Freshly harvested potato tubers contain up to 50 mg/100g ascorbic acid (AA), but levels decline rapidly during cold storage. Genetic alterations to boost the AA content of stored tubers would contribute to human nutrition and might improve tuber resistance to oxidative damage during chilling. While studying the breeding potential ofSolanum phureja germplasm, we identified a 24-chromosome Phureja-haploid Tuberosum hybrid (clone I, USW5295.7) that retained a twofold higher content of tuber AA than neighboring clones after storage at 5 C. Clone I produces 2n-pollen through a mechanism genetically equivalent to first-division restitution (FDR), which transmits much of the nonadditive genetic variance for tuber yield. We now report a survey of clone I progeny showing significant family and ploidy effects on tuber AA levels during cold storage, consistent with the transmission of information for higher AA by FDR 2n-pollen. A 7-year-old female Leonberger dog was referred to the National Veterinary School of Lyon Teaching Hospital with a 2-day history of anorexia and bleeding. A mammary mass had been removed 7 months earlier, but histologic examination was not performed. On physical examination, the dog was depressed and had pale mucous membranes and numerous petechiae and hematomas. Significant laboratory findings were moderate thrombocytopenia, prolonged prothrombin, activated partial thromboplastin, and thrombin times, hypofibrinogenemia, and increased concentration of fibrin(ogen) degradation products. A peripheral blood smear, buffy coat preparation, and bone marrow aspirate contained low numbers of large atypical cells that had moderate nuclear:cytoplasmic ratios, oval nuclei with multiple prominent nuclei, and basophilic cytoplasm with villous projections. A small nodule was found in the left inguinal mammary gland, and a fine-needle aspirate contained cells similar to those in blood and bone marrow. In samples of blood, bone marrow, and the mammary mass, the neoplastic cells were immunoreactive for cytokeratin. The diagnosis was mammary carcinoma with secondary disseminated intravascular coagulation (DIC) and disseminated tumor cells in bone marrow and circulating tumor cells in blood; this diagnosis was not confirmed by histopathologic examination. Owing to clinical deterioration and the poor prognosis, the dog was euthanized and a necropsy was not performed. Grasslands are often characterized by small-scale spatial heterogeneity due to the juxtaposition of grass tufts and bare ground. Although the mechanisms generating plant spatial patterns have been widely studied, few studies concentrated on the consequences of these patterns on belowground macrofauna. Our objective was to analyze the impact of grass tuft (Brachiaria bryzantha cv. marandu) spatial distribution on soil macrofauna diversity in Amazonian pastures, at a small scale (less than 9 m2). Soil macrofauna was sampled among B. bryzantha tufts, which showed a variable spatial distribution ranging from dense to loose vegetation cover. The vegetation configuration explained 69% of the variation in total soil macrofauna density and 68% of the variation in total species richness. Soil macrofauna was mainly found in the upper 10 cm of soil and biodiversity decreased with increasing distances to the nearest grass tuft and increased with increasing vegetation cover. The size of the largest grass tuft and the micro-landscape connectivity also had a significant effect on biodiversity. The density and species richness of the three principal soil ecological engineers (earthworms, ants and termites) showed the best correlations with vegetation configuration. In addition, soil temperature significantly decreased near the plants, while soil water content was not influenced by the grass tufts. We conclude that soil macrofauna diversity is low in pastures except close to the grass tufts, which can thus be considered as biodiversity hotspots. The spatial arrangement of B. bryzantha tussocks influences soil macrofauna biodiversity by modifying soil properties in their vicinity. The possible mechanisms by which these plants could affect soil macrofauna are discussed. 相似文献

Because of its pronounced estrogenicity, zearalenone may be of concern not only in the aqueous but also in the terrestrial environment. Therefore, we developed several analytical methods to quantify zearalenone in different solid matrices of agroenvironmental relevance (i.e., plant organs, soil, manure, and sewage sludge). The use of D(6)-zearalenone as the internal standard (IS) was essential to render the analytical method largely matrix-independent because it compensated for target analyte losses during extract treatment and ion suppression during ionization. Soil and sewage sludge samples were extracted with Soxhlet, whereas plant material and manure samples were extracted by liquid solvent extraction at room temperature. Absolute recoveries for zearalenone were 70-104% for plant materials, 105% for soil, 76% for manure, and 30% for sewage sludge. Relative recoveries ranged from 86 to 113% for all matrices, indicating that the IS was capable to largely compensate for losses during analysis. Ion suppression, between 8 and 74%, was in all cases compensated by the IS but influenced the method quantification levels. These were 3.2-26.2 ng/g(dryweightdw) for plant materials, 0.7 ng/g(dw) for soil, 12.3 ng/g(dw) for manure, and 6.8 ng/g(dw) for sewage sludge. Plant material concentrations varied from 86 ng/g(dw) to more than 16.7 microg/g(dw), depending on the organ and crop. Soil concentrations were between not detectable and 7.5 ng/g(dw), depending on the sampling depth. Zearalenone could be quantified in all manure samples in concentrations between 8 and 333 ng/g(dw). Except for two of the 85 investigated sewage sludge samples, zearalenone concentrations were below quantification limit. 相似文献

