Generation of Salmonella ghost cells expressing fimbrial antigens of enterotoxigenic Escherichia coli and evaluation of their antigenicity in a murine model

Salmonella Typhimurium ghost cells expressing K88ab, K88ac, K99, and FasA fimbriae of enterotoxigenic Escherichia coli (ETEC) in their envelopes were constructed. The genes encoding the fimbriae were individually cloned into an expression plasmid, pMMP81, carrying the asd gene, which was subsequently electroporated into the Δasd S. Typhimurium mutant. Plasmid pJHLP99, carrying the phiX174 lysis gene E, was also subsequently electroporated into the Salmonella mutant. The presence of the individual fimbriae on the ghost cells was examined by Western blot analysis. Forty BALB/c mice were equally divided into 2 groups of 20 mice each. Group A mice were intramuscularly vaccinated with a mixture of the 4 ghost cells expressing the individual fimbriae. The group B mice were inoculated with sterile phosphate-buffered saline as a control. The antigen-specific serum IgG concentrations were significantly higher in group A than in group B from week 2 until week 6 after inoculation. In addition, the antigen-specific IgA concentrations in fecal samples were significantly higher in group A than in group B at week 2 after inoculation. A large difference between the groups in the number of antigen-specific IgA-secreting cells in the small intestine was observed by immunohistochemical study. Also, the splenic lymphocyte proliferative responses were significantly greater in group A than in the control mice. These results suggest that vaccination with our Salmonella ghost cells can induce both humoral and cell-mediated immune responses and that the increased number of antigen-specific IgA-secreting cells in the small intestine may be correlated with the elevated fecal IgA immune response.     

桑枝屑香菇与杂木屑香菇的品质比较

以桑枝屑和杂木屑为试材,采用标准及实验室常用方法对香菇营养成分进行测定,研究桑枝屑和杂木屑2种培养料对香菇产量和营养成分(蛋白质、氨基酸、脂肪、多糖、纤维、部分矿质元素)含量的影响。结果表明:桑枝屑栽培的单袋香菇产量比杂木屑的低6.4%,但生物转化率比木屑的高1.7%,粗蛋白质含量是杂木屑栽培的1.03倍,但总氨基酸含量和必需氨基酸含量都比杂木屑香菇中的略低一些;脂肪含量是杂木屑栽培的1.82倍,纤维含量是杂木屑栽培的1.01倍,多糖含量是杂木屑栽培的1.34倍;重金属铅含量仅为杂木屑的11.3%,均达到国家绿色食品标准;矿质元素除钾元素外,钙、镁、铁、锌均高于杂木屑栽培的香菇。可见,用桑枝屑栽培的香菇子实体的营养性和药用保健性优于杂木屑栽培的香菇。     

基于风险分析的养殖场生物安全体系构建

本文介绍了WOAH《陆生动物卫生法典》中风险分析的内容和要求,并结合我国《无规定动物疫病小区管理技术规范》中生物安全的相关规定和作者在养殖场生物安全体系构建中的实践经验,总结了我国养殖场生物安全体系目前存在的主要问题,提出了以风险分析为基础的养殖场生物安全体系构建的具体做法及生物安全体系的主要内容,对指导养殖场建立科学、系统的生物安全体系,提升生物安全水平,科学防控重大动物疫病具有借鉴意义。     

山东省沂南县动物强制免疫“先打后补”现状分析

2021年,沂南县推进动物疫病强制免疫“先打后补”工作,依托“鲁牧云”信息平台,通过强化组织领导,加强政策宣贯,强化人员培训,放开强免疫苗经营渠道,提供抗体检测无偿服务等措施,提升了养殖场户“先打后补”的积极性、自主性、灵活性,强化了养殖场户动物防疫主体责任,满足了养殖场户疫苗多样化需求。但由于 “先打后补”补助金额少、社会化服务不规范、基层动物防疫体系弱化、缺少配套工作经费等原因,部分养殖场户实施“先打后补”的积极性不高。建议通过规范社会化服务组织,强化培训水平,提高补助标准,强化三级（县乡村）基础设施建设,升级“鲁牧云”等措施,确保“先打后补”政策惠及更多养殖场户。     

浅谈桑树在林业可持续发展中的优势

桑树抗逆性能较强,在干旱、寒冷、风沙较大的丰宁县5个月可长高1~1.5m,当年成林,生态效益明显;15kg桑叶用于养蚕,1个月时间可转化出1kg鲜蚕茧,毛收入8~10元,周期短,经济效益高;桑叶营养丰富,其蛋白质含量是小麦的2倍,可做畜禽饲料,蚕、桑均含药物成分,可开发保健食品和药品,社会效益潜力大。在林业可持续发展中有较强的优势。     

湿地松优良半同胞家系蛋白质及糖类对水分胁迫的生理响应

为研究湿地松优良半同胞家系蛋白质及糖类对水分逆境的生理响应，采用盆栽试验和吸光度测定结合的方法，以湿地松普通种子为对照（CK），测定了七个湿地松半同胞家系在正常水分状态（土壤相对含水量为70%）、弱度水分胁迫（土壤相对含水量为55%~60%）、中度水分胁迫（土壤相对含水量为35%~40%）和强度水分胁迫（土壤相对含水量为20%~25%）条件下的可溶性蛋白质、还原性糖、可溶性糖的含量，研究结果表明：无论是不同水分梯度还是不同家系水平，蛋白质、还原性糖、可溶性糖含量的差异均达极显著性水平。随着水分胁迫程度的加剧，各家系蛋白质含量呈先升后降的趋势，在水分胁迫初期，各家系蛋白质含量增加，水分胁迫中、后期各家系蛋白含量呈直线下降；随着水分胁迫的加剧，各家系还原性糖、可溶性糖含量呈直线上升。家系1027、101、464在水分胁迫条件下具有较高的可溶性糖含量，其抗旱性较其它家系要强，是这三个家系速生、高产的生理原因之一。     

NONLINEAR COUPLING IN THERMOELAST1C THEORY AND A SOLUTION TO THE PROBLEM IN THE HALF SPACE

It's recommended that the term coupled in temperature and strain is nonlinear and is significant in thermoelastic theory. Using the solution of linear coupled and uncoupled theory and selecting appropriate general coordinates, authers approach the nonlinear coupled problems with a variational method. A solution to the problem in half space, is given. It is shown that the difference between the nonlinear coupled theory and the linear coupled or uncoupled theory would be notable when the amplitute of temperature is much larger than that of the reference slate.     

A NEW METHOD BY USING MASS AND ACCELERATION PARAMETERS FOR BALANCING INERTIAL FORCE OF MECHANISM

This paper presents a new method by using mass and acceleration parameters for balancing inertial force of mechanism. Compared with other balance methods, this one is simple, clear and superior in regularity when the balancing equation of complex mechanism is established, and its solving procedure is also easy.
