AIMS: To evaluate a multivalent leptospiral and clostridial vaccine for prevention of renal colonisation and urinary shedding in sheep, following experimental challenge with New Zealand strains of Leptospira borgpetersenii serovar Hardjo type Hardjobovis and L. interrogans serovar Pomona.

METHODS: Two separate but similarly designed studies were conducted. In both studies, Romney-cross lambs, aged 9–11 weeks, were randomly allocated to a vaccinated group and a control group. Vaccinated lambs each received two 1.5-mL S/C doses of a multivalent leptospiral and clostridial vaccine, 4 weeks apart, and animals in the control groups received the same dose of saline. Groups of 12 vaccinated and 12 control lambs were randomly selected in each study for challenge with serovars Hardjo or Pomona. Challenge was initiated 16 weeks following the second vaccination with three daily doses of live leptospires by intranasal and conjunctival routes. Following challenge, urine samples were collected weekly for 6 weeks, for dark field microscopy and leptospiral culture; 6 weeks after challenge the lambs were slaughtered and kidneys collected for leptospiral culture.

RESULTS: In lambs challenged with serovar Hardjo, 8/12 unvaccinated lambs had ≥1 urine or kidney sample that was positive for leptospires following culture, compared with 0/12 lambs in the vaccinated group (p=0.001). In lambs challenged with serovar Pomona, 9/12 unvaccinated lambs had ≥1 urine or kidney sample that was positive following culture, compared with 0/12 lambs in the vaccinated group (p<0.001). Prevention of renal colonisation and urinary shedding, expressed as the prevented fraction, was 100 (95% CI=61.7–100)% and 100 (95% CI=68.3–100)% against challenge with serovars Hardjo and Pomona, respectively, at 4 months after vaccination.

CONCLUSIONS AND CLINICAL RELEVANCE: Use of a multivalent leptospiral and clostridial vaccine demonstrated protection against challenge from New Zealand strains of serovars of Hardjo and Pomona 4 months after vaccination in lambs first vaccinated at 9–11 weeks of age. Further studies are required to assess the duration of immunity against challenge in sheep.  相似文献

A reduction of arbuscules in roots of grapevines (Vitis vinifera) observed when ring nematodes were added to field microplots led to the hypothesis that nematode feeding suppresses arbuscules by competing for root carbohydrates. Support for this hypothesis was tested by growing ‘Pinot noir’ grapevines in a factorial experiment with three levels of initial nematode densities (0, 0.1, 1.0 nematodes g?1 soil), two levels of light (full sun, 50% sun), and two levels of AMF (nonAMF, +AMF). Effects on plant growth were primarily driven by a light and AMF treatment interaction, such that low light increased stem dry matter accumulation at the expense of roots in +AMF vines only. Nematodes had only a minor influence on plant growth (leaf mass was reduced at the highest nematode density), but nematodes did not affect overall plant dry matter accumulation. Since nonAMF vines were severely limited by P and their growth was so poor, the impact of nematode and light treatments was further analyzed in +AMF plants only. Nematode populations, AMF colonization, and root carbohydrates were differentially affected by initial nematode density or light levels. Root biomass, and reducing sugar and starch concentrations in fine roots were reduced by low light, but the final nematode populations and arbuscule frequencies in roots were unaffected by light. Nematodes reduced arbuscules and starch concentrations in fine roots, but did not affect total colonization by AMF (hyphae, vesicles or arbuscules). Nematodes reduced plant P and K uptake at the highest density, and low light reduced Mg uptake. These findings are consistent with the hypothesis that ring nematodes suppress arbuscules in roots via competition for root carbohydrates. However, the lack of a treatment interaction between light and nematodes in our study suggests that ring nematode–AMF interactions in grape roots are controlled by more than competition for photosynthate.  相似文献

Chemical reactivity is conventionally understood in broad terms of kinetic versus thermodynamic control, wherein the decisive factor is the lowest activation barrier among the various reaction paths or the lowest free energy of the final products, respectively. We demonstrate that quantum-mechanical tunneling can supersede traditional kinetic control and direct a reaction exclusively to a product whose reaction path has a higher barrier. Specifically, we prepared methylhydroxycarbene (H(3)C-C-OH) via vacuum pyrolysis of pyruvic acid at about 1200 kelvin (K), followed by argon matrix trapping at 11 K. The previously elusive carbene, characterized by ultraviolet and infrared spectroscopy as well as exacting quantum-mechanical computations, undergoes a facile [1,2]hydrogen shift to acetaldehyde via tunneling under a barrier of 28.0 kilocalories per mole (kcal mol(-1)), with a half-life of around 1 hour. The analogous isomerization to vinyl alcohol has a substantially lower barrier of 22.6 kcal mol(-1) but is precluded at low temperature by the greater width of the potential energy profile for tunneling.  相似文献

