The present study was conducted to study the impact of the virulence factors invC and sseD of the two type III secretion systems of Salmonella enterica serovar Typhimurium (S. Typhimurium) on the pathogenesis of the porcine S. Typhimurium DT104 infection. For this purpose, two S. Typhimurium mutant strains with a disrupted invC gene of the Salmonella pathogenicity island 1 or with a disrupted sseD gene of the Salmonella pathogenicity island 2 have been studied in experimental infection of pigs. Twenty-two 7-week-old male hybrid pigs were either infected with one of the mutants or the wild-type S. Typhimurium DT104 strain. Each group was examined for clinical signs, Salmonella shedding rate and the specific antibody response. Survival and replication were evaluated by qualitative and quantitative determination of the colonization rate. The humoral and cellular immune responses were examined using isotype-specific ELISA and quantitative real-time PCR of IL-2, IL-4, IL-10, IL-12 and IFN-gamma. The results proved that both mutants had a lower virulence (with marked differences between both mutants) than the wild-type and that both virulence factors have importance in porcine salmonellosis. Only pigs infected with the wild-type S. Typhimurium DT104 exhibited typical clinical symptoms of salmonellosis like anorexia, vomiting, disturbed demeanour, fever and diarrhoea. Deletion of the invC gene resulted in a significantly reduced colonization rate. Interestingly, the mRNA expression of both type-1 and type-2 cytokines were significantly decreased in pigs infected with either the invC-mutant and the sseD-mutant strain.  相似文献

Chromosome profiles of 10 Trypanosoma (T.) congolense populations with different isometamidium sensitivities were compared using the pulsed field gel electrophoresis technique. The aim was to elucidate whether there was a karyotype pattern specific to eight isometamidium resistant phenotypes. Analysis of the profiles indicated that all populations displayed several discrete bands at the region of small, intermediate and large chromosomes. The highest similarity was observed between two isolates originating from Burkina Faso, indicating that they had the same genetic origin. Other eight strains exhibited different patterns in terms of chromosome size and numbers such that there was no characteristic karyotype pattern that was established specifically to identify resistant populations and discriminate them from the sensitive ones. This study has revealed that isometamidium resistance is not correlated to karyotype profile in T. congolense.  相似文献

Enhanced blood glucose levels are a hallmark of diabetes and are associated with diabetic complications and a reduction of lifespan. In order to search for plant extracts that display preventive activities in such a scenario, we tested 16 extracts used in human nutrition for their survival enhancing activities in the nematode Caenorhabditis elegans. Nematodes were exposed for 48 h to 10 mM glucose in the absence or presence of 0.1 % extract. Thereafter, survival was measured at 37 °C. Extracts made from coffee, kola, rooibos and cinnamon, did not influence the glucose-induced reduction of survival. Those made from ginseng, camomile, lime blossom, paraguay tea, balm, rhodiola, black tea, or knotgrass all extended the lifespan of the glucose-treated nematodes significantly but did not rescue survival completely. Extracts from the leaves of blackberries, from hibiscus, elderberries, or jiaogulan completely countered the glucose-induced survival reduction. A potent activation of the proteasome was shown for the most preventive extracts suggesting a more efficient degradation of proteins impaired by glucose. In conclusion, we present a simple animal model to screen for plant extracts with potency to reverse glucose toxicity. Extracts from blackberry leaves, hibiscus, elderberries, and jiaogulan were identified as very potent in this regard whose exact mechanisms of action appear worthwile to investigate at the molecular level.  相似文献

Here we describe the use of Rutherford backscattering spectrometry (RBS) to measure quantitative in situ elemental profiles with high depth resolution, online and nondestructively, in volatile substances (liquid and frozen acids, ice). Samples for analysis are held in a chamber with controlled temperature and partial pressures designed to match conditions for aerosols in Earth's atmosphere. This technique is demonstrated in studies of water solubility in sulfuric acid, hydrochloric acid (HCl) on ice surfaces, the formation of a HCl-hexahydrate surface layer on evaporating HCl-doped ice, and the diffusion of water through this layer.  相似文献

