D-lactic acidosis occurs as a complication of short-bowel syndrome in humans and in a variety of other gastrointestinal disorders in monogastrics and ruminants. It is associated with signs of impaired central nervous system (CNS) function including ataxia and coma. The objective of this experiment was to determine if either acidification of nervous tissue or D-lactic acid is responsible for decreased neurological function. Eight Holstein calves (32 ± 11 d) were surgically catheterized with indwelling intravenous jugular and atlanto-occipital space cerebrospinal fluid (CSF) catheters and infused for 6 h in random order with iso-molar DL-lactic acid (DL-LA), L-lactic acid (L-LA), hydrochloric acid (HCl) or saline. DL-LA induced ataxia after 4 h of infusion and produced the greatest obtunding of CNS function (at 7 h, score 8.0 ± 0.4), whereas, the other infusions caused neither ataxia nor scores over 1.5 (p < 0.01 from DL-LA). After 6 h, DL-LA induced significantly less acidemia than HCl (pH 7.13 ± 0.06 and 7.00 ± 0.04, base excess −16 ± 1 and −23 ± 3 mmol/l, bicarbonate 11 ± 1 and 8 ± 1 mmol/L respectively, all p < 0.01), but greater than L-LA and saline (p < 0.01). CSF changes followed a similar but less pronounced pattern. Although HCl infusion produced a severe acidemia and CSF acidosis, only minor effects on neurological function were evident suggesting that D-lactate has a direct neurotoxic effect that is independent of acidosis. A single experiment with a completely randomized design was conducted to evaluate the effects of long- or short-term exposure to a calf identified as persistently infected with bovine viral diarrhea virus (PI-BVD) on feedlot performance and carcass characteristics of freshly weaned, transport-stressed beef heifers. Two hundred eighty-eight heifers that had been vaccinated for BVD before weaning and transport were processed and given a metaphylactic antibiotic treatment at arrival and were fed common receiving, growing, and finishing diets for a 215-d period. Treatments were designed to directly or adjacently expose the cattle to a PI-BVD heifer. Directly exposed treatments were 1) negative control with no PI-BVD calf exposure (control), 2) PI-BVD calf commingled in the pen for 60 h and then removed (short-term exposure), and 3) PI-BVD calf commingled in the pen for the duration of the study (long-term exposure); and spatially exposed treatments were 1) negative control with no PI-BVD calf exposure (adjacent pen control), 2) PI-BVD calf commingled in the adjacent pen for 60 h and then removed (adjacent pen short-term exposure), and 3) PI-BVD calf commingled in the adjacent pen for the duration of the study (adjacent pen long-term exposure). Exposure to a PI calf transiently (60 h) or for the duration of the feeding period (215 d) did not affect (P > or = 0.25) final BW compared with heifers that were not exposed. Neither period nor overall DMI was affected (P > or = 0.37) by PI-BVD calf exposure, and no differences (P > or = 0.44) were observed between short- and long-term exposed heifers in the direct or spatially exposed groups. Likewise, total trial ADG was not affected (P > or = 0.36) and overall efficiency of gain (P > or = 0.19) was unaffected by PI-BVD calf exposure in the direct or spatially exposed groups. The results from this study suggest that exposing previously vaccinated, freshly weaned, transport- stressed beef calves to a calf that is persistently infected with bovine viral diarrhea virus has little, if any, marked effects on health, performance, or carcass characteristics. 相似文献

The giant river shrimp (Macrobrachium rosenbergii), a native species of Thailand, is either exported for commercial purposes or supplied to meet the local requirements in Thailand. Limited pharmacokinetic information of the major antibiotic, oxytetracycline (OTC), is available for this freshwater shrimp. The purpose of the present study was to investigate the muscle tissue kinetics of OTC in M. rosenbergii following either intramuscular (i.m.) or oral (p.o.) administration at two dosages of 11 and 22 mg/kg body weight (b.w.). The concentration of OTC in shrimp tissues was measured using high‐performance liquid chromatography (HPLC) equipped with a fluorescence detector. Muscle tissue concentrations were below the detection limit (LOD, 0.1 μg/g) after 96 and 120 h, following i.m. and p.o. administration, respectively. Peak muscle concentrations (Cmax) were 3.47 and 1.73 μg/g after i.m. and p.o. administration at a single dose of 11 mg/kg b.w. whereas they were 6.03 and 2.51 μg/g at a single dose of 22 mg/kg b.w., respectively. A noncompartment model was developed to describe the pharmacokinetics of OTC in the giant freshwater shrimp. The terminal half‐lives of OTC were 28.68 and 28.09 h after i.m. and p.o. administration at a single dose of 11 mg/kg b.w., but 29.95 and 27.03 h at a single dose of 22 mg/kg b.w., respectively. The relative bioavailability was 82.32 and 64.67% following i.m. and p.o. administration, respectively. Based on the pharmacokinetic data, i.m. and p.o. administration with OTC at a dose of 11 mg/kg b.w. would be appropriate for use in giant freshwater shrimp farming. To avoid the OTC residue in shrimp muscle, it should take at least seven half‐lives (8 days) to wash out the drug from the muscle of M. rosenbergii. 相似文献

