Two new briarane-type diterpenoids, briarenolides K (1) and L (2), were isolated from an octocoral identified as Briareum sp. The structures of new briaranes 1 and 2 were elucidated by spectroscopic methods. Eight new cembranoids, crassarines A-H (1-8) were isolated from the Formosan soft coral Sinularia crassa. Compounds 1-3 represent the rare cembranoids with a 1,12-oxa-bridged tetrahydrofuran ring, while 4 and 5 are the firstly discovered 1,11-oxa-bridged tetrahydropyranocembranoids. The absolute configuration of 6 was determined using the Mosher's method. Compounds 6 and 8 were found to significantly inhibit the expression of both pro-inflammatory iNOS and COX-2 proteins at 10 μM, respectively, while compounds 4-8 were found to be non-cytotoxic toward the selected human liver cancer cells. 相似文献

Five new nardosinane-type sesquiterpenoids, paralemnolins Q-U (1-5), along with three known compounds (6-8), were isolated from the Formosan soft coral Paralemnalia thyrsoides. The structures of new metabolites were elucidated on the basis of extensive spectroscopic methods, and the absolute configuration of 1 was determined by the application of Mosher's method on 1. Among these metabolites, 1 and 3 are rarely found nardosinane-type sesquiterpenoids, possessing novel polycyclic structures. Melatonin, the major secretory product of the pineal gland, scavenges a variety of reactive oxygen and nitrogen species in vivo and in vitro, indicating that melatonin is a potent function as an antioxidant. The objective of this study was to investigate the effect of melatonin in the presence or absence of hydrogen peroxide (H2O2) on sperm characteristics (motility, viability, survival rate, membrane integrity, lipid peroxidation (LPO) and mitochondria activity) and also to examine the developmental rates to the blastocysts stage of porcine oocytes fertilized in vitro with semen treated with or without melatonin (100 nm ) in the presence or absence of H2O2 (250 μm ). The sperm were treated with melatonin in the presence or absence of H2O2 for 3, 6, 9 and 12 h at 37°C and then analysed for the sperm characteristics. The porcine embryos were produced by in vitro maturation and in vitro fertilization (IVM/IVF) using semen treated with or without melatonin (100 nm ) in the presence or absence of H2O2 (250 μm ) for 6 h. The semen characteristics, including motility, viability, survival rate, membrane integrity and mitochondria activity, were higher in the groups that were treated with melatonin in comparison to other groups, irrespective of incubation periods. Malondialdehyde levels in control, melatonin and melatonin + H2O2 groups were lower than H2O2 only group. A positive correlation was shown among motility, viability, survival rate and membrane integrity, but a negative correlation was observed between LPO and the other evaluation methods. The developmental rates to blastocysts of IVM/IVF porcine oocytes fertilized by semen treated with melatonin were significantly increased compared with any other groups, with the cell number of blastocysts shown to have a similar trend to the developmental rates. These results demonstrate that melatonin can improve the semen characteristics during in vitro storage and support the developmental ability of IVM/IVF embryos in pigs. 相似文献

The aim of this study was to systematically examine the inhibitory mechanisms of C-phycocyanin (C-PC), one of the major phycobiliproteins of Spirulina platensis (a blue-green alga), in platelet activation. In this study, C-PC concentration-dependently (0.5-10 nM) inhibited platelet aggregation stimulated by agonists. C-PC (4 and 8 nM) inhibited intracellular Ca2+ mobilization and thromboxane A2 formation but not phosphoinositide breakdown stimulated by collagen (1 microg/mL) in human platelets. In addition, C-PC (4 and 8 nM) markedly increased levels of cyclic GMP and cyclic GMP-induced vasodilator-stimulated phosphoprotein (VASP) Ser(157) phosphorylation. Rapid phosphorylation of a platelet protein of Mw 47,000 (P47), a marker of protein kinase C activation, was triggered by phorbol-12,13-dibutyrate (150 nM). This phosphorylation was markedly inhibited by C-PC (4 and 8 nM). In addition, C-PC (4 and 8 nM) markedly reduced the electron spin resonance (ESR) signal intensity of hydroxyl radicals in collagen (1 microg/mL)-activated platelets. The present study reports on a novel and very potent (in nanomolar concentrations) antiplatelet agent, C-PC, which is involved in the following inhibitory pathways: (1) C-phycocyanin increases cyclic GMP/VASP Ser157 phosphorylation and subsequently inhibits protein kinase C activity, resulting in inhibition of both P47 phosphorylation and intracellular Ca2+ mobilization, and (2) C-PC may inhibit free radicals (such as hydroxyl radicals) released from activated platelets, which ultimately inhibits platelet aggregation. These results strongly indicate that C-PC appears to represent a novel and potential antiplatelet agent for treatment of arterial thromboembolism. 相似文献

Betanin is an antioxidant pigment found in djulis, a grain native to Taiwan, and is a natural source food coloring, but the bright red color degrades rapidly if submitted to light, heat, or oxygen. The effects of nanogrinding on the stability of pigments and bioactive components are unknown. In this study, the color characteristics and bioactivity (antioxidant capacity and enzyme activity) of nanoparticle (NP) djulis was compared with those of intact granules (IG) and microparticles (MP). Results showed that the NP samples exhibited the highest betanin content (2.04 mg/g), which was almost twice that of IG. It was observed that nanogrinding resulted in higher pigment extraction efficiency. However, during storage (5-35 °C for 56 days), NP samples showed the most serious pigment degradation, and this color degradation, as expected, had the lowest activation energy. This was more evident when the storage temperature was high. Antioxidant capacities showed the same trends. MP and NP exhibited significantly higher activity of superoxide dismutaste-like activity, lactoperoxidase (LPO), and lysozyme than IG. Gel permeation chromatography confirmed the degradation of larger particles during nanogrinding, which might favor enzyme extraction and their activities. Effects of the 921 earthquake on the water quality in the upper stream at the Guandaushi experimental forest 总被引:1,自引:0,他引:1
A powerful 7.3 magnitude earthquake struck Taiwan on September 21, 1999. The stream water chemistry (pH, total alkalinity, conductivity, sodium, potassium, calcium, magnesium, ammonium, fluoride, chloride, sulfate, and nitrate) has been monitored since 1995 at the Guandaushi forestry riparian zone in central Taiwan. Collected data was used as a basis for comparing pre- and post-earthquake impacts. The pH, conductivity, and concentrations of Na, Ca, Mg, SO4, and HCO3 in stream water were lowest during the summer season, when stream water discharge was highest. On the other hand, the lowest concentrations of Cl, NH4, and NO3 in stream water occurred during the winter season, when stream water discharge was lowest. Also, K and F showed very little seasonal fluctuation in concentration. Downward trends in K and Ca were found 14 months prior to the earthquake; although, an upward trend occurred in NH4 at the same time. 相似文献
