Genetic Diversity in AA and CC Genome Oryza Species in Southern South Asia   总被引:3,自引:0,他引:3  
The CC genome of Oryza is found in nine species of Oryza that are distributed on all continents having a tropical climate. Three diploid Oryza species with CC genome are found in Africa and Asia to Papua New Guinea. In southern South Asia these three CC genome diploid species can be found, O. eichingeri and O. rhizomatis in Sri Lanka and O. officinalis in India. AA genome wild relatives of rice are also found in the same geographic region. Germplasm of both diploid CC and AA genome Oryza germplasm has recently been collected from Sri Lanka. AFLP analysis was used to compare the genetic diversity of the two Oryza genomes from a similar geographic region in southern South Asia. In addition, the diploid CC Oryza germplasm was also analyzed by RAPD and SSR methodologies and the combined results were analyzed. The results show that in southern South Asia the diploid CC genome species have a high level of genetic diversity compared to the diploid AA genome species. Molecular marker analysis revealed that populations of O. rhizomatis from northern and southeastern Sri Lanka are genetically differentiated. One accession of O. rhizomatis was aligned with O. eichingeri. This accession was collected from the site of O. rhizomatis that is the closest to a population of O. eichingeri. O. eichingeri showed lower genetic diversity than the other two diploid Oryza CC genome species. O. officinalis accessions from Assam, India, and China were genetically less diverged from O. eichingeri and O. rhizomatis than two accessions of O. officinalis from Kerala state, India. The genetic diversity of 58 wild and weedy populations representing taxa within the V. sativa aggregate from the former USSR, 4 cultivars of V. sativa, 2 accessions of V. cordata and 3 accessions of V. macrocarpa from Mediterranean countries were analysed using randomly amplified DNA fragments (RAPDs) and seed protein electrophoresis (SDS-PAGE). Interspecific variation between taxa in the V. sativa aggregate could readily be detected using both techniques. RAPDs and seed protein patterns were found to be an effective means of identifying accessions that cannot be identified clearly by morphological criteria alone. RAPD and seed protein analysis revealed a clear relationship between observed genetic variation of populations and their geographical distribution. Populations from each region had their own gene pools. Geographical variation was detected in V. segetalis. The degree of genetic divergence between local populations was usually related to proximity. In several locations where wild and weedy populations of different V. sativa agg. taxa grow sympatrically, intermediate forms could be detected at the DNA and protein levels. Both RAPD and seed protein analysis support the view that the V. sativa aggregate consists of 8 taxa warranting recognition at the species level. Several species in this aggregate are evolving intra-specific groups which can readily be detected at the molecular level.  相似文献

Land applications of manure from confined animal systems and direct deposit by grazing animals are both major sources of bacteria in streams. An understanding of the overland transport mechanisms from land applied waste is needed to improve design of best management practices (BMPs) and modeling of nonpoint source (NPS) pollution. Plots were established on pasturelands receiving phosphorus-based livestock waste applications to measure the concentrations of Escherichia coli (E. coli), fecal coliform (FC), and Enterococcus present in overland flow at the edge of the field. The flow-weighted bacteria concentrations were highest in runoff samples from the plots treated with cowpies (1.37×105 colony forming units (cfu)/100 ml of E. coli) followed by liquid dairy manure (1.84×104 cfu/100 ml of E. coli) and turkey litter (1.29×104 cfu/100 ml of E. coli). The temporal distribution of fecal bacterial concentrations appeared to be dependent upon both the animal waste treatment and the indicator species, with peak concentrations occurring either at the beginning of the runoff event or during peak flow rates. BMPs could be selected to reduce peak flows or first flush effects depending upon the litter or manure applied to the land. The commercial Biolog System was used to identify the dominant species of Enterococcus present in the cowpie source manure (Enterococcus mundtii 55%) and in the runoff collected from the transport plots treated with cowpies (Enterococcus faecalis 37%). The identification of predominant species of Enterococcus that are associated with specific sources of fecal pollution could greatly assist with identifying the origins of NPS pollution.  相似文献

