Immunodetection of allergens in dark chocolate is complicated by interference from the chocolate components. The objectives of this study were to establish reference materials for detecting multiple allergens in dark chocolate and to determine the accuracy and precision of allergen detection by enzyme-linked immunosorbent assay (ELISA) before and after chocolate processing. Defatted peanut flour, whole egg powder, and spray-dried milk were added to melted chocolate at seven incurred levels and tempered for 4 h. Allergen concentrations were measured using commercial ELISA kits. Tempering decreased the detection of casein and β-lactoglobulin (BLG), but had no significant effect on the detection of peanut and egg. Total coefficients of variation were higher in tempered than untempered chocolate for casein and BLG, but total and analytical CVs were comparable for peanut and egg. These findings indicate that processing has a greater effect on recovery and variability of casein and BLG than peanut and egg detection in a dark chocolate matrix.  相似文献

Among the major food allergies, peanut, egg, and milk are the most common. The immunochemical detection of food allergens depends on various factors, such as the food matrix and processing method, which can affect allergen conformation and extractability. This study aimed to (1) develop matrix-specific incurred reference materials for allergen testing, (2) determine whether multiple allergens in the same model food can be simultaneously detected, and (3) establish the effect of processing on reference material stability and allergen detection. Defatted peanut flour, whole egg powder, and spray-dried milk were added to cookie dough at seven incurred levels before baking. Allergens were measured using five commercial enzyme-linked immunosorbent assay (ELISA) kits. All kits showed decreased recovery of all allergens after baking. Analytical coefficients of variation for most kits increased with baking time, but decreased with incurred allergen level. Thus, food processing negatively affects the recovery and variability of peanut, egg, and milk detection in a sugar cookie matrix when using immunochemical methods.  相似文献

During rice milling, the bran and germ are successively removed from the caryopsis (kernel). Because bran and germ contain large quantities of lipid, the amount of lipid remaining on the kernel surface may be used as a method for the assessment of milling quality. Bulk samples of rice pureline varieties and an experimental hybrid were milled for 0, 10, 20, 30, and 40 s. Scanning electron microscopy (SEM) revealed that brown rice kernels had large contours of linear protuberances and depressions running lengthwise along the kernel surface. The protuberances were abraded successively during milling, but varying amounts of material remained in the depressions. Light microscopy combined with the lipid-specific probes Nile Blue A or Sudan Black B demonstrated that the material in the depressions observed with SEM was lipid. Sections of whole, milled rice kernels, prepared using a modified sectioning technique and stained with Nile Blue A, showed that portions of the embryo remain after milling and that lipid is located on or near the surface of the kernel. Differences in quantity and distribution of residual lipid as milling duration increased were documented photographically to indicate the extent to which the bran and embryo components were removed during milling. This paper provides proof of concept that residual lipid is a robust measure of the degree of milling.  相似文献

How land-application of digestate sourced from anaerobic digestion (AD) of animal waste influences the functioning of a mixed pasture agroecosystem is not well characterised, particularly with regard to the response of the actively growing microbial community. We studied the impact of the liquid AD digestate on the decomposer community in two different soils, seeded with two different common grassland crops; a mixture of either grass or grass/clover in a greenhouse experiment. We studied bacterial (leucine incorporation into bacteria) and fungal (acetate incorporation into ergosterol) growth responses to AD cattle slurry digestate, undigested cattle slurry, mineral fertiliser (NPK and N) added at a rate equivalent to 150?kg?N?ha?1, and a no-fertiliser control treatment. Differences in fungal and bacterial growth were evident between the soil and sward types. However, the fertilisers consistently stimulated a higher bacterial growth than the no-fertiliser control, and liquid digestate resulted in a level of bacterial growth higher or equal to that of mineral fertiliser, whilst undigested slurry resulted in lower bacterial growth. These fertiliser effects on bacterial growth mirrored the effects on plant growth. In contrast, the fungal community responded only marginally to fertiliser treatments. We conclude that the application of digestate stimulates the bacterial decomposer community in a similar way to that of mineral fertilisers. Our results suggest that mineral fertiliser can be exchanged for liquid digestate with limited impact on the actively growing soil microbial community that, in turn, regulate important soil processes including nutrient cycling in agricultural soils.  相似文献

