Te Runanga o Te Rarawa is the tribal council representing the interests of the marae (tribal commons) and hapu (a subtribal kin group) that make up the iwi (a Maori tribe) of Te Rarawa in the far north of Aotearoa/New Zealand. In April 2005, officials approached us to help them secure a valuable funding stream tagged to marshalling resources for material development in the area. They sought curriculum vitae and assistance in reframing the funding specifications. Intrigued, armed with a conceptual toolkit drawn from Gibson‐Graham's ideas of post‐development and asset‐based community mapping, and confident that we could add value, we agreed to help. This paper examines the complex politics of our involvement and our changing positioning as researcher subjects. We argue that negotiating a politics of knowledge for projects of this nature requires engagement in complex representational politics of place and divisive identity politics that rage around it. There are no easy protocols for outside researchers, but with appropriate humility and sensitivity to these politics, we can rely on, and should stand up for, the value of our work, which lies in commitments to excellence in scholarship. We cannot and should not seek to control these politics, which will chew us up and spit us out – humanely and with good grace or otherwise. However, good academic work will recognise and adapt to them. In our particular case, we argue that our work had significant value; and in this paper, we trace the production of this value. 相似文献

The intestinal microflora, typically equated with bacteria, influences diseases such as obesity and inflammatory bowel disease. Here, we show that the mammalian gut contains a rich fungal community that interacts with the immune system through the innate immune receptor Dectin-1. Mice lacking Dectin-1 exhibited increased susceptibility to chemically induced colitis, which was the result of altered responses to indigenous fungi. In humans, we identified a polymorphism in the gene for Dectin-1 (CLEC7A) that is strongly linked to a severe form of ulcerative colitis. Together, our findings reveal a eukaryotic fungal community in the gut (the "mycobiome") that coexists with bacteria and substantially expands the repertoire of organisms interacting with the intestinal immune system to influence health and disease. 相似文献

Seventy-five bovine radiographic examinations with definite periosteal response were analyzed for quality and pattern of new bone formation. Three patterns were recognized — amorphous, lamellar, and spiculated; they appeared singly or in various combinations. The lamellar and amorphous types seemed to be generally bone-related: with lamellar predominantly in the metacarpal and metatarsal bones and amorphous in the phalanges. The spiculated pattern in the bovine does not seem to be related to neoplastic processes. Bovine periosteum responded rapidly and with high magnitude. 相似文献

The presence of the zona pellucida has been perceived as a requirement for the oviductal transfer of cloned embryos at early stages of development while protecting the embryo from an immune system response. We hypothesized that steroid hormone therapy could reduce a potential cellular immune response after the transfer of zona‐free cloned embryos into the oviduct of recipient female goats. In Experiment 1, seven does were used to study the systemic immunosuppressant effect of the methylprednisolone administration (for 3 days) on blood cell counts. Whole blood was collected prior to treatment with methyprednisolone and then on Days 1, 2, 3, 4, 7, 14, 21 and 28 after the first dose of methylprednisolone for the analysis of haematological parameters. Methylprednisolone treatment significantly reduced circulating white blood cells and neutrophils in comparison with pre‐treatment levels, demonstrating a systemic immunosuppressant effect. In Experiment 2, a group of 58 does were used as recipient females to study the effect of administration of methylprednisolone for 3 days on the establishment of pregnancies after the transfer of zona‐free cloned embryos into the oviducts. No effects on pregnancy rates on Day 30 were observed regarding the distinct treatment groups (control vs. methylprednisolone), the source of oocytes (in vivo‐ vs in vitro‐matured) or the presence or absence of the zona pellucida in embryos. In summary, methylprednisolone was effective at inducing a systemic immunosuppressed state in goats, but the treatment prior to embryo transfer did not affect pregnancy rates. In 1992 and 1993, two studies were executed in Niger to evaluate the functioning and sustainability of village plant protection brigades. The history of these brigades, their functioning, and the way the training was put into practice are discussed, as well as the problems encountered by the brigadiers. Also, the division of benefits of the brigade within the villages is dealt with. The approach adopted by the programme is discussed. It is argued that It is unlikely that many brigades will continue to exist after the end of the programme, given the costs and availability of pesticides and equipment. The fact that the bigger and richer farmers within the village benefit most undermines community based‐support. The roots of these problems can be traced back to the top‐down approach. A more participatory approach, based on the enhancement of farmers’ decision‐making and their knowledge of the environment and its risks, is advocated. 相似文献

