A positive score on a modified acid-fast (MAF)-stained smear test of fresh placenta was used to identify a group of bovine abortion submissions believed to be infected with Coxiella burnetii. Immunohistochemical (IHC) testing for Coxiella and Chlamydia antigens was performed on 14 MAF smear-positive cases as well as 29 MAF smear-negative cases received during the study period. Nine MAF smear-positive cases as well as 1 MAF smear-negative case were Coxiella-positive via the IHC test. No placentas were positive for Chlamydia antigen. Various histopathologic features were categorized for all placentas and the presence or absence of selected risk categories was also graded for each case. The results between Coxiella IHC-positive cases and Coxiella IHC-negative/MAF-negative cases were compared using Fisher's exact test (P value at 95% confidence). Significant associations were found between Coxiella IHC-positive cases and the presence of placental inflammation (P = 0.0027), placental necrosis (P = 0.012), fetal pneumonia (P = 0.0152), and the visibility of Coxiella-like organisms within trophoblasts on hematoxylin and eosin-stained sections (P < 0.0001). Histopathologic features of Coxiella IHC-positive placentas included infiltration of the chorionic stroma by mononuclear cells, necrosis of chorionic trophoblasts, and focal exudation of fibrin and neutrophils. The results indicate that MAF smears are a good screening tool for the presence of Coxiella in placentas from bovine abortion cases and that the detection of this pathogen in aborted placentas via traditional staining or IHC methods is usually associated with placentitis.  相似文献

The effects of dietary β‐hydroxy‐β‐methylbutyrate (HMB) supplementation during gestation on reproductive performance of sows and the mRNA expression of myogenic markers in skeletal muscle of neonatal pigs were determined. At day 35 of gestation, a total of 20 sows (Landrace × Yorkshire, at third parity) were randomly assigned to two groups, with each group receiving either a basal diet or the same diet supplemented with 4 g/day β‐hydroxy‐β‐methylbutyrate calcium (HMB‐Ca) until parturition. At parturition, the total and live litter size were not markedly different between treatments, however, the sows fed HMB diet had a decreased rate of stillborn piglets compared with the sows fed the control (CON) diets (p < 0.05). In addition, piglets from the sows fed HMB diet tended to have an increased birth weight (p = 0.08), and a reduced rate of low birth weight piglets (p = 0.05) compared with piglets from the CON sows. Nevertheless, lower feed intake during lactation was observed in the sows fed the HMB diet compared with those on the CON diet (p < 0.01). The relative weights of the longissimus dorsi (LD) and semitendinosus (ST) muscle were higher (p < 0.05) in neonatal pigs from the HMB than the CON sows. Furthermore, maternal HMB treatment increased the mRNA levels of the myogenic genes, including muscle regulatory factor‐4 (MRF4, p < 0.05), myogenic differentiation factor (MyoD) and insulin‐like growth factor‐1 (IGF‐1, p < 0.01). In conclusion, dietary HMB supplementation to sows at 4 g/day from day 35 of gestation to term significantly improves pregnancy outcomes and increases the expression of myogenic genes in skeletal muscle of neonatal piglets, but reduces feed intake of sows during lactation.  相似文献

Staphylococcus aureus mastitis is an important cause of economic loss for the dairy industry. Control programs rely on the timely and accurate identification of positive quarters. The effects of sampling time and sample handling were examined in an attempt to improve the accuracy of detection of S. aureus. Premilking and postmilking milk samples were collected from 55 lactating quarters with subclinical S. aureus infection. Each sample was divided into 2 aliquots; one of which was cultured fresh, the other was frozen at -20 degrees C for 14 days before being cultured. Analysis of variance was used to determine the effect of sampling time (premilking vs postmilking) and sample handling (fresh vs frozen) on the detection of S. aureus, as measured by the mean category for colony-forming units per millilitre (cfu/mL). A stratified analysis was required, due to interaction between sampling time and sample handling. Only a fresh postmilking sample was inferior, yielding a lower mean category for cfu/mL (P < 0.05). The ability to detect S. aureus in quarters with subclinical intramammary infection, as measured by the mean category of cfu/mL, was maximized in fresh or frozen premilking samples and in frozen postmilking samples.  相似文献

Plant activators provide an appealing management option for bacterial diseases of greenhouse-grown tomatoes. Two types of plant activators, one that induces systemic acquired resistance (SAR) and a second that activates induced systemic resistance (ISR), were evaluated for control of Pseudomonas syringae pv. tomato and effect on plant defense gene activation. Benzothiadiazole (BTH, SAR-inducing compound) effectively reduced bacterial speck incidence and severity, both alone and in combination with the ISR-inducing product. Application of BTH also led to elevated activation of salicylic acid and ethylene-mediated responses, based on real-time polymerase chain reaction analysis of marker gene expression levels. In contrast, the ISR-inducing product (made up of plant growth-promoting rhizobacteria) inconsistently modified defense gene expression and did not provide disease control to the same level as did BTH. No antagonism was observed by combining the two activators as control of bacterial speck was similar to or better than BTH alone.  相似文献

