Effects of Medium-chain Triglyceride on Growth Performance and Serum Biochemical Indices in Broilers

The study was conducted to investigate the effects of medium-chain triglyceride replacing the soybean oil in the basal diet on growth performance and serum biochemical indices of broilers.One hundred and forty four healthy,similar body weights AA broilers at the age of 14 days were randomly assigned to four groups with six replicates ger group and six broilers per replicate.Groups A,B,C and D were respectively fed with 0,0.6%,1.2% and 2% medium-chain triglyceride substituted the equivalent amount of soybean oil (using corn flour to balance the energy levels in each group) .The test lasted for 28 days and the indexes were measured once every two weeks.The results showed that the average daily gain and average daily feed intake of groups B,C and D were higher than that of group A,and the F/G was lower than group A.The average daily gain of groups C and D at 28 to 42 days old and group D at 14 to 42 days old were significantly higher than that of group A (P < 0.05).The serum glucose,total content of amino acid,total protein and albumin of groups B,C and D at age of 28 and 42 days old were higher than that of group A,while the serum triglyceride,total cholesterol and urea nitrogen levels were lower.The total cholesterol content at 42 days old and triglyceride concentrations at 28 days old of group C were significantly lower than those of group A (P < 0.05).The results indicated that medium-chain triglyceride could raise average daily gain and reduce the total cholesterol content and triglyceride concentrations of broilers,which were useful for improving the growth performance.     

陇薯9号的品种特性及脱毒种薯繁育技术

1株鸭源非O1/O139群霍乱弧菌的分离鉴定与致病性分析

为探明引起贵州省某鸭场雏鸭发病的病原及其致病性和耐药情况，本研究对该鸭场送的疑似细菌感染病鸭进行剖检，取鼻黏膜、心脏和肝脏等组织器官接种于培养基中进行细菌分离鉴定，通过对分离菌进行药敏试验、动物回归试验和毒力基因检测研究其耐药情况和致病性。结果显示，分离菌在血琼脂培养基上生长16 h后呈现为边缘整齐、有光泽的乳白色菌落，伴有β-溶血现象，经革兰氏染色后在生物显微镜下呈两端钝圆、弧状、排列无规则的革兰氏阴性短小杆菌，与霍乱弧菌相符；16S rDNA基因序列同源性及系统进化树显示，该分离菌与霍乱弧菌同源性高达99.6%~99.7%聚为一支；药敏试验结果显示，分离菌对大部分药物都表现为耐药，其中对氨苄西林、克林霉素、复方新诺明、苯唑西林和克林霉素等抗菌药耐药性较强，对头孢哌酮和头孢曲松敏感；动物回归试验显示，分离菌可导致试验组雏鸭5 d内全部发病死亡，表明该分离菌对雏鸭具有较强的致病性；毒力基因PCR检测结果显示，检测的霍乱弧菌相关毒力基因hlyA、ompW和chxA为阳性，而检测的O1群rfb、O139群rfb、tcpA及ctxA基因为阴性，表明本次分离的霍乱弧菌携带有致病基因，但不属于O1和O139血清群。结果表明，该鸭场雏鸭发病的疫情病原为非O1/O139血清群霍乱弧菌，该菌致病性强且对多种抗菌药物耐药。本试验结果为贵州省鸭霍乱弧菌病的防控提供了参考依据。     

东北地区春玉米临界氮浓度稀释曲线的建立和验证

过量施氮是目前玉米栽培中存在的普遍现象,基于临界氮浓度稀释曲线计算得出的氮营养指数是诊断氮营养丰缺的重要手段。基于东北地区4个生态点的试验数据,构建了东北地区春玉米临界氮稀释曲线,并在此基础上建立了氮营养指数模型和需氮量模型,结果表明,东北地区春玉米地上部临界氮浓度与生物量符合幂函数关系。利用独立试验资料对建立的临界氮浓度稀释曲线进行检验,发现基于临界氮浓度稀释模型计算的氮营养指数可以准确诊断玉米植株的氮营养状况,并计算出实时的氮素需求量。该研究建立的东北地区春玉米临界氮稀释模型可以为该地区春玉米的氮营养诊断和动态调控提供较好的理论和技术指导。     

