Bovine ovarian stem cells differentiate into germ cells and oocyte‐like structures after culture in vitro

Stem cells have been isolated from ovaries, and their ability to differentiate into oocytes in vitro has been demonstrated for mice and human, but not for bovine species. The aims of this study were to isolate germline stem cells from bovine ovaries and to evaluate the effects of bone morphogenetic proteins (BMPs) 2 and 4, and follicular fluid on the differentiation of these stem cells into oocyte‐like structures. The ovarian stem cells were isolated and cultured in α‐MEM⁺ supplemented with BMP2, BMP4 or follicular fluid. On days 0 and 14, cells were evaluated for their morphological appearance, viability, expression of alkaline phosphatase and for markers of germ cell formation (VASA and DAZL) and oocyte development (GDF9, ZPA and SCP3) by qPCR. Levels of mRNA were analysed using ANOVA and Bonferroni test (p < .05). The results showed that at day 0, ovarian stem cells expressed specific markers of pluripotency (OCT4, SOX). In addition, these cells were positive for alkaline phosphatase, which is a marker commonly used to identify primordial germ cells (PGCs). After the period of differentiation, cells had morphological features that resemble PGCs and oocyte‐like cells (OLCs). An increase, ranging from five to 14 times, in the expression of VASA was observed in cells cultured in medium supplemented with BMPs and follicular fluid, while the increase in DAZL expression ranged from four to six times. In addition, OLCs had an increase in expression of mRNAs for GDF9, ZPA and SCP3 that ranged from two to eight times. In conclusion, OLCs can be differentiated in vitro from ovarian stem cells and BMPs and follicular fluid are effective in stimulating the expression of mRNAs for germ cell and oocyte markers.     

Resistance by the blue tick (Boophilus decoloratus) to the synthetic pyrethroid, fenvalerate

The Shaw larval test, in conjunction with adult tick immersion and stall tests, was utilized to confirm that a field strain of B. decoloratus, from Natal, is highly resistant to the ixodicide fenvalerate (Factor of resistance 4,744). This resistance developed over a reported 18 months of usage for cattle dipping.     

The electrocardiogram of anesthetized captive adult emus (Dromaius novaehollandiae)

ObjectiveTo characterize the electrocardiogram (ECG) of anesthetized adult emus (Dromaius novaehollandiae).AnimalsTen clinically healthy adult emus anesthetised for routine physical examination and an electrocardiogram, for both monitoring and investigation into any evidence of cardiac disease.MethodsThe ECGs for each emu were obtained in right lateral recumbency, using a modified electrode placement that replicated the standard bipolar leads used in small mammals. Lead II was used for waveform analysis.ResultsMedian P wave amplitude was 0.55 mV (range: 0.2–0.92 mV) and P wave duration was 0.06 s (0.04–0.09 s). S wave amplitude measured 1.42 mV (0.92–2.12 mV), T wave amplitude 0.67 mV (0.16–0.83 mV) and QRS duration was 0.07 s (0.07–0.12 s). Ninety percent of the QRS complexes were of rS type.ConclusionOur study provides electrocardiographic baseline data for anesthetized adult emus.     

Oocyte-specific deletion of Pten causes premature activation of the primordial follicle pool

In the mammalian ovary, progressive activation of primordial follicles from the dormant pool serves as the source of fertilizable ova. Menopause, or the end of female reproductive life, occurs when the primordial follicle pool is exhausted. However, the molecular mechanisms underlying follicle activation are poorly understood. We provide genetic evidence that in mice lacking PTEN (phosphatase and tensin homolog deleted on chromosome 10) in oocytes, a major negative regulator of phosphatidylinositol 3-kinase (PI3K), the entire primordial follicle pool becomes activated. Subsequently, all primordial follicles become depleted in early adulthood, causing premature ovarian failure (POF). Our results show that the mammalian oocyte serves as the headquarters of programming of follicle activation and that the oocyte PTEN-PI3K pathway governs follicle activation through control of initiation of oocyte growth.     

Venus: ionosphere and atmosphere as measured by dual-frequency radio occultation of mariner v

Venus has daytime and nighttime ionospheres at the positions probed by radio occulation. The main layers are thin by terrestrial standards, with the nighttime peak concentration of electrons being about two orders of magnitude below that of the daytime peak. Above the nighttime peak were several scale-height regimes extending to a radius of at least 7500, and probably to 9700, kilometers from the center of Venus. Helium and hydrogen at plasma temperatures of 600 degrees to 1100 degrees K seem indicated in the regimes from 6300 to 7500 kilometers, with cooler molecular ions in lower regions. Above the daytime peak a sharp plasmapause was discovered, marking a sudden transition from appreciable ionization concentrations near Venus to the tenuous conditions of the solar wind. This may be indicative of a kind of interaction of the magnetized solar wind with a planetary body that differs from the two different kinds of interaction characterized by Earth and by Moon. For Venus and probably for Mars, the magnetic field of the solar wind may pile up in front of the conducting ionosphere, form an induced magnetosphere that ends at the plasmapause, above which any ionosphere that tends to form is swept away by the shocked solar wind that flows between the stand-off bow-shock and the magnetopause. The neutral atmosphere was also probed and a surface reflection may have been detected, but the data have not yet been studied in detail. Results are consistent with a super-refractive atmosphere, as expected from Soviet measurements near the surface. Thus, two unusual features of Venus can be described in terms of a light trap in the lower atmosphere, and a magnetic trap in the conducting ionosphere.