Little information is available regarding the molecular epidemiology of Staphylococcus aureus-induced mastitis in dairy sheep. In this study, 4 different typing techniques were compared in typing 26 S. aureus isolates, predominantly from cases of subclinical mastitis in dairy ewes. The 4 techniques were pulsed-field gel electrophoresis (PFGE), restriction fragment length polymorphism (RFLP) on 2 genes (coagulase and clumping factor B), randomly amplified polymorphic DNA-polymerase chain reaction (PCR) (RAPD-PCR), and multilocus sequence typing (MLST). On the basis of discriminatory power as the key parameter of typing systems, MLST and PFGE were found to be the most powerful techniques. The MLST and PFGE could contribute to epidemiological surveillance and evaluation of mastitis control programs, by documenting prevalence and dissemination of endemic clones in infected populations. The results of this study show that a single clone of S. aureus is widely distributed in infected ewe mammary glands. 相似文献

Interest in the incorporation of riparian buffers and forest farming were modeled following agricultural conservation and
agroforestry adoption studies. Attitudes, individual characteristics, economic diversity of landowners’ household portfolio,
and physical and ecological conditions were explanatory variables in Logit regression models of interest. Habitus and field, the values and institutions of farm operators, were included in the framework. Knowledge of the practice, perceived problems
with the environment and attitude/habitus variables had a positive effect on the probability of being interested in riparian buffers. Knowledge of the practice was
the most important factor in explaining interest in the case of forest farming, followed by attitudes about trees and concern
for future generations, both habitus variables. Field variables like CRP payments representing familiarity with government programs that support conservation, had no significant
effect on interest in riparian buffers. Older farmers were less interested in both practices, consistent with other findings.
Existing economic diversification of the household portfolio had no effect on the probability of being interested in either
practice.

Whole blood and serum from 93 cats (44 pets and 49 shelter/stray cats) from Denmark were tested for the presence of feline Bartonella species by culture and for the presence of Bartonella antibodies by serology. Bartonella henselae was isolated from 21 (22.6%) cats. Bacteremia prevalence was not statistically different between shelter/stray cats (13/49, 26.5%) and pet cats (8/44, 18.2%), but varied widely by geographical origin of the cats, even after stratification for cat origin or age (p < 0.001). All isolates but one were B. henselae type II. The only cat bacteremic with B. henselae type I was not co-infected with B. henselae type II. None of the cats was harboring either B. clarridgeiae or B. koehlerae. Almost half (42/92, 45.6%) of the cats were seropositive for B. henselae and antibody prevalence was similar in shelter/stray cats (23/49, 46.9%) and pet cats (19/43, 44.2%). This is the first report of isolation of B. henselae from domestic cats in Denmark. This study also indicates that domestic cats, including pet cats, constitute a large Bartonella reservoir in Denmark. 相似文献

Streptococcus suis is an important pathogen of swine, causing meningitis, arthritis, polyserositis, septicemia, and sudden death in weaning piglets as well as fattening pigs. Recently, 3 molecular tests have been developed in our laboratory: a multiplex polymerase chain reaction (m-PCR) assay for the detection of S. suis species and serotypes 2 and 1/2, and 2 molecular typing methods, pulsed-field gel electrophoresis and an approach based on PCR amplification of a fragment of rRNA genes, including a part of the 16S and 23S genes and the 16S-23S rDNA intergenic spacer region (ISR), followed by restriction fragment length polymorphism (RFLP) analysis (ISR-RFLP). In the present study, we used these tests to analyze tonsil samples from clinically healthy pigs and to identify individual isolates of S. suis during epidemiologic investigations of 8 related herds with a history of septicemia caused by S. suis serotype 2. Capsular typing showed that 58% of the strains were nontypable. Of the 17 serotypes present, serotype 22 was the most prevalent. In the 7 farms without clinical signs on the day of sampling, we detected S. suis serotype 2 or 1/2, or both, in less than 5% of the pigs by m-PCR or by bacteriologic culture. In the 8th farm, on which 2 pigs had clinical signs of septicemia on the day of sampling, we detected S. suis serotype 2 or 1/2, or both, by m-PCR in the tonsils of 40% of fattening pigs (21 wk old) that lacked symptoms. Molecular typing of the serotype 2 strains showed a common origin of contamination in these herds, given that 1 pattern (C1) was detected in the isolates from 6 of the 8 herds. However, up to 4 patterns were associated with septicemia and sudden death. Several patterns of S. suis serotype 2 can be responsible for disease in the same herd. These molecular tools may be useful for confident studies of the transmission of S. suis, thereby contributing to the control of S. suis infection. 相似文献
