This study assessed the accuracy of the commercial BioPRYN® ELISA for the detection of pregnancy‐specific protein‐B (PSPB) using a single blood sample to determine pregnancy status in American bison (Bison bison). A total of 49 bison cows were used in the study, and sampled at two time‐points during the gestation period, fall and spring, correlating with early‐ to mid‐term gestation (average 62.9 days post‐mating) and mid‐ to late‐term gestation (average 229.2 days post‐mating), respectively. Sensitivity of the test during early‐ to mid‐term gestation sampling period (fall) was 87.1%, while specificity was 100%; sensitivity of the test during late‐term gestation sampling period (spring) was 96.3%, while specificity remained at 100%. In total, the test showed a total sensitivity of 91.4%, specificity of 100% and total accuracy of 93.8%, similar to domestic cattle. Use of the single‐sample BioPRYN® ELISA in American Bison for pregnancy diagnosis is economical and practical, minimizing animal handling time, frequency and subsequent stress while providing accurate results for pregnancy diagnosis at 62 days post‐mating. This method should be considered over more traditional pregnancy diagnosis methods for use in managed bison herds.  相似文献

Several remote observations have indicated that water ice may be presented in permanently shadowed craters of the Moon. The Lunar Crater Observation and Sensing Satellite (LCROSS) mission was designed to provide direct evidence. On 9 October 2009, a spent Centaur rocket struck the persistently shadowed region within the lunar south pole crater Cabeus, ejecting debris, dust, and vapor. This material was observed by a second "shepherding" spacecraft, which carried nine instruments, including cameras, spectrometers, and a radiometer. Near-infrared absorbance attributed to water vapor and ice and ultraviolet emissions attributable to hydroxyl radicals support the presence of water in the debris. The maximum total water vapor and water ice within the instrument field of view was 155 ± 12 kilograms. Given the estimated total excavated mass of regolith that reached sunlight, and hence was observable, the concentration of water ice in the regolith at the LCROSS impact site is estimated to be 5.6 ± 2.9% by mass. In addition to water, spectral bands of a number of other volatile compounds were observed, including light hydrocarbons, sulfur-bearing species, and carbon dioxide.  相似文献

Understanding animal responses to landscape elements helps forecast population reactions to changing landscape conditions. The challenge is that some behaviors are poorly known and difficult to estimate. We assessed how uncertainty in behavioral responses to dense woods, an avoided landscape structure, impacts functional connectivity among reproductive habitat patches for Fender’s blue butterfly, an endangered prairie species of western Oregon, USA. We designed a factorial simulation experiment using a spatially explicit individual-based model to project functional connectivity for female butterflies across current and alternative landscapes. We varied the probability of dense woods entry and turning angle standard deviation for movements within the dense woods over a range of biologically reasonable and observed values. Butterflies in the current landscape (46 % dense woods) and one with prairie encroached by forest (60 % dense woods) showed reductions in functional connectivity estimates consistent with the expectations of habitat fragmentation. Although dense woods entrance uncertainty impacted functional connectivity projections, uncertainty in the dense woods turning angle standard deviation had comparatively little impact on connectivity estimates. Reduction and reconfiguration of the current dense woods to 27 % cover (restored landscape) appeared to facilitate a corridor behavior in dispersing individuals, likely providing a functional connectivity estimate comparable to the historic landscape (<5 % dense woods). Our simulations suggest that additional study of butterfly movement within the dense woods is unnecessary and that a partial reduction in dense woods would be sufficient to achieve historic levels of functional connectivity for Fender’s blue across the study landscape.  相似文献

Under rainfed agriculture without supplementary supply, crop failure due to erratic rainfall has become a common phenomenon in many regions of Ethiopia. Spate irrigation development with storage provision at the 5,000 ha Boru Dodota Spate Irrigation Scheme is one of the initiatives to move away from exclusively rain-dependent agriculture. This initiative has faced several challenges. Lack of design experience and failure to fully grasp the unpredictable nature of the spate flow caused the Boru Dodota Spate Irrigation Scheme to be implemented without considering the necessity of storage ponds. In addition, absence of a systematic approach to assist planners has resulted in non-optimal design of ponds’ capacity and operation. The study, on which this article is based, aimed at optimum storage operation to irrigate the maximum possible area with the existing ponds’ capacity and available water resource. During the study, the surface storage and irrigation scheme planning model was developed and used to analyze several pond operation scenarios in Boru Dodota Spate Irrigation Scheme. The main findings were as follows: (1) Supplementing the rainfall with the operation of the existing 19 ponds that enable weekly irrigation frequency results in irrigating at least 6,600 ha. (2) An increase in the number of ponds does not always guarantee an increase in the size of irrigated land because the water resources, the operation, and management are defining factors. (3) It is not economical to only rely on spate flow for irrigation as even with 200 ponds, a maximum of 1,250 ha could be irrigated.  相似文献

The aim of the present study was to evaluate the effects of different medium replacement intervals on the viability, antral cavity formation, growth and in vitro maturation (IVM) of oocytes from caprine and ovine pre‐antral follicles. Pre‐antral ovarian follicles (≥150 μm) were isolated from the ovarian cortex of goats and sheep and were individually cultured for 24 days using two different medium replacement intervals [2 days (T1) or 6 days (T2)]. Follicle development was evaluated on the basis of antral cavity formation, increases in follicular diameter and the presence of healthy cumulus oocyte complexes and fully grown oocytes. For caprine species, results showed a higher percentage (p < 0.05) of viable follicles in T1 than T2 from day 6 until the end of the culture. In addition, when comparing both treatments after the same culture duration, the rate of antrum formation was significantly higher in T1 than in T2 from day 12 onwards. Yet, in ovines, when both treatments were compared on day 24 of the culture, there were more viable follicles in T2 than in T1 (p < 0.05). In the caprine species, percentages of fully grown oocytes (≥110 μm) acceptable for IVM after 24 days of culture were significantly higher in normal follicles cultured in T1 (30.0%) than in T2 (6.7%; p < 0.05). On the other hand, in ovines, at the end of the culture, the percentage of oocytes destined for IVM was higher in T2 than in T1 (23.5% vs 2.9%; p < 0.05). In conclusion, under the same conditions, the frequency of medium replacement significantly affected the in vitro development of caprine and ovine pre‐antral follicles. To improve the efficiency of the culture system, the medium must be replaced every 2 and 6 days for goat and sheep pre‐antral follicles, respectively.  相似文献
