The official AOAC manual Kjeldahl method for determining crude protein in animal feeds, 7.015, uses HgO as a catalyst in the digestion step. Because of environmental considerations, there is considerable interest in alternative catalysts. A collaborative study compares the official HgO-catalyzed method and an alternative using CuSO4. Fifty-four samples consisting of blind duplicates of closely matched pairs, representing a range of animal feed materials and 2 standard materials, were analyzed once by each method. Results were returned by 22 laboratories. Means and standard deviations between methods were comparable. The CuSO4-catalyzed method has been adopted official first action.  相似文献

Canine granulocytic anaplasmosis is a disease diagnosed in many areas of the United States, with the highest prevalence reported in dogs in the upper midwestern, northeastern, and western states. It is also found in several European countries. Anaplasmosis can manifest with a wide range of clinicopathologic findings. A 20 wk old puppy was presented with physical examination and laboratory findings atypical of the disease. In addition to other signs, abdominal effusion was detected. Diagnostic evaluation confirmed that the puppy was positive for anaplasmosis. No morulae were found in peripheral white blood cells, but morulae were apparent in the white cells of the abdominal effusion. Leukopenia was observed, characterized by neutropenia, as opposed to the lymphopenia and eosinopenia typically seen with the disease. Blast cells were also seen in the peripheral blood, and reactive lymphocytes were noted in the bone marrow. The patient responded well to doxycycline therapy. All hematologic and physical abnormalities resolved.  相似文献

SUMMARY A multivalent footrot vaccine has been developed, containing pilus antigens produced in recombinant Pseudomonas aeruginosa and representing all nine serogroups of Dichelobacter (Bacteroides) nodosus commonly recognised in the field. The responses of sheep to the multivalent vaccine have been compared with those to monovalent vaccines representing only a single serogroup. Antigenie competition between serogroups occurred in sheep immunised with the multivalent formulation, but high levels of protection were still achieved. The study showed that in multivalent footrot vaccines, antigenic competition is predominantly due to the presence of a family of immunologically-related pilus antigens rather than to interference by extraneous proteins.  相似文献

The plasma disposition kinetics of ivermectin following a single subcutaneous administration of 0.2 mg/kg was investigated in male and female Senegalese Peulh sheep. Ten clinically healthy animals (5 males and 5 females) weighing 38–45 kg were used in this trial. Blood samples were collected by jugular puncture at different times between 0.5 h and 30 days post treatment. After plasma extraction and derivatization, samples were analysed by HPLC with fluorescence detection. Computerized kinetic analysis was carried out and mean parameters were statistically compared with the Mann–Whitney U-test. The area under the concentration–time curve (AUC) was significantly higher (p < 0.0027) in females than in males. Although the differences in maximum concentration (C max), mean residence time (MRT) and half-life of elimination (t 1/2el) between males and females did not achieve statistical significance, values tended to be higher in females. Sex differences may be parallel with the level of storage in fat. Further investigations are required to improve the use of ivermectin in Senegalese sheep and findings may be used to predict optimal anthelmintic strategies for management of African species depending on the parasites present in a production system.  相似文献

To establish chronic infections, viruses must develop strategies to evade the host's immune responses. Many retroviruses, including mouse mammary tumor virus (MMTV), are transmitted most efficiently through mucosal surfaces rich in microbiota. We found that MMTV, when ingested by newborn mice, stimulates a state of unresponsiveness toward viral antigens. This process required the intestinal microbiota, as antibiotic-treated mice or germ-free mice did not transmit infectious virus to their offspring. MMTV-bound bacterial lipopolysaccharide triggered Toll-like receptor 4 and subsequent interleukin-6 (IL-6)-dependent induction of the inhibitory cytokine IL-10. Thus, MMTV has evolved to rely on the interaction with the microbiota to induce an immune evasion pathway. Together, these findings reveal the fundamental importance of commensal microbiota in viral infections.  相似文献

