Objective— To evaluate outcome after closed fluoroscopic-assisted application of spinal arch external skeletal fixators in dogs with vertebral column injuries.
Study Design— Retrospective case series.
Animals— Dogs with traumatic vertebral column injuries (n=5).
Methods— Medical records of dogs with vertebral column fractures and/or luxations stabilized with spinal arch external skeletal fixator frames applied using a closed fluoroscopic-assisted technique were reviewed. Owners were contacted to obtain long-term clinical outcomes.
Results— Five dogs (age range, 6–72 months; weight, 10–54 kg) had traumatic vertebral column injuries stabilized with spinal arch external skeletal fixators applied in closed fashion. Injuries involved vertebral segments of the thoracolumbar junction, lumbar spine, and lumbosacral junction. Immediately postoperatively, 4 dogs had anatomic alignment of their vertebral fracture/luxation; 1 dog had 1 mm of vertebral canal height compromise. Time to fixator removal ranged from 65 to 282 days (141±87 days). All dogs had regained satisfactory neurologic function by 3 months. At long-term follow-up (range, 282–780 days; mean 445±190 days) all dogs were judged to have good to excellent return of function by their owners.
Conclusion— Successful closed fluoroscopic-assisted application of external skeletal fixators using spinal arches provided satisfactory reduction with few complications in 5 dogs. Return to function was judged to be good to excellent in all dogs at long-term evaluation.
Clinical Relevance— Closed fluoroscopic-assisted application of ESF using spinal arches provided satisfactory reduction and effective stabilization of spinal fractures with few complications and should be considered as a treatment approach.  相似文献

The sensitivity and specificity of recombinant open reading frame 5 products used in the Western blotting assay for confirmation of porcine reproductive and respiratory syndrome virus (PRRSV) serologic status were evaluated. The recombinant antigen-based assays were specifically compared with a commercial enzyme-linked immunosorbent assay (ELISA) for PRRSV antibodies using 1) PRRSV antibody-negative reference sera (n = 30), 2) naturally infected pig sera (n = 40), 3) sequential sera obtained from 24 experimentally infected pigs, and 4) sera submitted to 3 state diagnostic laboratories (n = 200). The recombinant antigen assay yielded an average increased sensitivity of 10% over the commercial PRRSV ELISA. The negative controls (group 1 sera) showed no difference between the 2 assays. This comparison confirmed that the recombinant antigen-specific assay was more sensitive than the commercial ELISA and is well suited for routine confirmation of the presence of PRRSV antibodies.  相似文献

The objective of this experiment was to determine the effect of sarcomere length on postmortem proteolysis and meat tenderness. Eighteen Dorset market-weight sheep were slaughtered conventionally. The longissimus thoracis et lumborum and psoas major from each carcass were either left intact on the carcass (control), which was chilled at 0 degrees C, or excised from the carcass and chilled in an ice slurry (0 degrees C). At 24 h, control muscles were excised, and all muscles were cut into sections and assigned to 1 or 10 d of postmortem storage at 2 degrees C. Sarcomere length was shorter (P < .01), as intended, in the shortened relative to the control treatment and in longissimus relative to psoas major (1.36 vs 1.69 microm, raw longissimus; 1.45 vs 3.03 microm, raw psoas major). Sarcomere length was not affected (P > .05) by aging time. Western blot analysis of troponin-T and desmin indicated no effect (P > .05) of the shortened treatment compared to the control on the extent of proteolysis. Regardless of aging time or treatment, troponin-T was more degraded (P < .01) in longissimus than in psoas major (38.1 vs 23.5%) and desmin tended to be more degraded (P = .08) in longissimus than in psoas major (50.4 vs 35.1%). Regardless of muscle or treatment, aging 10 d compared to 1 d increased degradation of troponin-T (46.3 vs 15.3%) and desmin (69.3 vs 16.1%). Warner-Bratzler shear force was greater (P < .01) in the shortened treatment than in control (6.9 vs 3.8 kg), greater (P < .01) in longissimus than in the psoas major (6.5 vs 4.2 kg), and greater (P < .01) with 1 d than with 10 d of aging time (6.1 vs 4.6 kg). A muscle x aging time interaction (P < .05) indicated shear force declined more in longissimus than in psoas major during aging. We conclude that sarcomere length did not affect the extent of proteolysis. However, sarcomere length may have an indirect effect on tenderization during aging due to its effect on initial tenderness.  相似文献

