ABSTRACT: The effects of the different catching methods (gillnet, longline, harpoon) on sensory, chemical (pH, total volatile base nitrogen, K -value) and microbiological (total viable count [TVC]) changes in pike perch Sander lucioperca stored in ice were investigated. The same soaking time was used for both gillnet and longline fishing. The catching method had considerable influence on the freshness quality of pike perch. The acceptable shelf life was 15 days for pike perch caught by gillnet, and 22 days for longline and harpoon. The initial concentrations of inosine monophosphate (2.4 μmol/g) in pike perch caught by gillnet were significantly lower ( P < 0.05) than longline (4.1 μmol/g), and especially by harpoon (16.7 μmol/g). However, the initial K -values for fish caught by harpoon were significantly ( P < 0.05) lower (24.36%) than fish caught by longline and gillnet (57.69%, 64.41%, respectively). The average K , Ki, G and H -values at rejection day in terms of sensory assessment were approximately 90, 98, 156 and 40%, respectively, for all catching methods during ice storage. However, TVC reached 7.0 log cfu/g after approximately 11 days of storage for fish caught by gillnet, 19 days for fish caught by longline and 8 days for fish caught by harpoon. The result of this study suggests that the best catching method for preserving the freshness of pike perch is longline, based on the data obtained from the sensory and microbiological analysis. 相似文献

ABSTRACT: Post-release movement and diel activity patterns of hatchery-reared and wild black-spot tuskfish were examined using ultrasonic telemetry. Five hatchery-reared and four wild fish were released in the sandy bottom of Urasoko Bay, Ishigaki Island, Okinawa, Japan, and monitored using automated monitoring receivers from November 2005 to February 2006. Both hatchery-reared and wild fish tended to stay near the release site for over two weeks, before leaving the release site. Both hatchery-reared and wild tuskfish showed diurnal rhythm intermittently; signals were recorded more frequently in the daytime and less frequently in the nighttime, suggesting that the fish of both origins were active during the day and inactive during the night. These findings indicate that the one-year-old hatchery-reared tuskfish have some consistent behavioral characteristics with those of the wild. 相似文献

ABSTRACT: This study was undertaken to describe and quantitatively analyze the pattern of development of gametes in the Japanese scallop Patinopecten yessoensis . Incorporation of 5-bromo-2'-deoxyuridine (BrdU) into gonial cells was used to quantitatively detect mitotically active gonial cells. Oogonia increased in number from November to December and decreased rapidly to March. A small number of oocytes was detected in November. Oocytes steadily increased in number and size up to March. The number of spermatogonia slightly increased from November to December, and increased markedly from January to March. Both ratios of BrdU-immunopositive gonial cells in the ovary and testis to gonial cells moderately increased from September to December. The ratio of BrdU-immunopositive spermatogonia to gonial cells drastically increased from January to February and kept an elevated level in March, whereas the oogonia started to disappear in January. The results suggest that the pattern of proliferation of gonial cells can be divided into two phases: (i) phase I, oogonia and spermatogonia slowly proliferate through the growing stage; and (ii) phase II, oogonia develop into oocytes and spermatogonia begin to proliferate rapidly through the mature and spawning stages. The use of specific immunoglobulin heavy chain variable region (VH) genes has been associated with increased patient survival in human B-cell lymphomas (hBCL). Given the similarity of human and canine BCL (cBCL) in morphology and clinical treatment, we examined the choice of VH in cBCL and determined whether VH gene selection was a distinct feature associated with survival time in dogs. VH gene selection and mutational status in 52 cBCL, including 29 diffuse large B-cell lymphomas (cDLBCL, the most common subtype of cBCL), were analyzed by comparison with the 80 published canine germline VH gene sequences. We further examined the prognostic impact of the subgroups defined by these features on canine survival. We found that VH1-44 was preferentially expressed in the majority of the 52 cBCLs (60%) as well as in the majority of the cDLBCL subset (59%). VH1-44 gene expression was associated with a statistically better overall survival (p = 0.039) in cBCL patients, as well as in the cDLBCL subset of patients (p = 0.038). These findings suggest that VH gene selection in cBCL is not random and may therefore have functional implications for cBCL lymphomagenesis, in addition to being a useful prognostic biomarker. 相似文献

This study evaluated measures of sperm quality in relation to fertility achieved with fresh semen or semen cooled and stored. Semen from 1 stallion was collected and processed to provide 3 treatments: group 1 received fresh semen; group 2 received cooled semen containing 50% seminal plasma (SP) stored for 4 days; and group 3 received cooled semen containing 50% SP stored for 1 day, then centrifuged and resuspended in fresh extender containing 10% SP on days 1 to 3. Inseminates were evaluated for sperm motion characteristics and the percentage of sperm with intact membranes (SMI). Mares (n = 34) in estrus were treated with an ovulation-inducing drug and inseminated with 100 million membrane-intact sperm on the following day. Pregnancy status was determined via transrectal ultrasonography 2 weeks after ovulation. The mean percentage of SMI was higher in group 1 (81%, initial) than in group 2 (74%, day 4) or group 3 (74%, day 4) (P < .05). The median percentages of total sperm motility differed among the groups (77%, 5%, 59% for groups 1, 2, and 3 respectively; P < .05). Median values for the percentages of progressively motile sperm and curvilinear velocity for group 1 (55%, 216 μm/s) and 3 (37%, 186 μm/s) were higher than for group 2 (1%, 73 μm/s) (P < .05). Pregnancy rates did not differ among groups (5 of 11, 45% in group 1; 5 of 11, 45% in group 2; and 7 of 12, 58%, in group 3; P = .77). These data suggest that, at least for this stallion, sperm membrane integrity may be a more valuable means of assessing potential fertility of cooled-stored semen than sperm motion characteristics. 相似文献

Low pathogenicity avian influenza virus (LPAIV) and lentogenic Newcastle disease virus (lNDV) are commonly reported causes of respiratory disease in poultry worldwide with similar clinical and pathobiological presentation. Co-infections do occur but are not easily detected, and the impact of co-infections on pathobiology is unknown. In this study chickens and turkeys were infected with a lNDV vaccine strain (LaSota) and a H7N2 LPAIV (A/turkey/VA/SEP-67/2002) simultaneously or sequentially three days apart. No clinical signs were observed in chickens co-infected with the lNDV and LPAIV or in chickens infected with the viruses individually. However, the pattern of virus shed was different with co-infected chickens, which excreted lower titers of lNDV and LPAIV at 2 and 3 days post inoculation (dpi) and higher titers at subsequent time points. All turkeys inoculated with the LPAIV, whether or not they were exposed to lNDV, presented mild clinical signs. Co-infection effects were more pronounced in turkeys than in chickens with reduction in the number of birds shedding virus and in virus titers, especially when LPAIV was followed by lNDV. In conclusion, co-infection of chickens or turkeys with lNDV and LPAIV affected the replication dynamics of these viruses but did not affect clinical signs. The effect on virus replication was different depending on the species and on the time of infection. These results suggest that infection with a heterologous virus may result in temporary competition for cell receptors or competent cells for replication, most likely interferon-mediated, which decreases with time. 相似文献
