A new cleanup procedure is described for chlorinated hydrocarbon residues in butterfat. The method is based on the dropwise addition of H2SO4 to a fat solution column and continuous removal of the lipids and the acid. The cleanup of 0.25-2.0 g fat requires only 10-40 ml sulfuric acid and 12-17 ml petroleum ether. There is no need for any further cleanup step, solvent evaporation, or centrifugation. The method is easy to standardize and is suitable for automation. At least 30 fat samples can be cleaned up manually by one analyst in one day. Recoveries were complete (greater than 90%) for polychlorinated biphenyl compounds and for 13 chlorinated pesticides of 16 examined. The method was tested on chlorinated hydrocarbon residues in commercial butter and the results were compared with those obtained with the acetonitrile method. The versatility and limitations of the method were investigated by varying the sulfuric acid strength, initial fat solution concentration, and column dimensions.  相似文献

A laboratory technique is described that was developed to investigate the effects of hoof pressure on grassland topsoil. The effects of a vertically-applied pressure (300 kPa) of short duration on 4 soils equilibrated at 0, 5 and 10 kPa water tension are reported. Less vertical deformation and compression were produced at the lower water tension in 2 of the soils. In general, there was a greater recovery from deformation, measured immediately after the transient pressure, at lower water tensions. The most severe structural damage was recorded at depths of approximately 100 mm rather than at the surface.  相似文献

Methods to aid in the large-scale testing and characterization of Coastal Plain soils based on their susceptibility to root-limiting strength problems were developed and analyzed. They were basically regression equations modeled after a Taylor series expansion. The equations relate changes of soil strength, bulk density and soil water content between field and “critical rooting conditions”. Once equations wered eveloped from a data set of 426 laboratory samples, critical rooting bulk density was predicted for a separate set of laboratory and field samples. All laboratory samples and appropriate field samples were equilibrated at — 100 kPa soil-water potential. Soils used were sandy Ultisols, which may limit the scope of equations.

In many cases, changes in the water contents were not a significant factor in the prediction of soil strength. This may be a reflection of the limited capabilities of the equations, the uniform equilibration of soil-water potential of the soils, or the fact that the slope of the strength vs. bulk density curve is independent of water content over the range of samples considered. Nevertheless, it does simplify the equations and may suggest that a series of several equations for different soil types would be better than a single equation that requires soil-water content.  相似文献

The proteins of wheat flour have several biological activities that can affect human health and physiology when wheat-based foods are consumed. The modifications of bread crumb and crust proteins during an in vitro peptic/pancreatic digestion process were studied by electrophoresis and immunoblotting with polyclonal antibodies specific for single proteins or groups of homologous proteins of the wheat flour, and the results were compared to those obtained for an unheated dough sample. The results show that baking affects the extent of proteolysis and the immunological and physicochemical features of the digestion products in relation to the level of the heat treatment. Therefore, the results concerning the digestion of the unheated wheat flour or dough are not representative of what happens when baked products enter the human digestive tract.  相似文献

The oyster mushroom (Pleurotus ostreatus) is widely cultivated on wheat straw (Triticum aestivum); however, there is a need to better understand the relationship between the chemical composition of the compost and mushroom growth. Wheat straw was degraded over a period of 63 days by P. ostreatus during which time it was sampled at weekly intervals. Off-line thermochemolysis with tetramethylammonium hydroxide and solid-state (13)C NMR were then used in the molecular characterization of the undegraded wheat straw and the degraded samples. The degraded wheat straw samples had a lower proportion of syringyl- to guaiacyl-derived moieties and cinnamyl- to guaiacyl-derived moieties than the undegraded control. There were increases in both guaiacyl and syringyl acid to aldehyde ratios with composting time, which showed that side-chain oxidation has been mediated by P. ostreatus. The (13)C NMR spectra confirmed the increase in carboxyl content but indicated that the overall lignin and methoxyl contents remained relatively constant, although some nonsystematic variations were observed. The spectra also showed a decrease in amorphous noncellulosic polysaccharides in relation to the crystalline cellulose upon degradation.  相似文献

Kinetic studies are of great concern for understanding the processes and parameters involved in the sorption of pollutants by soils. Sorption kinetics of imidacloprid and diuron in eight soils of different characteristics, with very low organic carbon content were investigated. Pseudosecond-order kinetic reactions closely correlate with the experimental kinetic (R(2) > 0.98) in all soils. The sorbed amount of diuron was higher than that for imidacloprid. The low OC content of these soils correlated neither with the sorbed amount nor with the kinetic parameters for both pesticides. Imidacloprid sorption was correlated with silt and sand content and cation exchange capacity (CEC); meanwhile for diuron, no correlation was found. Thus, sorption kinetics take place throughout different mechanisms related mainly to the chemical character of the pesticides. Sorption kinetic parameters determined using three of the four models selected (pseudosecond-order kinetic reactions, Elovich equation, and Weber-Morris models) have been shown to be worthy to distinguish the process controlling the sorption kinetic of both pesticides.  相似文献

Male raccoons and red foxes were trapped alive and exposed to each other in captivity. Animals of each species trapped close to one another demonstrated a higher frequency of initial dominance-subordinance relationships and lower frequencies of more intense aggressive interactions than did animals trapped at greater distances from each other. This suggests the existence of neighbor recognition and thus a rudimentary social structure within these free-living "solitary" species.  相似文献

The current interest in locating new or alternative sources of energy has focused attention on solar energy capture by crops that can be subsequently utilized as a substitute for fossil fuels. The very high productivity of sugarepane and the fact that it accumulates sugars that are directly fermentable to alcohol may have caused seemingly less productive crops to be overlooked. We show here that recoverable alcohol from achievable commercial yields of pineapple can actually equal that of sugarcane, with the pineapple crop requiring only a fraction of the water used by sugarcane. Pineapple is well adapted to the subhumid or semiarid tropics and thus is particularly well suited for exploiting large areas not now under cultivation with any crop of commercial value.  相似文献

Tumor induction in athymic nude mice can be used to detect dominant transforming genes in cellular DNA. Mouse NIH 3T3 cells freshly transfected with either cloned Moloney sarcoma proviral DNA or cellular DNA's derived from virally transformed cells induced tumors when injected into athymic nu/nu mice. Tumors were also induced by cells transfected with DNA from two tumor-derived and one chemically transformed human cell lines. The mouse tumors induced by human cell line DNA's contained human DNA sequences, and DNA derived from these tumors was capable of inducing both tumors and foci on subsequent transfection. Tumor induction in nude mice represents a useful new method for the detection and selection of cells transformed by cellular oncogenes.  相似文献
