Positron emission tomography/computed tomography (PET/CT) utilizing 3′‐deoxy‐3′‐[18F]fluorothymidine (18FLT), a proliferation tracer, has been found to be a useful tool for characterizing neoplastic diseases and bone marrow function in humans. As PET and PET/CT imaging become increasingly available in veterinary medicine, knowledge of radiopharmaceutical biodistribution in veterinary species is needed for lesion interpretation in the clinical setting. The purpose of this study was to describe the normal biodistribution of 18FLT in adult domestic cats. Imaging of six healthy young adult castrated male cats was performed using a commercially available PET/CT scanner consisting of a 64‐slice helical CT scanner with an integrated whole‐body, high‐resolution lutetium oxy‐orthosilicate (LSO) PET scanner. Cats were sedated and injected intravenously with 108.60 ± 2.09 (mean ± SD) MBq of 18FLT (greater than 99% radiochemical purity by high‐performance liquid chromatography). Imaging was performed in sternal recumbency under general anesthesia. Static images utilizing multiple bed positions were acquired 80.83 ± 7.52 (mean ± SD) minutes post‐injection. Regions of interest were manually drawn over major parenchymal organs and selected areas of bone marrow and increased tracer uptake. Standardized uptake values were calculated. Notable areas of uptake included hematopoietic bone marrow, intestinal tract, and the urinary and hepatobiliary systems. No appreciable uptake was observed within brain, lung, myocardium, spleen, or skeletal muscle. Findings from this study can be used as baseline data for future studies of diseases in cats. 相似文献

A major problem facing ecologists is obtaining a complete picture of the highly complex soil community. While DNA-based methods are routinely used to assess prokaryote community structure and diversity in soil, approaches for measuring the total faunal community are not yet available. This is due to difficulties such as designing primers specific to a range of soil animals while excluding other eukaryotes. Instead, scientists use laborious and specialized taxonomic methods for extracting and identifying soil fauna. We examined this problem using DNA sequencing to profile soil animal diversity across two Alaskan ecosystems and compare the results with morphological analyses. Of 5267 sequences, representing 549 operational taxonomic units (OTU), only 18 OTUs were common to both sites. Representatives included 8 phyla, dominated by arthropods and nematodes. This is the most comprehensive molecular analysis of soil fauna to date, and provides a tool to rapidly assess a missing component of soil biodiversity. 相似文献

Following clearcutting applying the conventional stem-only harvesting method in a Norway spruce (Picea abies (L.) Karst.) stand and different levels of removal of logging residue, the nutrient fluxes from the heaps of logging residue and from the O horizon were monitored over four growing seasons and the soil nutrient pools were determined. Three levels of removal of logging residue were carried out using (i) conventional stem-only harvesting (no residues removed); (ii) residues removed; and (iii) removal of branches (foliage left on site). The heaps of logging residue were a minor source of inorganic N entering the soil in the water percolating through the heaps, but they were a significant source of organic N, P, Ca, Mg, and especially K. Nutrient fluxes from the O horizon were in general greater under the heaps of logging residue as compared to soils without overlying logging residue. The leaching of inorganic N from the O horizon under the heaps of logging residue resulted in a net loss of these compounds, while the O horizon without overlying logging residue gained N. The removal of logging residue significantly decreased the extractable K pools in the soil while it or conversely, the presence of residue heaps had no significant effect on the pools of organic matter and the pools of N, P, Ca, and Mg in the O horizon and in the 0–10 cm soil layer. The results show that the short-term effects of logging residue on nutrient dynamics in the soil can be complex and difficult to interpret in terms of site productivity as there are changes in the nutrient fluxes, which imply the opposite effects on site productivity. However, the results do indicate that, in the short-term, the removal of logging residue does not impair pools of N in the soil nor site productivity on sites where the availability of N limits productivity. 相似文献