Revegetation of mine tailings sites can require significant amounts of topsoil, the sourcing of which can be costly and have detrimental impacts. To address this problem at an Irish mine tailings site, engineered soils were created by mixing varying rates of glacial till with stockpiled peat and compost. Soil status was assessed using a range of soil parameters and vegetation growth characteristics and compared with locally sourced topsoil. Hordeum vulgare (Barley) germination and growth trials were assessed on engineered soils: compost with glacial till, peat with glacial till, compost/ peat with glacial till and topsoil. A range of soil quality parameters were examined including: nutrient status, dehydrogenase activity, metals availability and physical characteristics (bulk/particle density and porosity). Results demonstrate that compost derived soils yielded superior plant biomass and nutrient content, whilst peat derived treatments exhibited nutrient deficiency. Whilst the engineered soils offer potential as an alternative to sourcing topsoil for covering mine tailings, the phosphorus and metal content of composts should be assessed prior to inclusion. Combined growing- and dormant-season pasture use has potential to increase herbage harvest without causing the undesirable shift in species composition that occurs with excessive utilization. The objective of this study was to determine the effect of summer clipping on winter pastures and winter clipping on summer pastures regarding standing crop, plant community composition, and forage quality. The study was conducted from 2003–2006 at the Antelope and Cottonwood Research Stations located in the mixed-grass prairie of western South Dakota. At each location, the experimental design was a randomized complete block with three replications that included 18 clipping treatments arranged as a split-split plot. Whole plots consisted of four summer clipping dates (May–August). Subplot treatments were two clipping intensities (clipped to residual height to achieve 25% or 50% utilization). Sub-subplots consisted of two winter clipping intensities (unharvested or clipped to a residual height to achieve a total utilization of 65%). Two winter control treatments were arranged in the subplot and split into two clipping intensities of 50% and 65% utilization. Winter biomass for the May 25% clipping treatment was similar to winter biomass for winter-only clipping. No increase in forage quality resulted from summer clipping compared with winter clipping. Three consecutive yr of combined growing-season and dormant-season defoliation to 65% utilization resulted in no change in functional group composition compared with ≤ 50% utilization treatments. Clipping in June resulted in reduced midgrass biomass at both stations and increased shortgrass biomass at Cottonwood. Results suggest that producers could combine growing and dormant-season grazing to increase the harvest of herbage on mixed-grass prairie, but should change season of use periodically to avoid an undesirable shift in plant composition.  相似文献

Background: Urine specific gravity (USG) is used clinically to estimate urine osmolality (UOsm). Although USG has been shown to have a linear correlation with UOsm in dogs, the relationship is altered when there are significant numbers of high molecular weight (MW) molecules in the urine.
Hypothesis: USG would no longer predict UOsm in dogs given intravenous hetastarch (670/0.75)(HES).
Animals: Eight healthy employee-owned adult dogs.
Methods: Prospective, controlled experimental study. USG and UOsm were measured every 30 minutes from t=0 minutes to t=360 minutes. Dogs were administered 20mL/kg of either NaCl 0.9% (control group, n=4) or HES (treatment group, n=8) IV over 1 hour starting at t=90 minutes.
Results: There was a decrease in UOsm in both groups starting at t=120 minutes and continuing for the study duration, and there was no significant difference in UOsm between treatment and control groups across all time points. There was an appropriate decrease in USG from t=120 minutes for the control group. In the treatment group, USG increased significantly at t=120 minutes ( P = .0006), t=150 minutes ( P = .0002), and t=180 minutes ( P = .0044). The largest increase in USG occurred at t=150 minutes with a mean USG of 1.070 ± 0.021 (range 1.038-1.104).
Conclusions and clinical importance: Urine specific gravity should not be used to estimate urine solute concentration in dogs following the administration of 20mL/kg of HES. In a clinical setting, the evaluation of USG following this dose of HES may lead to an overestimation of urine concentration.  相似文献

Cane cuttings of the grapevine rootstock Vitis rupestris Scheele x V. riparia Michx. cv. 3309 Couderc were brought out of endodormancy by warming at 30 degrees C. Cane pieces (12 to 13 cm long) with nodes containing a primary bud were placed in a gas exchange system and monitored for net respiratory fluxes of CO2 and O2. Grapevine respiration rates expressed on a wood volume basis were 1.4 to 3.4 mmol CO2 or O2 m-3s-1, which is higher than stem respiration rates reported for many other woody taxa but similar to rates measured for ecodormant buds of other Vitis species. Passive water loss from canes was 0.7 to 1.2 mmol H2O m-3s-1. During a 7-day period, nonstructural carbohydrate concentrations in cane wood declined only slightly, whereas sucrose was nearly completely consumed. When ambient CO2 concentration ([CO2]) was raised from 300 to 750 micro molmol-1 and then 2000 micromol mol-1, net CO2 exchange rates declined by 5.9 +/- 0.6 and then 11.0 +/- 0.6%, whereas net O2 consumption rates remained about constant. The mean respiration quotient (net CO2/O2 flux) for canes with intact ecodormant buds was 0.99 +/- 0.03 when the [CO2] was 300 micromol mol-1, and decreased to 0.87 +/- 0.03 and 0.088 +/- 0.02 when the [CO2] was increased to 750 and 2000 micromol mol-1, respectively. The results support the hypothesis that, in Vitis canes, inhibition of respiratory CO2 efflux in response to high [CO2] is an indirect consequence of non-photosynthetic carboxylation reactions, and not a result of inhibition of respiratory metabolism.  相似文献
