Migrating duck populations in Wisconsin were evaluated during a one-year period for exposure to Aspergillus fumigatis by two serodiagnostic methods: immunodiffusion and countercurrent immunoelectrophoresis. Peak antibody reactivity to sporulating culture extracts of the fungus was detected in serum samples collected in winter. Brucella suis is an emerging, zoonotic disease predominantly affecting dogs and humans that engage in feral pig hunting in Australia and other countries. Although B. suis infection in dogs shares some clinical similarities to the host-adapted species (B. canis), B. suis remains an incompletely understood pathogen in dogs with limited published data on its pathogenesis and clinical features. This case series describes the presentations, diagnosis, and clinical management of B. suis infection in three dogs: (1) a bitch with dystocia, abortion and mastitis; (2) an entire male dog with septic arthritis and presumptive osteomyelitis; and (3) a castrated male dog with lymphadenitis. Unique features of these cases are reported including the first documented detection of B. suis from milk and isolation from lymph nodes of canine patients, as well as the follow-up of pups born to a B. suis-infected bitch. Consistent with previous reports, all three dogs showed a favourable clinical response to combination antibiotic therapy with rifampicin and doxycycline. Individually tailored drug regimens were required based on the clinical presentation and other factors, including owner expectations and compliance with therapy as well as a zoonotic risk assessment (generally considered low, except around time of whelping). The cropping systems model APSIM (Agricultural Production Systems sIMulator) was applied to assess the response of sorghum grain yield to inorganic fertilizers applications and residue retention in diverse farmers’ management systems (homestead fields and bush farms). The model was parameterized using data collected from experiments under optimum growth conditions (limited water or nutrient stress). Independent data from field experiments with three levels of P and four levels of N fertilizers conducted at two different locations and soils were used to evaluate the model. Soil water and fertility parameters measured were used for simulations while same starting conditions were assumed for unmeasured parameters for all trials. APSIM predicted the grain yield response of sorghum to both N and P applications with an overall modified internal coefficient of efficiency of 0.64. Following model parameterization, a long-term simulation study was conducted using a stochastic weather data derived from historical weather data to assess the effects of crop residue management on grain production. A gradual decline in sorghum grain yield was simulated over the 30-year simulation period in both the homestead fields and the bush farms, with yields being much lower in the latter under farmers’ management practices. Half the amount of mineral N fertilizer used in the bush farms was needed in the homestead fields to produce the average grain yields produced on the bush farm with full fertilization, if crop residues were returned to the fields in the homestead. Year-to-year variability in grain yield was consistently higher with the removal of crop residues, irrespective of management systems. APSIM was responsive to both organic and inorganic fertilizer applications in the study area and also highlighted the essential role of crop residues and inorganic fertilizer in influencing the temporal sorghum grain production and hence the impact of farmers’ management practices on food security. This was evident in the rapid decline in soil organic carbon (SOC) accompanied by a decline in grain yield over the 30 years of cropping. The crude protein content and amino acid profile of seven feedstuffs (linseed meal, maize gluten meal, rapeseed meal, rapeseed meal protected, soybean meal, fullfat soybean extruded and sunflower meal) were determined before and after ruminal incubation for 16 h in three bulls with large rumen cannulas. The intestinal disappearance of amino acids was measured using mobile bag technique. Ruminal incubation affected amino acid profile of all experimental feedstuffs. Crude protein degradation varied from 29.3% for maize gluten meal to 86.4% for rapeseed meal. A tendency towards increased disappearance was observed for glutamic acid, histidine, lysine and proline and decreased disappearance for branched-chain amino acids. The intestinal crude protein digestibility was higher than > 80%, except rapeseed meal (66.4%) and sunflower meal (77.8%). The least digestible individual amino acids were methionine and isoleucine in rapeseed meal, histidine and methionine in rapeseed meal protected and arginine in sunflower meal. In general, the lowest amino acid digestibilities were found in feedstuffs with the highest fibre content. The feedstuffs show that they have different potential for supplying of limiting amino acids. Of particular value are the feedstuffs with low crude protein degradability in the rumen and high intestinal digestibility of amino acids. 相似文献

Mycobacterium avium subspecies paratuberculosis (MAP) is a facultative intracellular pathogen that resides in host macrophage cells. Presently, little is known about how MAP is able to subvert the normal bacteriocidal functions of infected macrophages. Previously, we reported that ileal tissues from MAP infected cattle contained high levels of interleukin-1 alpha (IL-1alpha) and tumor necrosis factor receptor-associated factor 1 (TRAF1), relative to ileal tissues from uninfected cattle. High-level expression of these two proteins could have profound effects on macrophage function, intracellular signaling, and apoptosis. We now demonstrate that high levels of TRAF1 protein are located primarily within macrophages infiltrating areas of MAP infection. We have also utilized cultured bovine monocyte-derived macrophage cells (MDM) either infected with live MAP or stimulated with recombinant IL-1alpha (rIL-1alpha) to determine if there is a relationship between IL-1alpha and TRAF1 expression. These studies have identified a dose dependent increase in TRAF1 protein levels in bovine MDM in response to infection with live MAP or following treatment with rIL-1alpha. Sustained TRAF1 protein expression was dependent upon interaction of rIL-1alpha with it's receptor and rIL-1beta was also able to enhance TRAF1 gene expression. Our results suggest that MAP may use the IL-1-TRAF1 system to enhance TRAF1 protein expression in infected bovine MDM. These novel results provide evidence for a new avenue of research on the effect of MAP and other intracellular pathogens on macrophage signaling and apoptosis. 相似文献