The capacity of extracts from toxigenic and non-toxigenic ruminant strains of Fusobacterium necrophorum to protect against challenge with homologous and heterologous bacteria was examined in mice. The numbers of F. necrophorum which were infective or lethal for mice increased 5- to 8-fold in animals which had been previously inoculated with complete Freund's adjuvant (FCA). Although preparations containing lipopolysaccharide (LPS) and outer membrane proteins (OMP) from several strains gave protection against a non-toxigenic strain (FnB-3), they did not significantly immunize mice against a challenge infection with a toxigenic bovine strain, FnB-1. Only material which had been prepared by gel filtration of 18-h liquid culture supernates of toxigenic F. necrophorum elicited significant immunity against homologous challenge with FnB-1. This preparation contained LPS and the majority of the leucotoxic activity. However, passive protection was not afforded to mice inoculated with bovine or rabbit sera which possessed high neutralization titres against the leucocidin.  相似文献

A search was made for Common Seals (Phoca vitulina) around Shetland in September 1971. The coast was arbitrarily divided into eight areas and the count of Common Seals in each was adjusted to give an estimated total. 1,219 Common Seals were counted and a grand total of 1,800 estimated for the whole of Shetland. The question of whether or not the Shetland Common Seal stock is declining is discussed. It is concluded, on the basis of quantitative evidence and from discussions with local boatmen and seal hunters, that the stock is declining, although the rate of decline is unknown.  相似文献

Summary Bisulfite modification of DNA from potato leaves showing juvenile and mature leaf morphologies and from leaves of autotrophic and heterotrophic microplants showing different morphologies was carried out to deaminate cytosine residues while leaving 5-methylcytosine residues intact. RAPD analysis of the DNA was then performed using selective primers; firstly, with a 50–70% GC content and subsequently with a 50–70% AT content which contained CG or CA 3' ends. Three of the 4 primers used detected band polymorphisms in the juvenile versus mature leaves and all four primers detected band polymorphisms in the autotrophic·versus heterotrophic in vitro leaf samples. These results indicate differences in methylation in the different paired DNA samples. The hypothesised increase in band numbers in the relatively less mature samples was confirmed for both the in vivo and in vitro leaf samples; a higher number of bands was amplified from the DNA of the leaves from heterotrophic cultures compared with those from autotrophic culture when the results for all primer sets were grouped. however, this trend did not apply to the results from all the individual primers. The application of this approach to the evaluation of in vitro protocols for potato micropropagation is discussed.  相似文献

Data from field experiments were used to test whether disease observations on lower leaves of wheat were good predictors of future epidemic development in the upper canopy. Fungicide treatments to replicated plots in 1994 and 1995 caused variation in levels of initial inoculum of urediniospores of yellow rust ( Puccinia striiformis ). Observations of symptom severity were made on individual leaf layers throughout the season. Sporulating lesions on lower leaves were considered to be a measure of inoculum source strength for transfer to upper culm leaves. Low source strengths were associated with delays in epidemic development on the upper leaves, as quantified by a location parameter, t m, the time at which severity reached half of the asymptote value in logistic fits to disease progress data on the upper leaves. However, there was much unexplained variation, probably due to extraneous variation in upward transfer efficiency of inoculum and rates of epidemic development. Vanderplank's sanitation ratio theory was used to account for variation in inoculum transfer and epidemic rate. The analysis revealed that if the aim is to predict disease on a newly emerged culm leaf, during the period when it can be treated effectively with fungicide, then observations of disease two leaves further down the culm were of the greatest predictive value ( R 2 = 86%). These observations, however, need to be integrated with information about factors affecting upward inoculum transfer and rates of epidemic development if acceptable predictive precision is to be achieved.  相似文献

The toxicity of three second-generation rodenticides to Barn Owls (Tyto alba Scop.) has been investigated. Brodifacoum, difenacoum and flocoumafen were separately fed to owls over a period of 15 days via rodenticide-fed mice to simulate the potential route of exposure in the wild. The owls survived a cumulative dose of each rodenticide of at least 1.9 mg kg?1 owl body weight over 15 days. This is equivalent to the consumption of two 25 g mice with a rodenticide residue of 1 mg kg?1 each day for 15 days. Residue analysis confirmed that the liver is the organ which retains the largest residue of ingested rodenticide.  相似文献

OBJECTIVE: To assess radiologically the caudal vertebrae of a male alpaca with an abnormal tail, and nine of his offspring. PROCEDURE: Right lateral and ventrodorsal radiographs were taken of the caudal vertebrae. RESULTS: The caudal vertebral abnormalities in these alpacas included vertebral subluxation, hemivertebrae, wedge hemivertebrae and block vertebrae. CONCLUSION: These malformations in offspring of one sire suggest a hereditary basis for congenital caudal vertebral malformations in the alpaca.  相似文献