Experiments were conducted to develop an optimal protocol for measurement of slice shear force (SSF) and to evaluate SSF as an objective method of assessing beef longissimus tenderness. Whereas six cylindrical, 1.27-cm-diameter cores are typically removed from each steak for Warner-Bratzler shear force (WBSF) determination, a single 1-cm-thick, 5-cm-long slice is removed from the lateral end of each longissimus steak for SSF. For either technique, samples are removed parallel to the muscle fiber orientation and sheared across the fibers. Whereas WBSF uses a V-shaped blade, SSF uses a flat blade with the same thickness (1.016 mm) and degree of bevel (half-round) on the shearing edge. In Exp. 1, longissimus steaks were acquired from 60 beef carcasses to determine the effects of belt grill cooking rate (very rapid vs. rapid) and conditions of SSF measurement (hot vs cold) on the relationship of SSF with trained sensory panel (TSP) tenderness rating. Slice shear force was more strongly correlated with TSP tenderness rating when SSF measurement was conducted immediately after cooking (r = -.74 to -.76) than when steaks were chilled (24 h, 4 degrees C) before SSF measurement (r = -.57 to -.72). When SSF measurement was conducted immediately after cooking, the relationship of SSF with TSP tenderness rating did not differ among the belt grill cooking protocols used to cook the SSF steak. In Exp. 2, longissimus steaks were acquired from 479 beef carcasses to compare the ability of SSF and WBSF of 1.27-cm-diameter cores to predict TSP tenderness ratings. Slice shear force was more strongly correlated with sensory panel tenderness rating than was WBSF (r = -.82 vs -.77). In Exp. 3, longissimus steaks were acquired from 110 beef carcasses to evaluate the repeatability (.91) of SSF over a broad range of tenderness. Slice shear force is a more rapid, more accurate, and technically less difficult technique than WBSF. Use of the SSF technique could facilitate the collection of more accurate data and should allow the detection of treatment differences with reduced numbers of observations and reduced time requirements, thereby reducing research costs.  相似文献

A resource flock of 362 F2 lambs provided phenotypic and genotypic data to estimate effects of callipyge (CLPG) genotypes (NN, NC, CN, and CC) on meat quality traits. The mutant allele is represented as C, the normal allele(s) as N, and the paternal allele of a genotype is given first. Lambs of each genotype born in 1994 and 1995 were serially slaughtered in six groups at 3-wk intervals starting at 23 wk of age. Warner-Bratzler shear force and subjective evaluation of marbling were collected during both years from longissimus. Calpastatin activity was measured on longissimus from the 1994 group, and ELISA quantification of calpastatin protein was obtained from the 1995 group. Significant additive and paternal polar overdominance effects on meat quality traits were detected. This is in contrast to previous research that detected only polar overdominance effects on slaughter and carcass traits in this population. The magnitude of genotypic effects on shear force differed significantly between years; however, additive (P < .01), paternal polar overdominance (P < .001), and maternal dominance (P < .01) effects adjusted for variation in carcass weight were detected within each year. Shear force data adjusted to the mean slaughter age or carcass weight indicated that the means and variances of CN and CC genotypes were greater than values of NC and NN. Shear force values were greatest for CN and were intermediate for CC. The difference in shear force (adjusted for variation in slaughter age) between homozygous genotypes (additive effect) was supported by calpastatin activity data with 2-df F-tests of 3.66 (P < .05) and 11.84 (P < .001) at d 0 and 7 postmortem, respectively. Corresponding values for the paternal polar overdominance effects on calpastatin activity were 53.80 (P < .001) and 87.43 (P < .001). Calpastatin ELISA data (d 0, adjusted for slaughter age) exhibited a paternal polar overdominance effect exclusively with a 2-df F-test of 57.63 (P < .001). Additive and paternal polar overdominance effects on marbling adjusted for slaughter age had F-tests of 6.41 (P < .01) and 93.29 (P < .001), respectively. Consequences of increased longissimus shear force must be addressed if the advantages of CN lambs for dressing percentage and carcass composition are to be realized. Further research is needed to establish whether selection targeted at changing the background genome can mitigate the negative effects of the C allele on meat tenderness.  相似文献