Forty-two samples were taken from the contents of the proximal small intestine of two lactating dairy cows fitted with re-entrant duodenal cannula. Most samples were free of detectable amylase activity. The (chymo)trypsinogen present was only partially activated to (chymo)trypsin. The activation was continued in vitro: slowly at the original pH of the samples (between pH 2.8 and 4.2), and faster after neutralization or a slight alkalinization. The effect of Ca, EDTA and soybean inhibitor on the activation of trypsinogen was also studied. The pancreatic enzymes were inactive in the acid pH range of the samples, but pepsin was markedly active. At pH 3.8 casein was digested rapidly by purified pepsin and slowly by the samples (agar-plate experiments). In model experiments performed with purified enzymes, pepsin digested (chymo)trypsin rapidly at pH 1-2 and slowly at pH 3.8. In the intestinal juice (chymo)trypsin and their zymogens seemed to be unaffected by pepsin under the conditions of the samples. It is concluded that the conditions prevailing in the duodenum/upper jejunum of the experimental cows account for a gastric-type digestion, despite the presence of pancreatic enzymes. In vivo the intestinal contents pass in distal direction. Meanwhile the pH of the chyme gradually increases and gives rise to first an increase of enterokinase activity accounting for a faster activation of the zymogens; second a start of function of activated pancreatic proteases and third a gradual decrease of pepsin activity and finally to its irreversible denaturation. NH3 volatilization from surface-applied urea, diammonium phosphate (DAP), and calcium ammonium nitrate (CAN) was measured with chambers through which air was drawn continuously. Two sandy soils and two sandy loam soils, which had been treated with and without time for the last 25 years, were used for the experiments. The accumulated NH3 loss from CAN applied to an unlimed sandy soil was linearly related to time. For the other treatments the accumulated loss was exponentially related to time. The NH3 loss was exponentially related to the maximum soil pH of the fertilizer-amended soil, and was inversely related to the content of exchangeable H+. Due to the low cation exchange capacity of these light-textured soils the NH3 loss was not reduced as the soil CEC increased. The maximum pH after soil amendment was related to soil pH. Therefore, a model is proposed that relates the NH3 loss solely to fertilizers and soil pH. The NH3 loss was less than 5% from CAN, about 20% from DAP, and about 30% from urea, with the insignificant loss from urea applied to the unlimed sandy soil excluded. Wind erosion and dust storms are common phenomena in the semi-arid Xilingele grassland (Inner Mongolia) and contribute considerably to matter balances. Dust emission and dust deposition affect particles of the silt and clay fraction and the organic matter predominantly, the carriers for nutrients in soils. Based on wind erosion measurements in spring 2005 and 2006 the organic carbon and total nitrogen contents of dust were analyzed to determine the contribution of wind erosion processes on C- and N-ratios to the grasslands. 相似文献

Forest management requires a profound understanding of how tree species affect C and N cycles in ecosystems. The large C and N stocks in forest soils complicate research on the effects of tree species on C and N pools. In‐situ 13C and 15N labeling in undisturbed, natural forests enable not only tracing of C and N fluxes, but also reveal insight into the interactions at the plant‐soil‐atmosphere interface. In‐situ dual 13C and 15N pulse labeling of 20 beeches (Fagus sylvatica L.) and 20 ashes (Fraxinus excelsior L.) allowed tracing the fate of assimilated C and N in trees and soils in an unmanaged forest system in the Hainich National Park (Germany). Leaf, stem, root, and soil samples as well as microbial biomass were analyzed to quantify the allocation of 13C and 15N for 60 d after labeling and along spatial gradients in the soil with increasing distance from the stem. For trees of similar heights (≈ 4 m), beech (20%) assimilated twice as much as ash (9%) of the applied 13CO2, but beech and ash incorporated similar 15N amounts (45%) into leaves. The photosynthates were transported belowground through the phloem more rapidly in beech than in ash. Ash preferentially accumulated 15N and 13C in the roots. In contrast, beech released more of this initially assimilated 13C (2.0% relative 13C allocation) and 15N (0.1% relative 15N allocation) via rhizodeposition into the soil than ash (0.2% relative 13C, 0.04% relative 15N allocation), which was also subsequently recovered in microbial biomass. These results on C and N partitioning contribute to an improved understanding of the effects of European beech and ash on the C and N cycles in deciduous broad‐leaved forest. Differences in C and N allocation patterns between ash and beech are one mechanism of niche differentiation in forests containing both species. 相似文献