Cycling of nutrients, including nitrogen and phosphorus, is one of the ecosystem services we expect agricultural soils to deliver. Nutrient cycling incorporates the reuse of agricultural, industrial and municipal organic residues that, misleadingly, are often referred to as ‘wastes’. The present review disentangles the processes underlying the cycling of nutrients to better understand which soil properties determine the performance of that function. Four processes are identified (i) the capacity to receive nutrients, (ii) the capacity to make and keep nutrients available to crops, (iii) the capacity to support the uptake of nutrients by crops and (iv) the capacity to support their successful removal in harvested crop. Soil properties matter but it is imperative that, as constituents of ‘soil quality’, they should be evaluated in the context of management options and climate and not as ends in their own right. The effect of a soil property may vary depending on the prevailing climatic and hydrologic conditions and on other soil properties. We recognize that individual soil properties may be enhancing one of the processes underlying the cycling of nutrients but simultaneously weakening others. Competing demands on soil properties are even more obvious when considering other soil functions such as primary production, purification and flow regulation of water, climate modification and habitat provision, as shown by examples. Consequently, evaluations of soil properties and management actions need to be site‐specific, taking account of local aspects of their suitability and potential challenges. 相似文献

Forty-one dogs with resistant lymphoma were treated with a modified MOPP (mechlorethamine, vincristine, procarbazine and prednisone) protocol (MPP [mechlorethamine, procarbazine and prednisone] administered on a 21-day cycle, shortened from the 28-day MOPP cycle). The overall response rate to MPP was 34% for a median of 56 days (95% confidence interval 30–238). Seventeen percent of dogs had a complete response for a median duration of 238 days, 17% had a partial response for a median of 56 days and 32% had stable disease for a median of 24 days. Histological grade or cell morphology on cytology was associated with response. Minimal toxicity was observed with the MPP protocol, suggesting that further dose intensification or addition of another chemotherapeutic agent would be possible. 相似文献

Viking 2 lander began imaging the surface of Mars at Utopia Planitia on 3 September 1976. The surface is a boulder-strewn reddish desert cut by troughs that probably form a polygonal network. A plateau can be seen to the east of the spacecraft, which for the most probable lander location is approximately the direction of a tongue of ejecta from the crater Mie. Boulders at the lander 2 site are generally more vesicular than those near lander i. Fines at both lander sites appear to be very fine-grained and to be bound in a duricrust. The pinkish color of the sky, similar to that observed at the lander I site, indicates suspension of surface material. However, the atmospheric optical depth is less than that at the lander I site. After dissipation of a cloud of dust stirred during landing, no changes other than those stemming from sampling activities have been detected in the landscape. The aim of the study was to compare model-based approaches inthe prediction of timber assortment recovery with bucking simulationbased on detailed stem data. A correction function for the totallength of saw log fragments and two optional saw log reductionmodels, that is, the MELA96 version and MELA05 version, wereapplied. In the bucking simulation, the volumes by timber assortmentswere calculated using a bucking-to-value simulator. The predictionof saw log recoveries varied between the bucking simulationand different versions of saw log reduction models. The levelof the reduction from the MELA96 version was at the same levelas from the bucking simulation where defects were taken intoaccount, but the saw log reduction had a very low variance dueto a small amount of independent variables. The saw log reductionof the MELA05 version included more variation although the levelof the reductions was higher and the variation did not meetwith the bucking simulation. As a conclusion, the model-basedapproaches seem applicable at least for the prediction of sawlog recovery in the large area forest inventories where thevariance of the standwise timber assortment recoveries neednot be predicted. 相似文献

Antibody molecules, and antibody fragments in particular, have enormous potential in the development of biosensors for marine monitoring. Conventional immobilisation approaches used in immunoassays typically yield unstable and mostly incorrectly oriented antibodies, however, resulting in reduced detection sensitivities for already low concentration analytes. The 2H12 anti-domoic acid scFv antibody fragment was engineered with cysteine-containing linkers of two different lengths, distal to the antigen binding pocket, for covalent and correctly oriented immobilisation of the scFvs on functionalised solid supports. The Escherichia coli-produced, cysteine-engineered scFvs dimerised in solution and demonstrated similar efficiencies of covalent immobilisation on maleimide-activated plates and minimal non-covalent attachment. The covalently attached scFvs exhibited negligible leaching from the support under acidic conditions that removed almost 50% of the adsorbed wildtype fragment, and IC50s for domoic acid of 270 and 297 ng/mL compared with 1126 and 1482 ng/mL, respectively, for their non-covalently adsorbed counterparts. The expression and immobilisation approach will facilitate the development of stable, reusable biosensors with increased stability and detection sensitivity for marine neurotoxins. 相似文献