This paper examines the changing profile of water traders (both allocation and entitlement traders) in the Goulburn-Murray Irrigation District in Australia, and examines the efficiency of the water allocation and entitlement markets from 1998-99 to 2003-06. The results suggest that the profile of traders in the early and mature stages of the water allocation market differ greatly. In addition, the profile of allocation traders is significantly dissimilar from that of water entitlement traders at all stages of water market development. The decision to buy or sell water allocations was more likely to be associated with a farmer's socioeconomic characteristics and the type of farm, while the decision to buy or sell water entitlements was more likely to be associated with the extent of existing farm infrastructure and farm productivity. Finally, there was strong evidence to suggest that trading in the water allocation market has become more efficient over time, though there is no evidence to suggest the same for the water entitlement market.  相似文献

The new powdery mildew fungicide quinoxyfen belongs to the novel quinoline class of chemistry. Although its biochemical mode of action is unknown, quinoxyfen does not act in the same way as other cereal fungicides. It is a systemic protectant which inhibits the early stages of mildew infection on a wide range of crops, and provides season-long protection from a single early-season spray applied around GS 31. The base-line sensitivity profile of quinoxyfen was defined for barley powdery mildew (Erysiphe graminis f.sp. hordei) from over 340 field isolates collected from different parts of the UK from 1991 onwards. Sensitivities ranged from <0·0001→0·16 mg litre-1 with a mean of 0·003 mg litre-1. Current work is extending the base-line sensitivity studies to wheat powdery mildew (E. graminis f.sp. tritici), and includes isolates from European trials, but so far this new data set has shown no differences from barley powdery mildew. Quinoxyfen-resistant mutants were generated in the laboratory, and some similar resistant strains were obtained from treated field crops. These laboratory and field strains were always defective, in some way, for sporulation and, curiously, all required the presence of quinoxyfen for survival in culture. Attempts to generate resistant mutants that sporulated normally were unsuccessful. These studies suggested that the resistance risk for quinoxyfen is low. The recommended anti-resistance strategy accompanying introduction of quinoxyfen avoids seed treatments and late-season applications. Instead, a single early (GS 31) treatment using either pre-formulated mixtures or alternating with a fungicide with different mode of action is recommended. This strategy will be supported by continued monitoring of wheat and barley powdery mildew. ©1997 SCI  相似文献

It is believed that milk production is determined by the number and activity of mammary secretory cells. Secretory activity, as assessed by milk volume, depends on secretion of the major osmole in milk, lactose, which is produced by lactose synthase. The amount of either of the two proteins in lactose synthase may regulate milk production. The objective of this study was to determine whether the concentrations in milk of the two components of lactose synthase, α‐lactalbumin (α‐LA) and β1,4‐galactosyltransferase (B4GALT), were related to genetic background, stage of lactation, breed or parity of dairy cows. α‐Lactalbumin and B4GALT concentrations were measured by ELISA and by enzyme assays, respectively, from single milk samples. Two herds with a total of 279 cows were used in the analysis. One herd contained Ayrshire, Brown Swiss, Holstein and Jersey cows; the second herd contained two groups of cows; Holsteins selected for high milk production and Holsteins with 1960s genetics. The α‐LA concentration in milk was greater in Jerseys and Ayrshires than in Holsteins and Brown Swiss. However, no difference in α‐LA concentration was observed in milk from high and low genetic merit cows in the Minnesota herd or among different genetic backgrounds in the Illinois herd. β1,4‐Galactosyltransferase concentrations were similar for all groups that were analyzed. α‐Lactalbumin concentrations were positively correlated with milk protein concentration, milk fat concentration and lactose concentration. β1,4‐Galactosyltransferase concentration in milk exhibited a strong positive correlation with number of days in milk. Although the concentration of B4GALT increased as lactation progressed, the values did not show any correlation with persistency of lactation or late lactation milk production. In conclusion, this survey shows that the two components of lactose synthase are each correlated to protein concentration and individually correlated to the concentration of other milk components and stage of lactation.  相似文献