To test the hypotheses of modern human origin in East Asia, we sampled 12,127 male individuals from 163 populations and typed for three Y chromosome biallelic markers (YAP, M89, and M130). All the individuals carried a mutation at one of the three sites. These three mutations (YAP+, M89T, and M130T) coalesce to another mutation (M168T), which originated in Africa about 35,000 to 89,000 years ago. Therefore, the data do not support even a minimal in situ hominid contribution in the origin of anatomically modern humans in East Asia. 相似文献

A field trial was performed in order to evaluate the effect on fertility of different straw types, freezing protocols (one- or two-step) and thawing procedures (35°C and 70°C) using frozen–thawed ram semen. A total of 791 Norwegian Crossbred ewes were artificially inseminated during natural oestrus with semen collected from nine mature and proven Norwegian Crossbred rams. A milk-based extender was used for dilution. The ewes were allocated into one of the following three groups based on the different straw types and thawing temperatures: medium straw (0.5 ml) thawed at 35°C for 20 s (Med35), medium straw thawed at 70°C for 8 s (Med70) and mini straw (0.25 ml) thawed at 35°C for 15 s (Mini35). The semen to be frozen in mini straws was re-concentrated by centrifugation. Sperm number in each insemination dose was approximately 200 × 106 spermatozoa. The fertility results [as 25-day non-return rate (NRR)] for Med35, Med70 and Mini35 were 53.1%, 50.8% and 58.3%, respectively, and the lambing rates 49.8%, 46.8% and 53.8%, respectively. No significant main effects were seen for straw type/thawing temperature (p = 0.17), ram (p = 0.06) or age of the ewe (p = 0.18) on NRR or lambing rates (p = 0.19, p = 0.16 and p = 0.27, respectively). Here we discuss the novel application of handheld infrared thermal imagery as a learning tool to help smallholder farmers improve postharvest temperature management. Farmers in the Fiji Islands were trained in using a handheld thermal camera to view a series of on-farm demonstrations highlighting the consequence of poor postharvest practice. We observed that smallholder farmers accurately interpreted thermal imagery and the implication of poor practices. Our results showed thermal imagery has the potential to better communicate fundamental concepts of postharvest management. The availability of a range of iPhone, iPad, and Android platforms also means this technology can be widely accessible. 相似文献

This study assessed the effect of halal slaughter and anesthesia pre‐slaughter followed by bleeding on meat quality characteristics of goats. Eleven male Boer cross goats were divided into two groups and subjected to either halal slaughter (HS) or anesthesia with halothane and propofol pre‐slaughter (AS). At pre‐rigor, HS had significantly lower (P < 0.05) muscle pH and glycogen than AS. However, no significant difference was observed in the pH and glycogen content between the treatments on 1, 3 and 7 days post mortem. The drip loss of HS was significantly lower (P < 0.05) than that of AS at all aging periods. Treatment had no effect on sarcomere length, myofibrillar fragmentation index and shear force values, loss of thiol groups and degradation of major myofibrillar proteins. It can be concluded that HS did not have deleterious effect on meat quality traits of goat when compared to AS. 相似文献

Apoptosis in the testis is required to ensure an efficient spermatogenesis. However, sometimes, defective germ cells that are marked for elimination during this process escape elimination in the testes, giving rise to ejaculates with increased percentages of abnormal and apoptotic spermatozoa and a high percentage of apoptotic bodies. Apoptosis markers in the ejaculate have been associated with low fertility, either in animals or humans. Therefore, the goal of this study was to investigate whether fresh equine semen contains apoptotic bodies [initially named Merocyanine 540 (M540) bodies] and to study the relationship between the quantity of these bodies and cell concentration, the volume of ejaculate, viability and motility. Moreover, we also studied whether the presence apoptotic bodies in fresh semen was related to the resistance of the stallion spermatozoa to being incubated at 37°C or being frozen and thawed. Fresh equine semen was stained with fluorescent dyes such as M540 and Annexin‐V. Active Caspase 3 was studied in fresh semen through Western blotting and immunofluorescence with a specific antibody. Sperm kinematics was assessed in fresh, incubated and thawed samples using computer‐assisted semen analysis, and viability was evaluated with the LIVE/DEAD Sperm Viability Kit. Overall, our results demonstrate for the first time the presence of apoptotic bodies in equine semen. The quantity of apoptotic bodies was highly variable among stallions and was positively correlated with Caspase 3 activity in fresh samples and negatively correlated with the viability and motility of stallion spermatozoa after the cryopreservation process. 相似文献
