OBJECTIVE: To determine whether iontophoretic administration of dexamethasone to horses results in detectable concentrations in synovial fluid, plasma, and urine. ANIMALS: 6 adult mares. PROCEDURE: Iontophoresis was used to administer dexamethasone. Treatments (4 mA for 20 minutes) were administered to a tarsocrural joint of each mare. The drug electrode contained 3 ml of dexamethasone sodium phosphate at a concentration of 4 or 10 mg/ml. Samples of synovial fluid, blood, and urine were obtained before and 0.5, 4, 8, and 24 hours after each treatment. All samples were tested for dexamethasone using an ELISA. Synovial fluid also was evaluated for dexamethasone, using high-performance liquid chromatography. RESULTS: The lower and upper limits of detection for dexamethasone in synovial fluid with the ELISA were 0.21 and 1.5 ng/ml, respectively. Dexamethasone administered at a concentration of 10 mg/ml was detected by the ELISA in synovial fluid of 5 mares from 0.5 to 24 hours and in urine of 4 mares from 0.5 to 8 hours after each treatment, but it was not detected in plasma. Mean synovial fluid concentration of dexamethasone was 1.01 ng/ml. Dexamethasone administered at a concentration of 4 mg/ml was detected by the ELISA in urine of 2 mares at 0.5 and 4 hours after treatment, but it was not detected in synovial fluid or plasma. CONCLUSIONS AND CLINICAL RELEVANCE: Iontophoresis cannot be considered an effective method for delivery of dexamethasone to synovial fluid of horses, because drug concentrations achieved in this study were less than therapeutic concentrations.  相似文献

Circulating concentrations of estradiol (E2), vitellogenin (VTG), thyroxine (T4), triiodothyronine (T3) and insulin were measured in reproductively maturing four and five year-old Atlantic salmon. Blood samples were collected from the fish in seawater for one year prior to their spawning in November in fresh water. In females, E2 and VTG were low but detectable from December to July, and then increased to peak levels in September and October. Plasma levels of T4 and T3 were relatively constant in winter and spring, and decreased in July. Plasma concentration of T4 increased in November when the fish returned to fresh water. Plasma T3 levels remained low during the autumn. Both T4 and T3 levels tended to be higher in males than in females during September through November. Plasma insulin concentrations increased during the spring to peak values in May, and then decreased in June and July in fish of both sexes. There was a significant elevation of plasma insulin in males during October, and the levels in males tended to be higher than those found in females during final maturation.  相似文献

Ecology of Blue-Green Algae in Aquaculture Ponds   总被引:5,自引:0,他引:5  
Cyanobacteria (blue-green algae) in the genera Anabaena, Aphanizomenon, Microcystis, and Oscillatoria often form extensive and persistent blooms in freshwater aquaculture ponds. Bloom-forming cyanobacteria are undesirable in aquaculture ponds because: 1) they are a relatively poor base for aquatic food chains; 2) they are poor oxygenators of the water and have undesirable growth habits; 3) some species produce odorous metabolites that impart undesirable flavors to the cultured animal; and 4) some species may produce compounds that are toxic to aquatic animals. Development of cyanobacterial blooms is favored under conditions of high nutrient loading rates (particularly if the availability of nitrogen is limited relative to phosphorus), low rates of vertical mixing, and warm water temperatures. Under those conditions, dominance of phytoplankton communities by cyanobacteria is the result of certain unique physiological attributes (in particular, N2 fixation and buoyancy regulation) that allow cyanobacteria to compete effectively with other phytoplankton. The ability to fix N2 provides a competitive advantage under severe nitrogen limitation because it allows certain cyanobacterial species to make use of a source of nitrogen unavailable to other phytoplankton. The ability to regulate cell buoyancy through environmentally-controlled collapse ad reformation of intracellular gas vacuoles is perhaps the primary reason for the frequent dominance of aquaculture pond phytoplankton communities by cyanobacteria. Cyanobacteria that can regulate their position in the water column gain a distinct advantage over other phototrophs in poorly mixed bodies of water. In addition to the physicochemical interactions that influence phytoplankton community dynamics, cyanobacterial-microbial associations may play an important regulatory role in determining community structure. Cyanobacteria are always found in close association with a diverse array of microorganisms, including eubacteria, fungi, and protozoans. These associations, which in the past have often been viewed as antagonistic, are increasingly seen as mutualistic and may function in a positive manner during bloom development.  相似文献

