternative diagnostic method for canine demodicosis.  相似文献

The emerald ash borer (EAB) has killed millions of ash trees in Michigan and at least fourteen other states since its first detection near its introduction point in metropolitan Detroit in 2002. Despite overstory ash mortality near 100% in many areas, ash seedling and saplings remain unaffected and provide the potential for ash re-establishment into the canopy of deciduous forests of the region. We examined the potential for ash re-establishment in areas of heavy mortality by measuring the status and change of ash regeneration at 45 sites across southeastern Lower Michigan in 2007 and 2009. Ash regeneration was found to be abundant in all forest types, particularly in the smallest height classes, though it was more abundant in ash species of upland forests compared to those of lowland forests. New seedlings 1-2 years old were less common than other regeneration size classes, and declined substantially between 2007 and 2009, suggesting a depletion of the ash seed bank in these forests. Regeneration density was not explained well by the presence of overstory ash prior to EAB introduction, suggesting that competitive interaction with other tree species in higher canopy strata is an important driver of ash regeneration density. Regeneration is sufficient to replenish overstory ash to pre-EAB levels in upland forests, though ecological changes caused by overstory ash mortality in lowland areas may affect local hydrology in a way that reduces opportunities for tree regeneration. Despite abundant ash regeneration, it remains unclear whether ash will recover to the overstory of forests in southeastern Michigan because of uncertainties in the future dynamics of EAB in the region.  相似文献

Epidemiology of reticuloendotheliosis virus in broiler breeder flocks   总被引:1,自引:0,他引:1  
Six broiler breeder flocks from two companies in Mississippi were tested at intervals for reticuloendotheliosis (RE) virus infection. Virus was isolated and antibody demonstrated in all six flocks. Infection was first detected at ages ranging from 13 to 47 weeks. Studies showed that neither congenital transmission from grandparent flocks nor treatment with contaminated vaccines was a likely source of infection; thus, exposure to RE virus was assumed to come from the environment. Virus was isolated from litter samples from two of the flocks, but no specific environmental infection source was identified. Infection rates of flocks differed between the two companies. Although adequate controls were lacking, no performance problems due to RE virus infection were apparent in breeder or broiler progeny flocks. However, the RE viruses isolated from these flocks were immunosuppressive and oncogenic when inoculated into day-old chicks. A moderate (3-16%) incidence of neoplasms was induced by contact exposure to these field isolates in the laboratory.  相似文献

Mareks disease virus (MDV), a herpesvirus, and avian leucosis virus subgroup J (ALV-J), a retrovirus, were used for experimental coinfection of chickens. Chimeric molecules having sequences of both viruses were detected by the hotspot-combined polymerase chain reaction (HS-cPCR) system. The detection of chimeric molecules provided evidence for avian retroviral inserts in the herpesvirus genome. The persistence of chimeric molecules on in vivo passage served to indicate the infectivity of the recombinant virus. The evaluation of formation and persistence of the chimeric molecules was performed in two trials involving three in vivo passages. The chimeric molecules were identified according to the primer sets, their product length, and pattern. The persistence of chimeric molecules on in vivo passages served as an indication of their ability to replicate in and infect chickens. In the first experimental passage, MDV and ALV-J prototype strains, MD11 and HC-1, were intraperitoneally (i.p.) injected into 1-day-old chicks. The second trial included two passages. Passage II chicks were injected i.p. and passage III chickens were in contact with the chickens of passage II. For passage II, enriched white blood cells from blood samples of chickens from the first trial that had chimeric molecules were injected i.p. into 1-day-old chicks. For passage III, uninfected chicks were included together with the infected chicks. Synthesis evidence for the various species of chimeric molecules was assessed in the tissues of birds of the second trial. DNA was extracted from blood and feathers and analyzed by the hotspot-combined PCR and by pulsed field gel electrophoresis. To overcome the limits of detection, three amplification assays followed by hybridization of the products to specific viral probes were conducted. A variety of chimeric molecules were detected in low concentrations. Five species of chimeric molecules were characterized in blood, tumors, and feathers. Chimeric molecules were detected in 18 of 36 dually infected birds from the first trial and in 14 of 21 dually infected birds from the second trial. The findings show that, in four out of seven groups of the second trial, the chimeric molecule species persisted on passage.  相似文献
