The correct measurement of spermatic concentration in boar semen is one of the basic tasks routinely performed at the Swine Artificial Insemination Center (CIAS). This article explains two experiments whose objectives were: Experiment 1: This trial had two objectives: (1) to evaluate the precision of the spermdensimeter (SPM) with relation to the Neubauer count chamber (CN) which is considered the standard technique in the field and (2) to correct the scale on the SPM table. Experiment 2: This trial also had two objectives: (1) to evaluate the time and repetitions required in the SPM and (2) to compare the SPM adjusted table produced in Expt 1 with the SPM standard table. The methods in Expt 1 were used to measure total spermatic concentration in 141 boar ejaculates. The average spermatic concentrations (10(6) spermatozoids/ml) and average dose number were 229.1 and 22.6 for CN, and 283.6 and 28.0 for SPM. These results indicate that the SPM has a tendency to overestimate spermatic concentration with relation to the CN. The SPM produced a smaller coefficient of variation and needed lesser examination time than the CN (p < 0.05). The SPM adjusted table created in Expt 1 produced similar results as the SPM and CN standard tables (2.96 billion live spermatozoids/dose vs 2.36 billion live spermatozoids/dose). Incorporation and evaluation of supplemental data from other CIAS regarding the use of the SPM adjusted table in the field entitled 'Adjusted Table for Karras Spermdensimeter LPS-FMVZ-USP' would be valuable to prove its efficiency.  相似文献

The use of hormonal protocols in puberty induction and synchronization of oestrus has lead to an increase in the efficiency of replacement gilts. The aim of this study was to evaluate different doses of porcine LH in precocious puberty induction and oestrus synchronization in a homogeneous group of gilts. Sixty-seven homogeneous prepubertal gilts (Camborough 22) at 137 +/- 4 days of age and 87 +/- 7 kg were treated with three different hormonal protocols: T1--600 UI of equine chorionic gonadotrophin (eCG; Novormon) and after a 72-h period 5 mg of LH (Lutropin); T2--600 UI of eCG and a 72-h period 2.5 mg of LH; T3--600 UI of eCG and a 72-h period 1.25 mg of LH. The ovaries were examined at slaughter, on day 6 after the hormonal treatment. There were no statistical differences (p > 0.05) between the different LH doses in the percentage of the detected oestrus (T1 = 42.85%; T2 = 60.87%, T3 = 52.18%), oestrus duration (T1 = 41.44 +/- 16.30 h; T2 = 48.57 +/- 16.29 h, T3 = 39.33 +/- 11.42 h), number of corpora lutea (T1 = 9.61 +/- 5.43; T2 = 9.86 +/- 3.32, T3 = 8.13 +/- 5.52) and percentage of animals presenting ovarian cystic degeneration (T1 = 33.33%; T2 = 39.13%, T3 = 39.13%). The T2 (2.5 mg of LH) presented the lowest dispersion (p < 0.05) of the LH-ovulation interval (T1 = 37.17 +/- 4.07 h; T2 = 38.26 +/- 2.84 h; T3 = 36.25 +/- 5.69 h). The LH dose reduction to 2.50 and 1.25 mg presented equal results with the recommended dose of 5.0 mg, and could be used in the precocious induction of oestrus in gilts. The 2.5-mg LH dose showed the lowest dispersion of ovulation and it can be used in fixed-time artificial insemination programmes.  相似文献

X-ray fluorescence spectra obtained by the MESSENGER spacecraft orbiting Mercury indicate that the planet's surface differs in composition from those of other terrestrial planets. Relatively high Mg/Si and low Al/Si and Ca/Si ratios rule out a lunarlike feldspar-rich crust. The sulfur abundance is at least 10 times higher than that of the silicate portion of Earth or the Moon, and this observation, together with a low surface Fe abundance, supports the view that Mercury formed from highly reduced precursor materials, perhaps akin to enstatite chondrite meteorites or anhydrous cometary dust particles. Low Fe and Ti abundances do not support the proposal that opaque oxides of these elements contribute substantially to Mercury's low and variable surface reflectance.  相似文献

