Suspensions of mixed rumen bacteria (B), protozoa (P), and mixed rumen microorganisms (BP) prepared from rumen contents of fistulated goats were anaerobically incubated with 1 mM p‐hydroxyphenylacetic acid (HPA) at 39°C for 24 h. Tyrosine (Tyr), phenylalanine (Phe), tryptophan (Trp) and other related compounds in both supernatants and hydrolyzates of microbial cells in all incubations were analyzed by HPLC. Large amounts of Tyr (32.1, 42.7 and 36.1% of disappeared HPA in B, P and BP, respectively) were produced from HPA during a 12 h incubation period. The formation of Tyr in P (178.6 µmol/g MN) was 1.5 and 2 times higher than in B and BP, respectively. Phe (7–11% of the disappeared HPA) and Trp (3–6% of the disappeared HPA) were also synthesized from HPA in B, P, and BP. Phe synthesis in P (46.3 µmol/g MN) was 1.7 times higher than in B but, in contrast, Trp synthesis in B, was 1.6 times higher than in P. The metabolites p‐hydroxyphenylpyruvic acid (in the range of 5–14% of disappeared HPA), phenylacetic acid (1–11%), p‐hydroxybenzoic acid (3–7%) and benzoic acid (1–6%) were produced from HPA in B, P and BP. This paper describes a new in-cell method for pursuing accelerated solvent extraction (ASE) prior to lipid analysis from food samples. It is difficult to pursue direct ASE with acid- or base-hydrolyzed samples due to the corrosive nature of the reagents and material limitations. In this study ion exchange based materials were used to remove acid or base reagents in-cell without compromising the recovery of lipids. The performance data are presented here for the new methods for lipid extraction for a variety of food samples and compared to the Mojonnier method. NIST Standard Reference Materials (SRM-1546 and SRM-1849) were used to validate the ASE methods. Excellent fat recoveries were obtained for the ASE methods. The new methods presented here enhance the utility of ASE and eliminate labor intensive protocols. 相似文献

Shoot:root (S:R) response in four winter C3-grasses (cereals) viz. wheat (Triticum aestivum L.), rye (Secale cereale L.), barley (Hordeum vulgare L.) and oats (Avena sativa L.) was investigated under eight different NPK sources (S1 = 20-20-20, S2 = 20-27-5, S3 = 7-22-8, S4 = 10-10-10-20S, S5 = 11-15-11, S6 = 31-11-11, S7 = 24-8-16, and S8 = 19-6-12) in pot experiment at Dryland Agriculture Institute, West Texas A&M University, Canyon, Texas, USA during winter 2009–2010. The experiment was performed in completely randomized design (CRD) with three repeats. The objective of this experiment was to investigate whether the S:R of winter cereals differ or not when applied with different NPK sources. Considerable variations in the shoot: root was noticed in the four grasses at different growth stages, i.e., 30, 60 and 90 days after emergence (DAE) when applied with different NPK fertilizers sources. Wheat had the highest S:R of 3.4 at 30 days after emergence (DAE), barley at 60 DAE (6.5), while oats at 90 DAE (3.9). Among the NPK sources, crops had the highest S:R of 3.6, 6.2 and 3.7 when applied with S3, S2, and S6 at 30, 60 and 90 DAE, respectively. Shoot to root ratio showed positive relationship with increase in shoot weight and negative relationship with increase in root weight. The NPK fertilizer S6 (31: 11: 11), an acid loving fertilizer had negative effects on the shoot and root development of cool season cereals, but among these cereals under study, barley and oats roots were affected more than wheat and rye. It was concluded from this present study, that the four cool season cereals responded differently in terms of shoot: root to different NPK fertilizers at different growth stages. 相似文献

Abstract Apparent digestibility coefficients were determined for six feedstuffs, subsequent to their processing into pelleted feed for tropical freshwater catfish, Mystus nemurus (Cuvier & Valenciennes), using a reference diet and test diets composed of 70% reference diet and 30% test ingredient. It was found that most ingredients were fairly well digested except maize and chicken viscera. Digestibility coefficient for protein (P), dry matter (DM) and energy (E) were: (1) fish meal: 97·8% P,97% DM. 77·88% E: (2) soybean meal: 86% P,95·55% DM, 67·89% E: (3) rice bran: 81% P,85·81% DM, 67·93% E: (4) copra meal: 79·90% P, 86% DM, 71·89% E: (5) maize: 51·93% P, 49·0% DM, 63·40% E and (6) chicken viscera: 37% P,29% DM, 52% E. Digestibility coefficients for feedstuffs indicated that animal protein product, such as fish meal and grain by-product, such as rice bran were more digestible than starchy and fibrous feed, e.g. maize. Chicken viscera was poorly digested largely due to inefficient processing. As most feedstuffs were well digested, ingredient selection should, therefore, be based primarily on their protein content, amino acid composition and palatability. 相似文献