AIM: To determine if there are sex- or age-related differences in the increase in skinfold thickness in response to the mitogen phytohaemagglutinin (PHA) in red deer. METHODS: One dose of 250 mug PHA was injected intradermally in the right side of the neck, and phosphate buffered saline (PBS) was injected at a second site as a control, in 110 (51 males and 59 females) captive Iberian red deer (Cervus elaphus hispanicus), ranging in age from 21 months to > or =5 years. Skinfold thicknesses were measured immediately before and 72 h following injection. RESULTS: There was a significant effect of gender on the average increase in skinfold thickness; males had greater increases (8.8 (SEM 0.57) mm) than females (4.23 (SEM 0.39) mm) after correcting for other confounding variables. No age-related differences were evident, but differences between sexes were more marked with increasing age. CONCLUSIONS AND CLINICAL RELEVANCE: Effects of gender, probably due to differences in energetic and reproductive constraints in red deer, should be taken into account when interpreting skinfold-test data, both in ecology and in the control of tuberculosis (Tb). Males tend to have a thicker skin than females, so skinfold increase relative to the thickness of the skin, rather than skinfold increase per se, should be used as a more appropriate measure of skinfold increase. Real-time PCR has become a powerful tool for the detection of inflammatory parameters, including cytokines. Reference or housekeeping genes are used for the normalization of real-time RT-PCR results. In order to obtain reliable results, the stability of these housekeeping genes needs to be determined. In this study the stability of five genes, including beta-actin, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), hypoxanthine phophoribosyl-transferase (HPRT), ubiquitin (UB) and glucose-6-phosphate dehydrogenase (G6PDH), was determined in a lipopolysaccharide inflammation model in chickens. beta-Actin appeared to be the most stable single gene in our model. Because the use of a single gene for normalization can lead to relatively large errors, the use of the geometric mean of multiple reference genes or normalization factor is preferred. The most stable combination for gene expression analysis in this lipopolysaccharide inflammation model in chickens is G6PDH and UB, since their correlation coefficients were 0.953 and 0.969, respectively (BestKeeper) and an M value of 0.34 and a low V(2/3) value of 0.155 (geNorm) were obtained. The use of HPRT and GAPDH should be avoided. The stable housekeeping genes, G6PDH and UB together, can be used to normalize the expression of pro-inflammatory cytokines in a lipopolysaccharide inflammation model in chickens. 相似文献

Epidemiological investigations implicated the semen of artificial insemination (ai) bulls as the only plausible source of infection with bovine viral diarrhoea virus (bvdv) in 10 Finnish dairy herds. The infection was traced back to two northern Finncattle bulls that had been transiently infected when their semen had been collected while they were in a gene bank herd containing persistently infected (pi) animals. The isolates of bvdv from the animals in the gene bank herd, from the semen of the two bulls and from a pi calf born in one of the herds using the semen belonged to a rare genetic type in Finland and, on the basis of the nucleotide sequences in the 5' untranslated region, were identical. Cross-contamination of batches of semen at the ai station and an external source of bvdv were ruled out for the recipient herds. It was concluded that bvdv infection can be transmitted through the semen of transiently infected bulls under field conditions. 相似文献

This case report describes the findings in a seven-month-old heifer with diabetes insipidus attributable to internal hydrocephalus. The heifer was referred to the clinic because of reduced appetite, polydipsia, decreased faecal output and weight loss. The heifer was examined daily for 8 days. She was thin and weak and had a dull dry hair coat and decreased appetite. The heifer urinated frequently; the urine was clear and yel low, had a specific gravity of 1.015. A complete blood cell count, biochemical profile and blood gas analysis revealed increased serum urea, increased serum creatinine, hypernatraemia, hyperchloraemia, hypercalcaemia and hypophosphataemia. The heifer received 10 litres of water and 3 litres of ruminal fluid from a healthy cow per os daily for 5 days. The heifer had access to fresh water ad libitum. The general condition of the heifer did not improve after this treatment. Although the concentration of serum urea and creatinine decreased, the concentrations of sodium, chloride and calcium remained higher than normal. Based on the findings, a diagnosis of diabetes insipidus was made and the heifer was euthanatized. Postmortem examination revealed severe internal hydrocephalus, and a definitive diagnosis of central diabetes insipidus attributable to internal hydrocephalus was made. 相似文献

This case report describes the findings in a 10-year-old goat with metastasis of an adenocarcinoma in the iris. Two weeks before admission, the owner had noticed blepharospasm of the left eye. Clinical examination by the referring veterinarian revealed unilateral intraocular hemorrhage. The goat was referred to our clinic for further work-up. The rectal temperature was 40 degrees C. The most important haematological result was severe thrombocytopenia. There was mild corneal oedema of the left eye. Approximately 75 per cent of the anterior chamber was filled with non-coagulated blood. The fluid in the anterior chamber dorsal to the blood was cloudy, and the corpora nigra could not be seen clearly. All other internal parts of the eye could not be seen. Ultrasonography of the left eye confirmed cloudiness of the anterior chamber and revealed moderate thickening of the iris. The right prescapular lymph node was markedly enlarged. Cytological examination of a fine needle aspirate of the lymph node showed a mixed population of neoplastic cells. Based on immunohistochemical evaluation of the cells metastasis of a carcinoma was diagnosed. The goat was subjected to euthanasia, and a postmortem examination was carried out. The anterior chamber of the left eye contained blood, and the iris was thicker than normal and adhered to the posterior surface of the cornea. There were neoplastic alterations in the iris, the oesophagus, the lung lobes, the liver, the kidney and in the prescapular, retropharyngeal, mediastinal and hepatic lymph nodes. Histologically, a diagnosis of carcinoma was confirmed, but the origin of the tumour could not be determined. 相似文献
