The role of calcium in regulating the contractile state of smooth muscle has been investigated by measuring calcium and contraction in single smooth muscle cells with the calcium-sensitive dye fura-2 and the digital imaging microscope. The concentration of free calcium in the cytoplasm increased after stimulation of the cells by depolarization with high potassium or by application of carbachol. Changes in calcium always preceded contraction. The increase in calcium induced by these stimuli was limited to less than 1 microM. Calcium within the nucleus was also subject to a limitation of its rise during contraction. Intranuclear calcium rose from 200 nM at rest to no more than 300 nM while cytoplasmic calcium rose to over 700 nM. These apparent ceilings for both cytoplasmic and intranuclear calcium may result either from negative feedback of calcium on cytoplasmic and nuclear calcium channel gating mechanisms, respectively, or from the presence of calcium pumps that are strongly activated at the calcium ceilings.  相似文献

The terminal navigation of the Near Earth Asteroid Rendezvous (NEAR) spacecraft during its close flyby of asteroid 253 Mathilde involved coordinated efforts to determine the heliocentric orbits of the spacecraft and Mathilde and then to determine the relative trajectory of the spacecraft with respect to Mathilde. The gravitational perturbation of Mathilde on the passing spacecraft was apparent in the spacecraft tracking data. As a result of the accurate targeting achieved, these data could be used to determine Mathilde's mass as 1.033 (+/- 0.044) x 10(20) grams. Coupled with a volume estimate provided by the NEAR imaging team, this mass suggests a low bulk density for Mathilde of 1.3 grams per cubic centimeter.  相似文献

A four-year study of site-specific weed control is presented in this paper. Weed seedling distribution was sampled and mapped prior to and after post-emergence herbicide application in four fields planted with maize, sugar beets, winter wheat and winter barley, rotating on one site. Herbicides and other weed control strategies were applied site-specifically according to the spatial variation of weed populations. Different decision algorithms were used for chemical weed control methods in each crop. A weed treatment map was created to direct location and dosage of herbicide application. The sprayer was coupled with a differential Global Positioning System (DGPS). The solenoid valves of the sprayer were opened automatically when the tractor entered a weed patch characterized in the weed treatment map. For site specific herbicide application, a patch sprayer was developed that allowed variable rate application and the selective control of each section of the spray boom.  相似文献

With modern machinery and modem techniques of grass production, mechanical-or zero-grazing offers a means of utilizing the crop to perfection. Whether this results in enhanced profits depends largely upon the farm and the specific circumstances. It may, it may not. What is evident is that the health of the cows is not affected by an unnatural manner of summer feeding; the sward, per se, may be improved by the regular and uniform defoliation; and under conditions of intensive production and the vital need to use profitably every blade of grass, as for instance on small, highly-rented farms, the practice of zero-grazing merits the most careful consideration. It cannot lightly be dismissed as merely a new fad.  相似文献

An experiment is reported in which two bunker silos were filled with about 12 t of ryegrass; one was compacted with a surface pressure of 4·1 kPa and the other was not. Both were instrumented to record temperatures and gas compositions during fermentation and feed-out. Cores of silage (100 mm diameter) were removed from the bunkers and subjected to forced aeration in insulated cylinders. Simple mathematical models were used to simulate CO2 leakage from the silos and the associated loss of dry matter, and temperature changes during the forced aeration of silage cores. The leakage of CO2 from the silos, as shown by the fall in CO2 concentration with time, could be described empirically by an exponential equation and could be simulated with a simple mathematical model. Experimental evidence supported the hypothesis that permeation was the main method of gaseous exchange during fermentation and feed-out, but gas mixing by diffusion and/or convection also occurred within the silos during fermentation. Dry-matter losses by aerobic activity during storage of 120–150 d were conservatively estimated to be about 0·3% and 0·9% for the uncompacted and compacted bunkers respectively. These were considered to be lower than those that could be expected in farm silos, because the sealing was likely to be more thorough. The silages were also more stable, when subject to aeration, than others examined in this laboratory, probably because of high contents of acetic, propionic and butyric acids. Simple mathematical models were of value, but greater sophistication is needed (e.g. multi-compartmental models) to deal comprehensively with the heat and gas flows found in the complex biophysical systems of silage.  相似文献