While remediation and storage of contaminated dredged materials is a key issue at harbour sites, there is another type of sediment pollution problem, which mainly originates from large-scale dispersion of contaminants in flood-plains, dike foreshores and polder areas. In recent years, catastrophic cases of sediment contaminations have occurred in connection with the failure of tailing dams from mines. Unlike problems related to conventional polluted sites, the risks here are primarily connected with the transporting and depositing of contaminated solids in a catchment area, especially in downstream regions. A special example demonstrating the dispersion of highly contaminated sediments in a large catchment area will be shown from the so-called Chemistry Triangle of the upper Elbe River system, Germany. The Spittelwasser area, situated there, was chosen by the organisers of the international conference ConSoil 2000 for a case comparison and four expert teams from Denmark, Germany, the Netherlands and the UK were invited to participate in this Case Study. Evaluation of the plan was done by members of the networks of NICOLE (Network for Industrially Contaminated Land) and CLARINET (Contaminated Land Rehabilition Network). In the study of the German team, five major groups of technical measures have been identified by the environmental authorities to be discussed in relation to the Spittelwasser case or for similar problem solutions in contaminated flood-plain areas. The team came to the conclusion that none of these techniques would be applicable as an individual measure. Instead, a stepwise approach combining different monitoring techniques and remediation measures was proposed. These would include point excavations of critical material, promotion of plant growth as an element for stabilising the soil and flood sediments, as well as the installation of sediment traps. At the Spittelwasser site, investigations are planned on the effects of natural attenuation processes of organic and inorganic contaminants in flood-plain sediments and soils. In the practice of this concept, non-destructive, ‘intrinsic’ bonding mechanisms and their temporal development have thus far found much less recognition compared to destructive processes such as biological degradation. Yet these so-called ‘diagenetic’ effects, which apart from chemical processes involve an enhanced mechanical consolidation of soil and sediment components by compaction, loss of water, and mineral precipitations in the pore space, may induce a quite essential reduction of the reactivity of solid matrices [see Part I ‘Improving Chemical and Biological Criteria’ (JSS - Journal of Soils and Sediments, Vol 1, No 1, pp 30-36)]. Abstract

The aim of the study was to analyse aspects of fen soil quality for grassland use with regard to the different topsoil structure and their status of earthification/moorshification (degradation). Fifty fens of different origin, structural status and land use intensity were sampled, analysed and scored by different methods: Visual Soil Assessment, Peerlkamp test and Muencheberg Soil Quality Rating. Suitable soil structure scores were found at different land use intensities with the exception of stock tracks on pastures. These had lower water and air permeability and lower soil strength. Highest visual scores of macrostructure were found where the water table was deeper; while highest overall soil quality scores occurred where the water table was optimum. The accelerated status of moorshification had no effect on the soil quality scores and on the crop yield. At lower levels of soil development (earthification) the crop yield was slightly lower due to higher proportions of inedible plants. It may be concluded that degraded peat soils will have no loss of soil quality and have relatively high soil quality for grassland use if the water table can be managed in a suitable range and the sward quality is maintained.  相似文献

Knowledge of the soil hydraulic functions is required for various hydrological studies and for the simulation of water and solute fluxes in unsaturated soils. Sand/kaolin boxes are frequently used to determine these properties in the low‐tension range. For higher tensions the pressure plate extractor is mainly applied. The extended evaporation method allows a more efficient determination of the water‐retention curve in an adequate range. Besides this method enables to quantify simultaneously the unsaturated hydraulic‐conductivity function. The objective of this study was to compare the water‐retention curves obtained from the standard methods (STM) with those determined with the extended evaporation method (EEM). A set of 90 natural soil samples of different texture and origin was analyzed, and the agreement between the methods was statistically evaluated. The average water‐content deviation (AWCD) of all samples was 1.83 vol.%, and the root mean square error (RMSE) 2.08 vol.%. The deviation of soil water‐storage capacity in the pore‐size classes 0–6, 6–30, 30–500, 500–1500 kPa varied between minimum –0.17 and 0.25 vol.% and maximum –2.89 and 2.36 vol.%, and confirmed the good comparability among the adopted methods. Systematic deviations between the methods were not found.  相似文献

The rhizosphere of two flooding‐resistant plant species (Arundinella anomala Steud., Alternanthera philoxeroides Mart.) from Three Gorges Reservoir area (China) has been examined for reactions to waterlogging and submergence. Rhizosphere parameters were monitored in natural sediment substrate by means of a dual‐access floodable rhizobox, which allows monitoring of oxygen and pH dynamics noninvasively with planar optodes in high temporal and spatial resolution, as well as simultaneous low‐invasive soil‐solution sampling. Analysis of samples for low‐molecular‐weight organic acids (LMWOA) was done by capillary electrophoresis. Roots could be observed easily in situ during growth and exposure to flooding. The floodable rhizobox is therefore considered a valuable tool for root‐reaction monitoring also under flooding conditions. During waterlogging, both species exuded oxygen into their rhizosphere and showed diurnal rhythms of rhizospheric acidification. The pH of the rhizosphere of growing root tips decreased up to 0.8 units corresponding to higher LMWOA concentrations. These rhythms weakened during flooding, but gained maximum amplitude again rapidly after resurfacing. We conclude that the root system was still fully functioning during and after flooding, and that flooding poses no threat to the physiology of the root system of the study species.  相似文献