Egg yolk is routinely used as a cryoprotectant in semen extenders. However, it may contain cryoprotective antagonists, and there are hygienic risks associated with its use. Proteins of plant origin, like soya-lecithin, lack these hazards. The aim of this study was to use soya-lecithin as a cryoprotectant in extender and to investigate its effects on in vitro quality and in vivo fertility of buffalo semen. Semen from three buffalo bulls was frozen in tris-citric extender containing 5.0%, 10% or 15% soya-lecithin or 20% egg yolk. Sperm motility, plasma membrane integrity and viability were assessed post-dilution, pre-freezing and post-thaw. In Post-dilution and pre-freezing, the values for motility, plasma membrane integrity and viability remained higher (p ≤ 0.05) in extenders containing 10% soya-lecithin and control compared with extender containing 5% and 15% soya-lecithin. However, motility, plasma membrane integrity and viability were higher (p < 0.05) in extender containing 10% soya-lecithin compared with control and extenders containing 5% and 15% soya-lecithin. Semen from two buffalo bulls was frozen in tris-citric extender containing either 10% soya-lecithin or 20% egg yolk. Higher (p < 0.05) fertility rate was recorded in buffaloes inseminated with semen containing 10% soya-lecithin (56%) compared with 20% egg yolk (41.5%). The results suggest that 10% soya-lecithin in extender improves the freezability and fertility of buffalo bull spermatozoa and can be used as an alternate to egg yolk in cryopreservation of buffalo semen. 相似文献

This research study evaluated the effect of different additives on the bait consumption by Indian crested porcupine, a serious forest and agricultural pest, under field conditions. Different additives (saccharin, common salt, bone meal, fish meal, peanut butter, egg yolk, egg shell powder, yeast powder, mineral oil and coconut oil) at 2 and 5% each were tested for their relative preference, using groundnut–maize (1:1) as basic bait. All the additives were tested under a no‐choice test pattern. For control tests, no additive was mixed with the basic bait. Saccharin at 5% concentration significantly enhanced the consumption of bait over the basic bait, while 2% saccharin supplemented bait resulted in a non‐significant bait consumption. All other additives did not enhance the consumption of the bait material; rather, these worked as repellents. However, the repellency was lowest with the common salt, followed by egg yolk, egg shell powder, bone meal, peanut butter, mineral oil, fish meal and yeast powder, while coconut remained the most repellent compound. The present study suggested that groundnut–maize (1:1) supplemented with 5% saccharin was the preferred bait combination, and can be used with different rodenticides for the management of Indian crested porcupine. 相似文献

Kalam Kohistan is a dry,temperate hilly region in the Hindu Raj Series of the Greater Hindu Kush Mountains with diverse forests.As plant distribution and composition is expression of a range of various environmental variables,ecological and floristic attributes of vegetation in the Laikot Forests were therefore evaluated via consecutive explorative trips.These forests have not been evaluated ecologically before due to harsh climatic conditions and issues of accessibility and the study in hand is one of the first in its nature.Quantitative ecological techniques were used to sample forest vegetation and identify species in quadrats of 2 m×2 m for herbs,5 m×5 m for shrubs and 10 m×10 m for trees.All the data of 195 plant species and environmental factors were analyzed via Two-way Cluster Analysis and Canonical Correspondence Analysis using PCORD and CANOCO software.Among 195 plant species from 63 families were 27 species of Asteraceae,the most-speciose family,followed by Poaceae(20 spp.).The dominant life form was therophyte(86 spp.;44%share),followed by phanerophyte(41 spp.;21%share).For the leaf size spectrum,most prevalent was nanophyll(73 spp.;44%)followed by microphyll(66 spp.,34%)and mesophyll(44 spp.;23%).Calcium carbonate,pH,potassium,sand,silt,organic matter,slope aspect and grazing significantly influenced species composition,distribution and habitat.The floristic variation in the region was diverse due to elevational and aspect gradients and thus devided into three zones.This vegetation zonation can be utilized for forest management,species and habitat conservation.Both in situ and ex situ conservation of threatened plant species may improve their conservation status in future if address properly. 相似文献
