Anatid herpesvirus 1 (AHV-1) CH virulent strain was first isolated from an infected duck and it was found that this virus strain could induce cytopathic effect (CPE) in duck embryo fibroblast (DEF). Following AHV-1 infection, DEF showed morphological changes such as cell rounding, improved refractivity and detachment from the culture surface. However, its pathological characteristics were not adequately known. Related studies were performed and the results showed that syncytium formation could be observed as the other type of CPE in AHV-1 infection. Hematoxylin-eosin staining and 4’, 6-diamidino-2-phenylindole (DAPI) staining of infected DEF were each used to visualize the shape and distribution of chromatin within nuclei and nuclear fragmentation was observed. Chromatin condensation and margination, as well as formation of apoptotic bodies were observed by transmission electron microscopy (TEM). DNA ladder formation was detected in AHV-1 infected cells and apoptosis of the infected DEF was also detected by flow cytometry analysis of Annexin V-FITC/PI staining method. Therefore, it was suggested that AHV-1 virulent strain can induce syncytium and apoptosis in DEF. Syncytium formation and apoptosis observed in this study may contribute to the elucidation of AHV-1 pathogenesis. 相似文献

为了探索不同酶系组成对玉米弯孢叶斑病菌原生质体制备的影响,建立该病菌原生质体遗传转化系统,采用酶系混合物裂解菌丝体制备原生质体,采用PEG介导方法进行原生质遗传转化,通过PCR和Southern blotting技术对转化子进行验证。结果表明:1%溶壁酶+1%蜗牛酶+1%纤维素酶混合酶系为玉米弯孢叶斑病菌原生质体制备的最佳酶系,可以产生原生质体6.78×106 cfu/mL。用PEG介导方法转化共获得16个稳定的转化子,从中随机挑取5个转化子发现质粒pV2已被成功整合到基因组中。本研究获得了制备玉米弯孢叶斑病菌原生质体的最佳酶系,建立了PEG介导的原生质体遗传转化体系,为开展该菌致病相关基因克隆和基因功能研究提供了一种手段。 相似文献

鸡球虫病是影响养鸡生产的重要寄生虫病,试验采用单味钩藤和以钩藤为主的中草药复方制剂进行抗球虫病的疗效试验。按给药剂量分为9组,每组10羽,1、2、3组给药剂量分别为0.5ml、1.0ml、1.5ml的单味钩藤中药组,4、5、6组则以钩藤为主的中草药复方制剂组及地克珠利组、感染不给药组和不感染不给药组。结果表明,单味钩藤中药1、2、3组和以钩藤为主的中草药复方制剂4、5、6组、地克珠利组的抗球虫指数(ACI)分别为177.1、181.4、182.1;179.9、182.4、188.5;185.4,单味钩藤中药1组和以钩藤为主的中草药复方制剂4组为轻度抗荮陛,其余均为敏感。结果表明,钩藤对抗鸡柔嫩艾美尔球虫具有显著疗效。由于中草药具有毒性低,使用安全、残留少等优点,在防制鸡球虫病方面具有良好的应用前景。 相似文献

对不同含水量的菜豆种子( 2.1%、3.4%、3.8%、6.9%和9.0% ) 老化过程中的生理特性进行了研究。结果表明, 菜豆种子在含水量为318%时, 发芽率, 保护酶系统( SOD、POD和CAT) 活性和脱氢酶活性明显高于其它含水量的种子, 而脂氧合酶(LOX) 活性和丙二醛(MDA) 含量显著低于其它含水量的种子, 表现出较高的生活力。此结果表明适度降低菜豆种子的含水量可以提高其抗老化劣变能力;不同生活力的菜豆种子POD同工酶酶谱表现不同, 而SDS2PAGE蛋白质谱带则基本相同。 相似文献

This study was conducted to investigate the effects of compound p1ant nutrients (CPN) on rumen fermentation parameters, rumen microbiota and fatty acid composition of longissimus dorsi musc1e in finishing sma11 tai1 Han sheep. Sixteen 4-month-o1d finishing sma11 tai1 Han sheep with an initia1 body weight (BW) of (24.18±0.31) kg were random1y divided into two groups, name1y, with 8 rep1icates per group and 1 sheep per rep1icate. The sheep in the contro1 group were fed a basa1 diet, whereas the sheep of the contro1 group (CPN group) was fed the basa1 diet supp1ementation with 3‰ CPN. The experiment 1asted for 97 days after 7 days adaption. The resu1ts showed as fo11ows: compared with the contro1 group, 1) adding CPN decreased the concentration of ammonia nitrogen (NH3-N) (P<0.05); 2) CPN supp1ementation affected beta diversity of rumen microbiota; 3) the re1ative abundance of Acidobacteriota, Erysipe1otrichaceae_UCG-002, Lactobacillus, and Megasphaera were enhanced by adding CPN (P<0.05), whereas, the re1ative abundance of Bacteroidetes and Prevotella was 1ower (P<0.05); 4) the supp1ementation of CPN had no significant effect on fatty acid composition of longissimus dorsi musc1e of fattening sma11-tai1ed Han sheep (P>0.05); 5) the contents of tota1 MUFA, C18:1n9c, C14:1, C16:1, C18:2n6c, C18:3n3 and n-3PUFA in the longissimus dorsi musc1e were corre1ated with the re1ative abundance of Megasphaera, Erysipe1otrichaceae_UCG-002, Succiniclasticum and Ruminococcus (P<0.05). In conc1usion, CPN can regu1ate the rumen microbiota structure and reduce the rumen NH3-N concentration of fattening sma11-tai1ed Han sheep. A field experiment was conducted to study the effects of combination of straw incorporation and water management on fluxes of CH4, N2O and soil heterotrophic respiration (Rh) in a paddy field in subtropical central China by using a static opaque chamber/gas chromatography method. Four treatments were set up: two rice straw incorporation rates at 0 (S1) and 6 (S2) t ha?1 combined with two water managements of intermittent irrigation (W1, with mid-season drainage) and continuous flooding (W2, without mid-season drainage). The cumulative seasonal CH4 emissions for the treatments of S1W2, S2W1 and S2W2 increased significantly by 1.84, 5.47 and 6.63 times, respectively, while seasonal N2O emissions decreased by 0.67, 0.29 and 1.21 times, respectively, as compared to S1W1 treatment. The significant increase in the cumulative Rh for the treatments S1W1, S2W1 and S2W2 were 0.54, 1.35 and 0.52 times, respectively, in comparison with S1W2. On a seasonal basis, both the CO2-equivalents (CO2e) and yield-scaled CO2e (GHGI) of CH4 and N2O emissions increased with straw incorporation and continuous flooding, following the order: S2W2>S2W1>S1W2>S1W1. Thus, the practices of in season straw incorporation should be discouraged, while mid-season drainage is recommended in paddy rice production from a point view of reducing greenhouse gas emissions. 相似文献
