Weeds resistant to the s-triazine herbicide atrazine also show resistance to the triazinone herbicide metribuzin. However, with highly lipophilic triazinones, thylakoids isolated from atrazine-resistant Amaranthus retroflexus (mutation at position Ser264 of the photosystem II D-1 reaction centre protein) in general show a higher pI50 value in photosystem II electron transport than those from the wild type (i.e. negative cross-resistance; ‘supersensitivity’). A quantitative structure–activity relationship (QSAR) can be established, wherein the lipophilicity of the compound plays a major role. In in-vivo experiments, it was found that the triazinone DRW2698 killed resistant Amaranthus retroflexus and Chenopodium album whereas the wild type was almost unaffected. Triazinones were further investigated in five different mutants of Chlamydomonas rheinhardtii (mutations in the D-1 protein at positions Ser264, Ala251, Leu275, Phe255, and Val219). Inhibitory activity of all triazinones was generally enhanced in the Phe255 mutant but decreased in the Val219 mutant. In the other mutants, biological activity was decreased when position 3 of the triazinone was substituted by CH3, OCH3, SCH3, NHCH3 or N(CH3)2. However, negative cross-resistance was again observed when this position was occupied by free thiol. It is therefore suggested that these two groups of triazinones orient themselves differently within the herbicide binding niche of the photosystem II D-1 protein.  相似文献

The effects of adjuvants on the performance of a dispersible concentrate formulation (DCI) of a novel powdery mildew fungicide, 1-(4-chlorobenzyl)-4-phenylpiperidine (I) were investigated. The method involved assessment, under glasshouse conditions, of the therapeutic (curative) control of infections of powdery mildew (Erysiphe graminis DC f.sp. hordei Marchal) on barley (Hordeum vulgare, L.) eight to nine days after spray application to plants that had been inoculated one day prior to spraying. The results from the first trial showed that marked improvements (∽five-fold) in the performance of DCI could be obtained by the spray tank addition of some types of surfactant adjuvants and a series of further trials investigating a wide range of adjuvants was conducted. Nonylphenol, alkylamine and alcohol ethoxylates varying in mean ethylene oxide content between 5 and 20 moles were highly effective. There were indications that optimum performance enhancements were obtained with these surfactants containing between 5 and 10 moles ethylene oxide. Lower enhancements, sometimes only marginal, were obtained from trisiloxane, phosphate ester, propylene oxide, alkylamine-propylene oxide and castor oil ethoxylates and also alkyl polyglucoside biosurfactants. Negligible adjuvant enhancements were observed with emulsifiable paraffinic/naphthenic and rape seed oils, though slightly better enhancements were seen with an emulsifiable transmethylated rape seed oil and, interestingly, larger enhancements with an emulsifiable lipophilic alcohol. A final trial identified the alcohol ethoxylate, ‘Dobanol’ 91-6, as the most effective adjuvant and that its optimum application rate under glasshouse conditions was 250 g ha-1. Genome-sequencing studies indicate that all humans carry many genetic variants predicted to cause loss of function (LoF) of protein-coding genes, suggesting unexpected redundancy in the human genome. Here we apply stringent filters to 2951 putative LoF variants obtained from 185 human genomes to determine their true prevalence and properties. We estimate that human genomes typically contain ~100 genuine LoF variants with ~20 genes completely inactivated. We identify rare and likely deleterious LoF alleles, including 26 known and 21 predicted severe disease-causing variants, as well as common LoF variants in nonessential genes. We describe functional and evolutionary differences between LoF-tolerant and recessive disease genes and a method for using these differences to prioritize candidate genes found in clinical sequencing studies.  相似文献

