Culture techniques of antral follicle–like structure (AFLS) derived from cumulus–oocyte complexes (COCs) might provide important insights into follicular development and oocyte maturation. This study was undertaken to investigate the effects of embedding bovine COCs individually (one COC) or in groups (4–5 COCs) in collagen gels on the formation of AFLS and the meiotic status of oocytes. The observations of AFLS formation were performed every second day for 14 days. The AFLS was formed at Day 2 or 4 after the start of culture (Day = 0), irrespective of the culture methods. The mean diameters of AFLS during Days 4–14 using the individual culture method were significantly higher (p < 0.05) than those using the group culture method. However, the AFLS formation rate in the individual culture method was significantly lower compared to that in the group culture method (26.1% vs 62.7%, p < 0.01). Almost all oocytes had undergone the germinal vesicle breakdown stage, irrespective of the culture method or AFLS formation. In conclusion, comparison with the individual culture method revealed that the mean diameters of AFLS in the group culture method were smaller, but more COCs formed AFLS. The group culture method might be useful for evaluating the various hypotheses of follicular formation and interfollicular communication. However, improvement of the group culture system is necessary to prevent the meiotic resumption of oocytes, because the AFLS formation is dependent on the cumulus/granulosa cells surrounding oocytes.  相似文献

The production of sorghum is hampered by the parasitic weed Striga asiatica L. Mulching is promoted as a component of the conservation agriculture systems in sub Saharan Africa. The objective of the study was to assess the effects of mulching and Striga infestation on sorghum chlorophyll content, Striga attachments, sorghum height, and yield. The 2?×?2?×?10 factorial experiment was replicated three times for two seasons. The pot experiment was laid out as a completely randomised design. The first factor was mulching at 0 and 3?tons/ha. The second factor was infestation at two levels: infested and uninfested while the third factor was sorghum varieties at 10 levels. Data collected was plant height, chlorophyll content, Striga counts and sorghum yield. Results indicated that mulching increased chlorophyll content in the second season and not first. Infestation reduced chlorophyll content in both seasons. For the second season, mulching increased chlorophyll content in all genotypes except Ruzangwaya, Mukadziusaende and SC Sila. The genotypes Mukadziusaende, Chiredhi and Hlubi were able to maintain height despite infestation by Striga. Generally yield was higher in uninfested pots across all varieties except Mukadziusaende for the first season. Sorghum arundinaceum was highly susceptible to Striga and hence cannot be a source of resistant traits for Sorghum bicolour breeding. Mulching in Striga infested plants increased plant height and chlorophyll content. Mulching tended to diminish the pathological effects of the Striga parasite in some varieties. Therefore, mulching negated the effect of Striga parasitism and resulted in yield maintenance in some genotypes and hence can be taken up by farmers in Striga infested areas as a beneficial cultural practice.  相似文献

Phosphoinositide 3-kinases (PI3Ks) regulate fundamental cellular responses such as proliferation, apoptosis, cell motility, and adhesion. Viable gene-targeted mice lacking the p110 catalytic subunit of PI3Kgamma were generated. We show that PI3Kgamma controls thymocyte survival and activation of mature T cells but has no role in the development or function of B cells. PI3Kgamma-deficient neutrophils exhibited severe defects in migration and respiratory burst in response to heterotrimeric GTP-binding protein (G protein)-coupled receptor (GPCR) agonists and chemotactic agents. PI3Kgamma links GPCR stimulation to the formation of phosphatidylinositol 3,4,5-triphosphate and the activation of protein kinase B, ribosomal protein S6 kinase, and extracellular signal-regulated kinases 1 and 2. Thus, PI3Kgamma regulates thymocyte development, T cell activation, neutrophil migration, and the oxidative burst.  相似文献

1. Seventy-two per cent of the Flathead River catchment (22 241 km2) is federally designated and protected as wilderness or national park. Thus, the catchment remains one of the more pristine areas of its size in the temperate latitudes of the world. 2. Discharge in the downstream reaches of the river system outside the protected areas is regulated by three dams for flood control and hydropower production. These dams have blocked natural migration of native fish from Flathead Lake (496 km2) and isolated populations in sub-catchments. Temperature and erratic flow fluctuations have altered phenologies of river zoobenthos and fish, and in dam tailwaters aquatic biodiversity is drastically reduced in comparison to unregulated segments. 3. Ecological problems caused by changing water quality conditions, altered land-use patterns and introductions of non-native biota are interactive with the impacts of stream and lake level regulation, thereby emphasizing the complexity of this river–lake ecosystem. 4. Mitigation of the effects of regulation is compromised by differing management priorities and regulatory mandates of County, State, Tribal, and Federal agencies responsible for natural resource management within the catchment. Moreover, economic and ecological interests outside the Flathead influence the way flows are regulated within the catchment. 5. The most pervasive influences of stream and lake regulation can be ameliorated by retrofitting the hypolimnial release dam with a selective depth outlet structure to allow temperature control, and by controlling changes in flow rates to create a more natural hydrograph in the tailwaters of the large dams. Allowing fish passage by construction of fish ladders is problematic because upstream passage will commingle native species that were isolated upstream by construction of the dams with non-native species that were introduced subsequently below the dams. Cascading food web interactions elicited by invasions of non-native biota may offset any advantage to native stocks gained by passage and/or augmentation with hatchery stocks. 6. Mitigation must be adaptive in the sense that unanticipated effects and interactions with other management objectives can be documented and alternative action can be implemented. 7. This case history of the effects of stream and lake level regulation, and the approaches to management reviewed in this paper, should serve as a lesson in river conservation.  相似文献

The aim of this study was to test the use of mechanical and mechanical‐enzymatic methods, saline solution (SS), and PBS solution for the manipulation and isolation of mare ovarian preantral follicles (PAFs). The ovaries were subjected to mechanical isolation (mixer) alone or in association with enzymatic digestion (collagenase). Incubation times of 10 and 20 min were employed. In the first group, 4.1 ± 4.9 PAFs were harvested with the mechanical‐enzymatic method vs 71.1 ± 19.2 with the mechanical procedure, showing a significant difference between methods; using SS and PBS, these numbers were 35.7 ± 34.3 and 39.6 ± 39.6, respectively, with no significant difference between solutions. In the second group, there was significant difference between methods, with 7.1 ± 10.6 follicles harvested with the mechanical‐enzymatic method vs 63.2 ± 22.9 with the mechanical procedure; using SS and PBS, means were 35.5 ± 36.4 and 34.9 ± 31.1, respectively. The mechanical method proved more effective than the mechanical‐enzymatic approach. Both SS and PBS can be used as a media for equine PAFs preparation.  相似文献