The fabrication of improved ceramic-matrix composites will require a better understanding of processing variables and how they control the development of the composite microstructure. Noninvasive, high-resolution methods of x-ray tomography have been used to measure the growth of silicon carbide in a woven Nicalon-fiber composite during chemical vapor infiltration. The high spatial resolution allows one to measure the densification within individual fiber tows and to follow the closure of macroscopic pores in situ. The experiments provide a direct test of a recently proposed model that describes how the surface area available for matrix deposition changes during infiltration. The measurements indicate that this surface area is independent of the fiber architecture and location within the preform and is dominated by large-scale macroporosity during the final stages of composite consolidation. The measured surface areas are in good agreement with the theoretical model.  相似文献

Mutations in anc-1 (nuclear anchorage defective) disrupt the positioning of nuclei and mitochondria in Caenorhabditis elegans. ANC-1 is shown to consist of mostly coiled regions with a nuclear envelope localization domain (called the KASH domain) and an actin-binding domain; this structure was conserved with the Drosophila protein Msp-300 and the mammalian Syne proteins. Antibodies against ANC-1 localized cytoplasmically and were enriched at the nuclear periphery in an UNC-84-dependent manner. Overexpression of the KASH domain or the actin-binding domain caused a dominant negative anchorage defect. Thus, ANC-1 may connect nuclei to the cytoskeleton by interacting with UNC-84 at the nuclear envelope and with actin in the cytoplasm.  相似文献

Abstract

Field average based recommendations have been a common practice for recommending the major crop nutrients nitrogen (N) and phosphorus (P). The problem is yield will not be the same from year to year with application of the same amount of recommended rate of fertilizer. The objectives of this study were to demonstrate how recommendations generated using nutrient response experiments were dynamic; and to assess the relative contribution of temporal variability, N and P fertilizers on winter wheat grain yield and N concentration. Twelve factorial combinations of four N (0, 56, 112, and 168 kg ha?1) and three P (0, 14.5, and 29 kg P ha?1) rates were evaluated in a randomized complete block design with three replications at Perkins, Oklahoma. To address the first objective, ANOVA and orthogonal polynomial contrasts were used. To address the second objective, a ten predictor variable multiple linear regression model with two quantitative variables and their interaction (N, P and N×P) and seven-year variables was evaluated and a reduced model containing seven variables was generated. Wheat grain yield showed three distinct responses to N rates: Linear, quadratic and no response. These individual year data show that it is not always appropriate to use results of nutrient response experiments to estimate next year's N fertilizer requirement due to apparent temporal variability in the results. Wheat only responded to P during the first two years of the study. The reduced model from the regression analysis revealed that most of the variability in grain yield was accounted for by five individual indicator years and N only. High variability across years in grain yield and fertilizer (N and P) response, even between years of similar grain yield, is an indication of a given season's production dependence on factors other than N and P.  相似文献

Improved quantification is needed for long‐term soil organic carbon (SOC) transport in runoff at watershed scales. Coshocton wheel samplers were used to collect runoff samples from no‐till and chisel‐till watersheds in corn (Zea mays) and soybean (Glycine max) rotations over 13 years. Samples were analyzed for SOC, N, P, K, and soil losses. The SOC losses, ranging from 0 to 357 kg ha−1 event−1, were correlated (r2 = 0·80–0·94) in power law relationships with N, P, K, soil loss, and runoff. Two events occurring in corn when soybean and cover crop residue were present in no‐till had combined SOC transport of 460 kg ha−1, nearly double the no‐till losses of a previous 11‐year period and 20 times higher than chisel‐till in the same events. Infrequent, extreme transport events that are not well characterized empirically, particularly in no‐till, can strongly influence hydrologic C transport from agriculture watersheds.