菊花花瓣伸长相关基因CmXTHs的克隆与功能验证

从杭白菊(Chrysanthemum morifolium‘Hangbaiju’)中克隆了4个细胞壁松弛和伸展相关的木葡聚糖内转糖苷酶/水解酶基因,分别命名为CmXTH1、CmXTH2、CmXTH3和CmXTH4,序列分析表明其编码的氨基酸序列N端都含有23~30个氨基酸组成的信号肽区域和保守的催化活性位点DE(I/L)DEFLG以及紧邻的N(R/A)T组成的N端糖基化位点,CmXTH1/2/3的C端均含有4个保守的半胱氨酸残基,CmXTH1/2/3/4蛋白的相似度为66.2%。系统进化分析结果表明,CmXTH1/2/3属于XTH家族Ⅰ组,CmXTH4属于Ⅱ组。实时荧光定量结果表明:(1)菊花CmXTH1/2/3/4在根、茎、叶和花蕾中均有表达。CmXTH1在根中表达量较高,CmXTH2/3在茎中表达量较高,CmXTH4在花蕾中表达量最高。(2)CmXTH1/2/3/4在花序不同部位表达量有差异,CmXTH1/4在舌状花中表达量最高,CmXTH2/3在筒状花中表达量较高。(3)CmXTH1/2/3/4在舌状花不同发育阶段表达量差异显著,CmXTH1/2在盛花期表达量最高,CmXTH3在衰老期表达量最高,CmXTH4在初开期表达量最高。病毒诱导基因沉默结果表明,CmXTH1/2/3/4沉默系的花序直径和舌状花长度与对照相比均有所减小,其中CmXTH4沉默系减小最大,分别减小了25.67%和10.42%,舌状花花瓣表皮细胞与对照相比明显变小,影响了舌状花花瓣的伸长;CmXTH2沉默系的筒状花雄蕊长度和CmXTH4沉默系的花萼直径分别与对照相比显著减小。这表明菊花CmXTH1/2/3/4基因参与了花序的开放,促进了舌状花花瓣的伸长,对于菊花的花序增大具有重要作用。

Effects of Jiedu-Qingfei mixture on NF-κB and p38 MAPK pathways in lung tissues of rats with Mycoplasma pneumoniae infection

AIM: To observe the therapeutic effect of Jiedu-Qingfei mixture on Mycoplasma pneumoniae (MP)-infected rat lung tissues and to explore its mechanism. METHODS: SD rats (n=40) were randomly divided into 4 groups:blank control group, model group, Jiedu-Qingfei group and positive control group, with 10 rats in each group. The rats in experimental groups were slowly dripped with 1×10⁹ CFU/L MP solution into their nostrils for 4 d. One rat in each group was sacrificed for MP nucleic acid detection at the second day after inoculation, and the other rats were given gavage therapy. The rats in blank control group and model group were intragastrically given the same volume of normal saline, the rats in Jiedu-Qingfei group were given 8 mL/kg Jiedu-Qingfei mixture daily for 4 weeks, and the rats in psoitive control group were given dexmethasone sodium phosphate (0.5 mg·kg^-1·d^-1). After the experiment, the rats were killed. The serum and bronchoalveolar lavage fluid (BALF) were collected for detecting the levels of interleukin-12 (IL-12), IL-13 and TNF-α by ELISA. The right lung tissues were used for pathological observation and HE staining, while the left lung tissues were used to detect the expression of NF-κB p50, I-κBα and p38 mitogen-activated protein kinase (p38 MAPK) at mRNA and protein levels. RESULTS: The results of MP nucleic acid detection showed that all the rats except blank control group were MP nucleic acid positive, indicating that the rat model of MP infection was successfully established. On the 1st day of the treatment, the pathological scores of the lung tissues in model group and Jiedu-Qingfei group were significantly higher than those in blank control group (P<0.05). After treatment, the pathological scores of the lung tissues in mo-del group were significantly higher than those in blank control group and Jiedu-Qingfei group. The levels of IL-12 in the serum and BALF in model group were significantly lower than those in blank control group after MP infection (P<0.05), while those after treatment with Jiedu-Qingfei mixture were significantly higher than those in model group (P<0.05). The levels of IL-13 and TNF-α in the serum and BALF of MP-infected rats were increased significantly, while those after treatment with Jiedu-Qingfei mixture were significantly lower than those in model group (P<0.05). The mRNA expression levels of NF-κB p50 and p38 MAPK in model group were increased significantly (P<0.01). After treatment, the mRNA expression levels of NF-κB p50 and p38 MAPK were decreased significantly compared with model group (P<0.01). The mRNA expression level of I-κBα in model group was significantly lower than that in control group. After treatment, the mRNA expression of I-κBα in Jiedu-Qingfei group was significantly higher than that in model group (P<0.05). The protein levels of NF-κB p50 and p38 MAPK in the lung tissues of model group were significantly higher than those of blank control group. After treatment, the protein expression of NF-κB p50 and p38 MAPK was decreased significantly. The protein level of I-κBα in model group was significantly lower than that in blank control group, and after treatment with Jiedu-Qingfei mixture, the protein expression level of I-κBα was increased significantly (P<0.05). CONCLUSION: Jiedu-Qingfei mixture may attenuate lung tissue inflammation caused by MP through NF-κB and p38 MAPK pathways.

非线性、分形与期锌市场价格

从非线性角度研究期锌市场，运用R/S分析法对期锌市场进行分析，发现期锌市场的Hurst指数为0.6183,其价格收益率序列是分维时间序列，具有分形结构，维数为1.6174。期锌市场的平均周期长度为176天，具有强持续性和相关性。关联系数为19.47%，远远大于国外相关市场。在风险方面，期锌市场的风险比期铜市场的要小，而比股票市场的风险要大。