Summary Experiments were conducted on sandy and deep silty soils to assess the effect of physiological age of seed tubers and planting date on the response of potatoes to nitrogen fertiliser. Nitrogen optima commonly had large standard errors which made interpretation of the data difficult. On the sandy soil with cv. Record, early planting increased yield in two years but had no effect on mean N optimum (303 kg ha−1, NOpt). Mean NOpt was less on the silty soil where the cv. Pentland Squire was grown, and was reduced from 273 kg ha−1 to 241 kg ha−1 when planting was delayed by 4 weeks. Physiological age had no effect on NOpt. On the sandy soil, residual soil mineral nitrogen (RSMN) increased by 0.05 kg ha−1 per kg N fertiliser applied; on the silty soil the increase was four times as large. RSMN was larger following the late planted crop on the silty soil in one year.  相似文献

This paper explores whether spatial variation in the biodiversity values of vertebrates and plants (species richness, range-size rarity and number or proportion of IUCN Red Listed threatened species) of three African tropical mountain ranges (Eastern Arc, Albertine Rift and Cameroon-Nigeria mountains within the Biafran Forests and Highlands) co-vary with proxy measures of threat (human population density and human infrastructure). We find that species richness, range-size rarity, and threatened species scores are all significantly higher in these three tropical African mountain ranges than across the rest of sub-Saharan Africa. When compared with the rest of sub-Saharan Africa, human population density is only significantly higher in the Albertine Rift mountains, whereas human infrastructure is only significantly higher in the Albertine Rift and the Cameroon-Nigeria mountains. Statistically there are strong positive correlations between human density and species richness, endemism and density or proportion of threatened species across the three tropical African mountain ranges, and all of sub-Saharan Africa. Kendall partial rank-order correlation shows that across the African tropical mountains human population density, but not human infrastructure, best correlates with biodiversity values. This is not the case across all of sub-Saharan Africa where human density and human infrastructure both correlate almost equally well with biodiversity values. The primary conservation challenge in the African tropical mountains is a fairly dense and poor rural population that is reliant on farming for their livelihood. Conservation strategies have to address agricultural production and expansion, in some cases across the boundaries and into existing reserves. Strategies also have to maintain, or finalise, an adequate protected area network. Such strategies cannot be implemented in conflict with the local population, but have to find ways to provide benefits to the people living adjacent to the remaining forested areas, in return for their assistance in conserving the forest habitats, their biodiversity, and their ecosystem functions.  相似文献

A statistical mixed effects model was applied to data from a two-phased experiment in which cholinesterase activity (CA) in red blood cells (RBCs) of rats was monitored following single-and double-gavagead ministrations of the carbamate pesticide, Aldicarb. Our goals were to develop and estimate a nonlinear mixed effects model for describing the inhibition and recovery pattern of CA as a continuous function of time and Aldicarb dose, and to use the results to characterize both the typical-animal CA response and its variability across animals. The Phase 1 experiment involved adult male CD® rats randomly assigned to a control and two dose groups. Blood samples were taken via jugular cannula prior to exposure and at 11 post-dose time points (over six hours) for determination of RBC CA. The Phase II experiment was similar, but involved a second administration approximately 4.5 hours after the first. An open, one-compartment model with first-order kinetics appeared to provide an adequate structural basis for the statistical nonlinear mixed effects model, which was fit to the RBC data of the 26 treatment-group rats from both phases. For the “typical” animal, the model predicts that about a 20% (40%) maximal reduction in activity will occur for the low (high) dose group and that recovery to 90% of the baseline level will occur in 105 minutes (179 minutes) for the low (high) dose group. Based on simulated distributions derived from the model’s variance-covariance matrix of random effects, we estimated that about 99% of animals are expected to require less than 143 minutes (267 minutes) to recover to 90% of their baseline level for the low (high) dose group. Because virtually all animals had returned to near pre-exposure, levels by the time of the second administration, the level of the first dosing had little impact on the CA patterns following the second dose.  相似文献