The average particle size of ground grains is known to influence properties related to processing (e.g. water absorption and solubility) and nutritional value (e.g. starch digestion rate) of human foods and animal feeds. The purpose of this study was to identify the contributions made by individual size fractions of hammer-milled barley and sorghum grains to average bulk compositional, hydration, rheological, and enzyme susceptibility properties. Barley and sorghum grains were each hammer-milled through a 4 mm screen and subsequently fractionated on a set of eight sieves ranging from 0.125 mm to 2.8 mm. Individual fractions were characterised for (1) starch, aNDF, and water content, (2) water absorption index (WAI) and water solubility index (WSI), (3) viscosity profile during cooking and cooling in excess water, and (4) in vitro starch digestibility. Weighted average values based on fraction yields and property values for WAI, WSI, and starch digestibility were not significantly different from values obtained for non-fractionated ground grains of both barley and sorghum. Glucose yields from starch digestion varied about ten-fold between the smallest and largest particle fractions, and WAI and WSI had value ranges of 1.9–2.8 g/g (sorghum), 2.1–4.0 g/g (barley) and 1.3–4.5% (sorghum), 0.7–10.3% (barley), respectively. Viscosity profiles for milled sorghum grain fractions were dominated by starch swelling which became increasingly restricted as particle sizes increased. Viscosity profiles for milled barley grain fractions did not exhibit typical starch-based behaviour and were most likely dominated by soluble fibres. Taken together, the results show that there is considerable potential for designing combinations of hydration, rheological and digestibility properties of ground grains through informed selection of appropriate grains and particle size distributions.  相似文献

BACKGROUND: Water-soluble polymers are increasingly added to herbicide and pesticide formulations at very low concentrations (100-1000 mg L(-1)) in order to control the spray characteristics, notably to reduce spray drift and influence droplet bounce. The incorporation of polymeric adjuvants improves the efficacy of the spray solutions, thus enabling crop growers to maximise the performance of agrochemical sprays at lower dose rates of active ingredient. It is important to establish a fundamental understanding of how polymers influence the processes involved in droplet deposition.RESULTS: The shear and extensional viscosities of a series of high molecular mass (M(w)) poly(acrylamides) (M(w) approximately 10(6)-10(7)) have been determined at very low concentrations (100-1000 mg L(-1)). The polymer solutions demonstrated typical shear thinning characteristics under shear, and strain hardening behaviour under extension above a critical strain rate. The presence of the polymers was shown to increase the size of droplets produced in atomisation using an agricultural spray nozzle, as measured by laser diffraction. This was attributed to the increase in the extensional viscosity at the strain rates generated under pressure in the spray nozzle and was a function of both polymer concentration and M(w). In addition, the presence of polymer was found to have a significant influence on droplet bounce.CONCLUSIONS: The presence of very low concentrations of high molecular mass poly(acrylamides) significantly influences the size of droplets formed on atomisation and subsequent bounce characteristics. Large extensional viscosities generated above a critical strain rate are responsible for both processes.  相似文献

Marine protected areas can enhance fish stocks within their boundaries, but the circumstances in which they might also supplement stocks or enhance fisheries outside their boundaries are less clear. Using visual survey and fishery data, we assess the impacts of increasing fishing effort, and of the establishment in Hawaii of a network of areas closed to aquarium fishing, on the prime-target species, yellow tang (Zebrasoma flavescens), and draw conclusions about MPA impacts on long-term fishery sustainability. Between 1999, when 27.8% of the coastline was closed to collecting, and 2007, the number of active fishers and total catch of yellow tang doubled. Prior to MPA establishment, yellow tang densities were similar at sites open to fishing and those slated for closure. By 2007, closed areas had five times the density of prime targeted sized fish (5-10 cm), and 48% higher density of adults than open areas. Densities of adults in ‘boundary’ areas (open areas <1 km from nearest MPA boundary) were significantly higher than in open areas far from MPA boundaries, which was indicative of spillover at that scale. Given the long life-span of yellow tang (>40 years) relative to the duration of protection and the increasing intensity of fishing, the likelihood is that protected areas will become increasingly important sources for the adult fishes which will sustain stocks and the fishery over the longer term.  相似文献