Echinococcus granulosus is an important zoonotic infection of dogs. The purpose of the present study assessed the performance of two laboratory diagnostic methods with arecoline purgation and necropsy in infected dogs. In total 65 dogs were successfully experimentally infected with protoscoleces of E. granulosus from ovine infection. At 14-34 days post-infection groups of dogs were purged with arecoline hydrobromide and then necropsied. Faecal samples were tested at weekly intervals by coproantigen ELISA and coproPCR. The necropsy infection rate with E. granulosus was 89.2 per cent. Only 43 per cent of dogs were successfully purged after one arecoline dose; this percentage increased to 76.9% for two doses of arecoline purgation. E. granulosus coproantigen was detected by coproELISA in 82.8% of faeces. The positive and negative predictive values for coproantigen ELISA were 96 and 44.4% respectively. E. granulosus DNA was detected in pre-patent faecal samples by coproPCR in 25.9% of dogs. These results indicate that coproELISA is more sensitive than arecoline purgation for the detection of pre-patent E. granulosus infection in dogs. CoproPCR detected E. granulosus DNA in dog faeces by 21 days post-infection before egg production.  相似文献

The present study was conducted to determine the safe and toxic levels of dietary copper in juvenile olive flounder, Paralichthys olivaceus, fed Mintrex® copper, a chelated dietary copper source. Fish averaging 3.8 ± 0.13 g (mean ± SD) were fed 1 of 10 diets (n = 3) containing 7 (Cu0), 10.4 (CuM5), 15.8 (CuM10), 24.9 (CuM20), 43.4 (CuM40), 82.1 (CuM80), 158 (CuM160), 308 (CuM320), 658 (CuM640), and 1267 (CuM1280) mg Cu/kg diet. At the end of 12 wk of feeding trial, weight gain (WG), specific growth rate, and protein efficiency ratio of fish fed CuM5 and CuM10 diets were significantly higher than those fed CuM80, CuM160, CuM320, CuM640, and CuM1280 diets (P < 0.05). Survival of fish fed Cu0, CuM5, CuM10, CuM20, and CuM40 diets was significantly higher than those of fish fed CuM320, CuM640, and CuM1280 diets. Whole‐body lipid content of fish decreased while whole‐body ash increased with dietary copper levels. Whole‐body and tissue copper concentrations increased with dietary copper levels. Although ANOVA test suggested that the toxic level of dietary Cu in juvenile olive flounder, P. olivaceus, could be 320 mg/kg diet, broken‐line analysis of WG indicated a level of 286 mg/kg diet when Mintrex®Cu is used as the dietary copper source.  相似文献

Abstract – Understanding movement patterns and habitat utilisation is critical for the management of diadromous fishes. An acoustic telemetry array was used to monitor 33 estuary perch, Macquaria colonorum and 39 Australian bass, Macquaria novemaculeata in the freshwater and estuarine reaches on the Shoalhaven River, south‐east Australia. On average, tagged M. novemaculeata were detected for a considerably shorter period than M. colonorum, and evidence suggested that fishing pressure may have impacted on their survival. Macquaria colonorum displayed significant shifts in seasonal and size‐related habitat use, with fish predominantly residing in deep (>5 m) areas within the middle (mesohaline) reaches of the estuary during the austral spring to autumn months. In winter, M. colonorum individuals made frequent downstream migrations, often to localised areas, within the lower estuary (LE). In contrast, M. novemaculeata were distributed in shallow (<2 m) habitats throughout the year, within the upper (oligohaline) estuarine reaches of the river, as well as in fresh water. Like M. colonorum, M. novemaculeata made extensive downstream and upstream movements, often coincident with reproductive behaviour, water temperature and increased freshwater inflows. It is postulated that the high site fidelity and repetitive homing displayed by both species is influenced by ontogenetic behaviour and prey availability. Furthermore, the extent of instream distribution by both species, and the lack of observed annual spawning migrations by some M. novemaculeata individuals, indicates the once considered ‘catadromous’ life cycle of these fishes may not be obligatory. A management approach is recommended to ensure that both these species are not over‐exploited within a portion of their instream range, thus maintaining their full reproductive potential.  相似文献

