The starch granule surface is a frontline of microbial attack and defence, operating in the background of normal starch granule metabolism. Puroindoline, a wheat protein which binds starch granule surfaces, contains a unique tryptophan-rich domain likely responsible for this property, though direct evidence is lacking. To test puroindoline’s tight association, prime starch granule extracts were water-washed 8 or 20 times and residual puroindoline removed using a solution of 50% isopropanol/50 mM NaCl. We found that this solvent was consistent in the amount of protein extracted from wheat flour and washed starch, regardless of initial protein content. Relative quantification of puroindoline following water-washing was performed using dot blot. Washing more than 8 times did not further reduce puroindoline content of starch granules suggesting a strong association with the starch granule surface. To identify the tryptophan-rich domain tightly associated with the starch granule surface, a combination of in situ tryptic digestion and mass spectrometry was used. Following digestion and water-washing, 50% isopropanol/50 mM NaCl was used to remove tightly-associated peptides for identification by mass spectrometry. Using this method, we identified the tryptophan-rich domain of puroindoline directly bound to the starch granule surface of wheat.  相似文献

Abstract

The use of one source of cytoplasmic male steriity, leading to widespread uniformity of maize, coupled with the appearance of a new virulent race of Helminthosporium maydis made the Southern Corn Leaf Blight epidemic possible. The weather during 1970 was very suitable for the spread of the disease and thus the epidemic occurred. The possible role of cytoplasmic DNA, carried most probably by the mitochondria, in the susceptibility or resistance of various maize lines in discussed. The relationship of cytoplasmic male sterility and disease resistance or susceptibility is considered. The effects of the pathotoxin, produced by H. maydis, its uses as an experimental tool in research, and its potential uses in further investigations are described in detail.  相似文献

The productive potential of potatoes (Solanum tuberosum L. cvs. Norland, Superior, Norchip, and Kennebec) was assessed for life support systems being proposed for space stations and/or lunar colonies. Plants were grown in walk-in growth rooms for 15 weeks at 20 C under 12-, 16- and 20-h photoperiods of 400 μmol mt-2st-1 photosynthetic photon flux (PPF). Norland yielded the greatest tuber fresh weight, producing 2.3, 2.4, and 2.9 kg/plant under 12-, 16-, and 20-h photoperiods, respectively. The respective yields for the other cultivars under 12-, 16-, and 20-h were: Superior, 1.9, 1.5, and 1.8 kg/plant; Norchip, 1.8, 1.4, and 2.0 kg/plant; and Kennebec, 2.3, 0.2, and 0.8 kg/plant. Shoot and total plant biomass increased with lengthening photoperiods except for Kennebec, which showed increased shoot growth but no change in total growth with the longer photoperiods. Kennebec shoot growth under the 20-h photoperiod, and to some extent under 16-h, was noticeably stunted with shortened internodes. In addition, leaves of these plants showed mild chlorosis with rusty “flecking” of the surfaces. The harvest index (ratio of tuber yield/total biomass) was highest for all cultivars under the 12-h photoperiod, with a maximum of 0.69 for Norland. Similarly, the tuber yield per input of irradiant energy also was highest under 12-h for all cultivars. The tuber yield expressed on an area basis for the highest yielding treatment (Norland under 20-h) equaled 2.2 kg dry matter mt-2. Over 15 weeks this equates to a productivity of 20.7 g tuber dry matter mt-2 dayt-1. Assuming 3.73 kcal per g tuber dry matter and a daily human dietary requirement of 2800 kcal, then 36 m2 of potatoes could supply the daily energy requirement for one human. The differences in soil fertility in a ley as a result of treatments applied for four years in a sheep-grazing trial were measured by growing crops of wheat and kale. The pasture treatments had been: control, dung, urine and dung plus urine, each combined with four levels of fertilizer nitrogen.
Yields of winter wheat harvested in 1956 ranged from 27.1 to 38.6 cwt. per acre. Plots to which faeces had been allowed to return during the pasture phase outyielded (p <0.001) those from which it was withheld. Urine was relatively ineffective except in combination with nitrogen. In the absence of animal returns, nitrogenous fertilizer depressed grain and straw yields. A top-dressing of K applied to the wheat in spring as a sub-treatment had no significant effect. The percentages of N and K in the grain were unaffected by the former pasture treatments, or by the K top-dressing.
A subsidiary small-plot experiment in which the above pasture treatments had been combined with P and K, each at two levels, was cropped with marrow-stem kale. In general, N applied to the pasture increased kale yields except where dung and urine had been withheld. Neither P nor K had a significant effect, except when combined. The leaf/stem ratio of the kale was reduced by applied N and by P (both p <0.05).

This study evaluated the use of visible and near-infrared (VISNIR) spectroscopy to predict lean color stability in pork loin chops. Spectra were collected immediately after and approximately 1 h after rib removal on 1,208 loins. Loins were aged for 14 d before a 2.54-cm chop was placed in simulated retail display. Spectra were collected on aged loins immediately after removal from the vacuum package and on chops 10 min after cutting. Instrumental color measurements [L*, a*, b*, hue angle, chroma, and E (overall color change)] were determined on d 0, 1, 7, 11, and 14 of display. Principal components analysis of display d 0 and 14 values of these traits identified a factor (first principal component; PC1) explaining 67% of the variance that was related to color change. Partial least squares regression was used to develop 3 models to predict PC1 values by using VISNIR spectra collected in the plant, on aged loins, and on chops. Loins with predicted PC1 values less than 0 were classified as having a stable color, whereas values greater than 0 were classified as having a labile lean color. Loins classified as stable by the in-plant model had smaller (P < 0.05) L* values than those classified as labile. Hue angle and ΔE values were less (P < 0.05) and a* and chroma values were greater (P < 0.05) after d 7 of display in loins predicted to have a stable color than in loins predicted to have a labile lean color. Similarly, chops from loins classified as stable using the aged loin model had smaller (P < 0.05) L* values than those from loins classified as labile. Furthermore, loins predicted to be stable had smaller (P < 0.05) hue angle and ΔE values and greater (P < 0.05) a* and chroma values after d 7 of display than did loins predicted to be labile. Results for the chop model were similar to those from the 2 loin models. Chops predicted to have a stable lean color had smaller (P < 0.05) L* values than did those predicted to have a labile lean color. Chops classified as stable had smaller (P < 0.05) hue angle and ΔE values and greater (P < 0.05) a* and chroma values after d 7 of display compared with chops classified as labile. All 3 models effectively segregated chops based on color stability, particularly with regard to redness. Regardless of the model being used, d 14 display values for a*, hue angle, and ΔE in loins classified as stable were similar to the d 7 values of loins classified as labile. Thus, these results suggest that VISNIR spectroscopy would be an effective technology for sorting pork loins with regard to lean color stability.  相似文献

Chlamydia psittaci was detected by PCR in the lung and equine foetal membranes of two aborted equine foetuses and one weak foal from two different studs in Victoria, Australia. The abortions occurred in September 2019 in two mares sharing a paddock northeast of Melbourne. The weak foal was born in October 2019 in a similar geographical region and died soon after birth despite receiving veterinary care. The detection of C. psittaci DNA in the lung and equine foetal membranes of the aborted or weak foals and the absence of any other factors that are commonly associated with abortion or neonatal death suggest that this pathogen may be the cause of the reproductive loss. The detection of C. psittaci in these cases is consistent with the recent detection of C. psittaci in association with equine abortion in New South Wales. These cases in Victoria show that C. psittaci, and the zoonotic risk it poses, should be considered in association with equine reproductive loss in other areas of Australia.  相似文献