Land use and mineral characteristics affect the ability of surface as well as subsurface soils to sequester organic carbon and their contribution to mitigation of the greenhouse effect. There is less information about the effects of land use and soil properties on the amount and composition of organic matter (OM) for subsurface soils as compared with surface soils. Here we aimed to analyse the long‐term (≥ 100 years) impact of arable and forest land use and soil mineral characteristics on subsurface soil organic carbon (SOC) contents, as well as on amount and composition of OM sequentially separated by Na pyrophosphate solution (OM(PY)) from subsurface soil samples. Seven soils with different mineral characteristics (Albic and Haplic Luvisol, Colluvic and Haplic Regosol, Haplic and Vertic Cambisol, Haplic Stagnosol) were selected from within Germany. Soil samples were taken from subsurface horizons of forest and adjacent arable sites continuously used for >100 years. The OM(PY) fractions were analysed for their OC content (OCPY) and characterized by Fourier transform infrared spectroscopy. Multiple regression analyses for the arable subsurface soils indicated significant positive relationships between the SOC contents and combined effects of the (i) exchangeable Ca (Caex) and oxalate‐soluble Fe (Feox) and (ii) the Caex and Alox contents. For these soils the increase in OC (OCPY multiplied by the relative C=O content of OM(PY)) and increasing contents of Caex indicated that OM(PY) mainly interacts with Ca2+. For the forest subsurface soils (pH < 5), the OCPY contents were related to the contents of Na‐pyrophosphate‐soluble Fe and Al. The long‐term arable and forest land use seems to result in different OM(PY)‐mineral interactions in subsurface soils. On the basis of this, we hypothesize that a long‐term land‐use change from arable to forest may lead to a shift from mainly OM(PY)‐Ca2+ to mainly OM(PY)‐Fe3+ and ‐Al3+ interactions if the pH of subsurface soils significantly decreases to <5.  相似文献

Dendroclimatological studies in a dry inner Alpine environment (750 m a.s.l.) revealed different growth response of co-occurring coniferous species to climate, which is assumed to be caused by a temporal shift in wood formation among species. The main focus of this study therefore was to monitor intra-annual dynamics of radial increment growth of mature deciduous and evergreen coniferous species (Pinus sylvestris, Larix decidua and Picea abies) during two consecutive years with contrasting climatic conditions. Radial stem growth was continuously followed by band dendrometers and modelled using Gompertz functions to determine time of maximum growth. Histological analyses of tree ring formation allowed determination of temporal dynamics of cambial activity and xylem cell development. Daily fluctuations in stem radius and radial stem increments were extracted from dendrometer traces, and correlations with environmental variables were performed. While a shift in temporal dynamics of radial growth onset and cessation was detected among co-occurring species, intra-annual radial growth peaked synchronously in late May 2011 and early June 2012. Moist atmospheric conditions, i.e. high relative air humidity, low vapour pressure deficit and low air temperature during the main growing period, favoured radial stem increment of all species. Soil water content and soil temperature were not significantly related to radial growth. Although a temporal shift in onset and cessation of wood formation was detected among species, synchronous culmination of radial growth indicates homogenous exogenous and/or endogenous control. The close coupling of radial growth to atmospheric conditions points to the importance of stem water status for intra-annual growth of drought-prone conifers.  相似文献

The wheat (Triticum aestivum L.) plant type in major producing areas of the U.S. is changing rapidly from tall cultivars to high‐yielding semidwarf cultivars. Objectives of experiments were to determine if nitrogen and phosphorus nutritional requirements differ between traditional tall cultivars and modern semidwarf cultivars under dryland and irrigated conditions. ‘Larned’, a tall cultivar; ‘Newton’, a semidwarf cultivar; and ‘Plainsman V, a high‐protein semidwarf cultivar, were grown with all combinations of three nitrogen fertilizer levels (0, 84, and 168 kg N/ha) and two phosphorus fertilizer levels (0 and 90 kg P2O5/ha) at Colby, Kansas for two years. Three levels of irrigation—dryland, limited irrigation, and full irrigation—were applied. Grain yields were highest with 84 kg N/ha under dryland and with 168 kg N/ha under irrigation. Phosphorus increased grain yield under dryland conditions one year, but had no effect under irrigated conditions. Cultivar X nutrition interactions from differential yield responses to fertility levels occurred under the dryland and limited irrigation regimes one year. Grain protein content was increased by nitrogen fertilization under all regimes both years and was decreased only by phosphorus fertilization under dryland conditions one year. Cultivar X nitrogen interactions for grain protein occurred under all irrigation regimes. We concluded that nutrient requirements do not differ between tall and semi dwarf wheat culti‐vars under any irrigation regime. Raising the recommended level of nutrients, particularly nitrogen, should be considered for all cultivars, both tall and semidwarf.  相似文献