Poultry litter leachate (PLL) is known to contain a variety of contaminants including endocrine disrupting compounds (EDCs). This study analyzed the presence of steroids and contaminants in samples of poultry litter from a broiler poultry operation in Maryland, USA. Litter samples were homogenized, hydrated, incubated for two time periods (4 and 24?h) at two temperatures (20°C and 37°C), filtered, and analyzed for steroids and anthropogenic contaminants. In addition, duplicate samples were spiked with 17?\?? estradiol (E2) and testosterone (T), and ??-glucuronidase and aryl sulfatase, to measure steroid recovery and the presence of conjugates, respectively. A steroid recovery rate of 71 and 73% was obtained from E2 and T spiked samples, respectively. Increased incubation duration demonstrated an increased trend in E2 and a decreased trend in androgen (T and/or dihydrotestosterone [DHT]) concentrations, regardless of temperature. In contrast, increased incubation temperature displayed different trends in E2 and androgen concentrations. High temperature with a 4-h incubation resulted in an increased trend in androgen with no effect on E2. However, after 24?h of incubation at high temperature, an increased trend in E2 was observed with no effect on androgen. The presence of de-conjugating enzymes resulted in a greatly increased trend in T concentrations with a slight increased trend of E2 concentrations. Trace amounts of several metals and anthropogenic compounds were detected. Arsenic, barium, endosulfan, and bis (2-ethylhexyl) phthalate were detected at quantifiable levels. This study demonstrates that PLL contains potential EDCs and contaminants that can be toxic to, and bioaccumulate in, aquatic fauna. Determination of EDC concentrations in environmental samples is important to elucidate potential detrimental effects of agricultural runoff on aquatic wildlife.  相似文献

The shear-pile-disk and chop-herbicide-burn treatments had similar overall growth pattern, and will approach the same level of pine volume as the herbicide-burn treatment.

    In southeastern pine–oak ecosystems, ecological restoration targets oaks for removal by chemical, mechanical, burning, or combinations of treatments. Managers often pursue oak removal indiscriminately despite the poorly understood historical structure, cover, and ecological function within these ecosystems. Restoration treatments often cite the impediment that oak litter represents to prescribed fire spread and effectiveness. We evaluated the burning characteristics of eight southeastern Quercus spp. by burning collected litter under controlled conditions in a combustion chamber. Replicated burns consisted of 15 g of litter on a 35 cm × 35 cm grid of xylene-soaked cotton strings. Burning characteristics measured included maximum flame height (cm), flaming duration (s), smoldering duration (s), residual ash, and mass loss rate (g s−1). We compared all 8 oaks using ANCOVA, with litterbed depth as a covariate. The oaks differed for all burning characteristics measured (P < 0.001). Rank comparisons placed Quercus stellata and Quercus laevis as the species with greatest fire intensity, sustainability, and consumability, equivalent in many measures to longleaf pine and other fire resisters. Quercus virginiana and Quercus hemisphaerica burned with the least intensity, sustainability, and consumability, burning similarly to sand pine and other fire evaders. These results show that oaks common to southeastern United States ecosystems have litter properties, similar to pines, which vary in their ability to sustain fire. Understanding the pyric properties of oak species also suggests that managers prioritize removal of species that hinder prescribed fire effectiveness for restoration of southeastern USA pine–oak ecosystems.  相似文献

    Ophidiomyces ophiodiicola, the causative agent of snake fungal disease, is proposed as a serious threat to the conservation of several snake populations. The objective of this study was to determine the pharmacokinetic parameters of terbinafine administered through nebulization and a sustained subcutaneous implant as potential treatments of Ophidiomyces in reptiles. Seven adult cottonmouths (Agkistrodon piscivorus) were used in single‐dose trials. Each snake was nebulized with terbinafine (2 mg/ml) for 30 min and had blood collected before nebulization and up to 12 hr after nebulization. Following a 5‐month washout, the same snakes were administered a subcutaneous implant containing 24.5 mg terbinafine; blood was collected at baseline, 1 day postimplant placement, and then once weekly for 9 weeks. Plasma for both studies was analyzed by high‐performance liquid chromatography. The mean plasma concentrations of nebulized terbinafine peaked between 0.5 and 4 hr. The subcutaneously implanted terbinafine reached therapeutic concentrations on day 1 and maintained therapeutic for over 6 weeks. These methods and doses are recommended as potential treatment options for snake fungal disease in reptiles.  相似文献