Abstract – The role of male colouration in the behavioural interactions of haplochromine cichlids has received much attention. Previous studies supported the role played by visual signals during interactions of rock‐dwelling cichlids, or mbuna, from Lake Malawi. It has largely been assumed that hue, and not pattern, is the primary signal encoded in male colouration; thus, the role of colour pattern has received less attention. We explored the use of visual signals during aggression in Labeotropheus fuelleborni by using identically and differently coloured opponents and by offering these opponents under coloured illumination. We presented two simultaneous opponents to males and found that the focal male’s responses depended upon opponent colour pattern; similarly patterned opponents received more aggression, even if presented under a hue‐obscuring light source. These results support findings that gross colour pattern may be more important than hue in interactions among the mbuna and further support an important role for visual communication in these fishes.  相似文献

Uterus masculinus (persistent Mullerian duct) is a vestigial embryological remnant of the paramesonephric duct system in males and has been associated with clinical signs such as dysuria, incontinence, tenesmus and urethral obstruction in dogs. The radiological appearance of cystic uterus masculinus in dogs has been described previously with the aid of retrograde positive or negative contrast cystography. The purpose of this retrospective study was to describe ultrasonographic features of confirmed or presumed uterus masculinus in a group of dogs with confirmed or presumed disease. Ultrasonographic findings were recorded based on a consensus opinion of two readers. A uterus masculinus was defined as cylindrical when no lumen was observed and tubular when it had lumen that was filled with anechoic fluid. Six dogs met the inclusion criterion with a mean age of 8 years and 9 months. Uterus masculinus appeared as single (four dogs) or two (two dogs) horn‐like, tubular (four dogs) or cylindrical (two dogs) structures, originating from the craniodorsal aspect of the prostate gland and extending cranially. The walls of the uterus masculinus were isoechoic to the urinary bladder wall. The diameter of the observed uterus masculinus varied from 0.3 cm to 1 cm. The length of the uterus masculinus varied from 2 cm to 6.5 cm but the cranial terminal end was not identified in two dogs. Concomitant prostatomegaly was seen in five dogs (83.3%) and urinary tract infection was noted in three dogs (50%). Findings indicated that uterus masculinus should be included as a differential diagnosis for male dogs with these ultrasonographic characteristics.  相似文献

AIM: To determine the amount of ergovaline and lysergic acid retained or excreted by geldings fed endophyte-infected seed containing known concentrations of these alkaloids, and the effects of exposure time on clinical expression of toxicosis. METHODS: Mature geldings (n=10) received diets containing either endophyte-free (E-) or endophyte-infected (E+) tall fescue seed during three experimental phases. The first phase (Days -14 to -1) was an adaptation phase, to allow all horses to adapt to a diet containing E- tall fescue seed. The second (Days 0 to 3) was the initial exposure phase to E+ tall fescue seed, used for the delivery of ergovaline and lysergic acid at 0.5 and 0.3 mg/kg of diet, respectively, to test the initial effects of exposure on routes and amounts of elimination of alkaloid. During this phase, half the geldings were exposed to an E+ diet while the rest served as controls by remaining on the E- diet. Once assigned to treatments, geldings remained on the same diet through the third phase (Days 4 to 21), which served as the extended exposure phase. Total outputs of faeces and urine were collected within each phase, to determine retention of ergovaline and lysergic acid and nutrient digestibility. Serum was collected weekly and analysed for activities of enzymes and concentrations of prolactin. Bodyweights (BW) and rectal temperatures were recorded weekly. RESULTS: BW, rectal temperature, enzyme activities and concentrations of prolactin in serum, and nutrient digestibility were not affected by treatment. Total intake of ergovaline by geldings on the E+ diet was 3.5 and 3.6 (SE 0.20) mg/day, and 2.1 and 2.3 (SE 0.11) mg/day were not accounted for in initial and extended phases, respectively. Lysergic acid was excreted in the urine (4.0 and 4.9 (SE 0.97) mg/day) and faeces (2.5 and 2.7 (SE 0.35) mg/day) at greater amounts than that consumed (2.0 and 1.9 (SE 0.09) mg/day) during the initial and extended exposure phases, respectively. Animals exposed to E+ seed for a period of 20 days appeared to excrete more (1.5 vs 1.2 mg/day; SE 0.08; p=0.03) ergovaline in the faeces than those exposed for only 4 days. CONCLUSIONS: Exposure time to the ergot alkaloids had a limited effect on the route of elimination or the amounts of ergovaline or lysergic acid excreted by horses. The primary alkaloid excreted was lysergic acid, and urine was the major route of elimination. These data will aid future research to improve animals' tolerance to toxic endophyte-infected tall fescue.  相似文献