Arbuscular mycorrhizal fungi (AMF) may exert profound influences on ecosystem resilience and invasion resistance in rangelands. Maintenance of plant community structure through ecological feedback mechanisms such as facilitation of nutrient cycling and uptake by host plants, physical and chemical contributions to soil structural stability, and mediation of plant competition suggest AMF may be important facilitators in stressful arid environments. Plant-AMF interactions could influence succession by increasing native plant community resilience to drought, grazing, and fire and resistance to exotic plant invasion. However, invasive exotic plants may benefit from associations with, as well as alter, native AMF communities. Furthermore, questions remain on the role of AMF in stressful environments, specifically the mycorrhizal dependency of sagebrush (Artemisia spp.) steppe plant species. Here, we review scientific literature relevant to AMF in rangelands, with specific focus on impacts of land management, disturbance, and invasion on AMF communities in sagebrush steppe. We highlight the nature of AMF ecology as it relates to rangelands and discuss the methods used to measure mycorrhizal responsiveness. Our review found compelling evidence that AMF mediation of resilience to disturbance and resistance to invasion varies with plant and fungal community composition, including plant mycorrhizal host status, plant functional guild, and physiological adaptations to disturbance in both plants and fungi. We conclude by outlining a framework to advance knowledge of AMF in rangeland invasion ecology. Understanding the role of AMF in semiarid sagebrush steppe ecosystems will likely require multiple study approaches due to the highly variable nature of plant-AMF interactions, the complex mechanisms of resilience conference, and the unknown thresholds for responses to environmental stressors. This may require shifting away from the plant biomass paradigm of assessing mycorrhizal benefits in order to obtain a more holistic view of plant dependency on AMF, or lack thereof, in sagebrush steppe and other semiarid ecosystems.  相似文献

Nutrient composition and yield of soybean [Glycine max (L.) Merr] seeds are heritable traits affected by environmental factors. This study determined the effects of arbuscular‐mycorrhizal (AM) fungi on seed protein, lipid, and phosphorus (P) composition and yield in soybean grown under a high nitrogen (N) regime. Plants were grown in pot cultures without AM fungi in P‐fertilized (+P) or unfertilized (‐P) soil, or in ‐P soil inoculated with one of the AM fungi Glomus mosseae (Nicol. & Gerd.) Gerd, and Trappe (Gm), Glomus etunicatum Becker and Gerd.(Ge), or Gigaspora rosea Nicol. and Schenck (Gr). Seed yields of+AM plants, as a group, were halfway between those of the +P and ‐P plants. Seed size was highest in Gm plants. Differences in protein concentrations between Ge and Gr and the other treatments were highly significant. Seed P and protein concentrations were not significantly correlated (p=0.162), but a highly significant (r =‐0.949) negative correlation between seed P and lipid concentrations was observed. Phosphorus concentration was highest and that of lipids lowest in +AM plants. Seed yield and nutrient composition were independent of the intensity of root colonization. The seed protein/lipid ratio was highly correlated with seed P concentration and was significantly higher for +AM plants, as a group, than for both +P and ‐P ‐AM plants. Differences in seed dry weight, size, seed/ stem ratio, P content, and protein concentration among +AM plants showed mycorrhiza‐specific host responses. These responses suggest that AM fungi can modify soybean seed development and chemical composition.  相似文献

Abstract

AIM: To determine whether viable Mycobacterium avium subsp. paratuberculosis (Map) is present in skeletal muscle and blood in ewes with and without Johne's disease confirmed histologically.

METHODS: A total of 51 mixed-aged ewes in poor body condition from a farm with a history of clinical Johne's disease were culled and examined at necropsy. BACTEC radiometric culture was performed on samples of skeletal muscle from the biceps femoris, mononuclear cells in peripheral blood (hereafter referred to as blood), and ileum. Histological sections and Ziehl-Neelsen (ZN)-stained impression smears of terminal ileum and mesenteric lymph nodes were examined. Ewes were defined as having confirmed Johne's disease if there was histopathological evidence typical of the disease within the ileum and adjacent lymph nodes.

RESULTS: Eighteen of 21 (86%) ewes with confirmed clinical Johne's disease were culture-positive for Map from sites peripheral to the alimentary tract, comprising 15 from skeletal muscle and 13 from blood. Five of 30 (17%) ewes that did not have Johne's disease were culture-positive, with four from skeletal muscle and one from blood. The likelihood that ewes with confirmed Johne's disease had systemic Map infection compared with ewes without was determined as OR=30 (95% CI=6.3–142.0; p<0.001).

CONCLUSION: The prevalence of Map infection of skeletal muscle and blood in ewes with confirmed Johne's disease was 71% and 62% respectively, and in unaffected ewes was 13% for muscle and 3% for blood.

CLINICAL RELEVANCE: Skeletal muscle and blood are potential sources of exposure of humans to Map, and the risk appears higher from sheep with Johne's disease.  相似文献

AIMS: To determine the effects of liveweight and condition score of ewes at mating, and shearing mid-pregnancy, on birthweights and growth rates of twin lambs from birth to the time of weaning, and; to determine whether any effect of condition score is independent of liveweight.

METHODS: Romney ewes (n=435) which conceived within a 17-day mating period and successfully gave birth to and reared twin lambs to weaning were used. Ewes were managed during pregnancy so that their total liveweight increased at a level equivalent to the expected growth of the conceptuses. Approximately half of the ewes were shorn 70 days after the mid-point of mating, and the remainder were left unshorn, at which time they had 6-months' wool growth. All lambs were weighed within 12 h of birth and again at weaning.

RESULTS: There was a significant linear relationship (R2=0.99) between mean group liveweight and condition score of ewes at mating (difference of 7.3 kg per 1.0 unit condition score). Liveweight of ewes at mating had small but significant (p<0.05) positive effects on both birthweights and growth rates of twin lambs to weaning. Ewes that had condition scores of 3.5–4.0 (scale 0–5) at mating gave birth to significantly (p<0.05) lighter lambs than ewes that had condition scores <3.5. The condition score of ewes at mating was positively associated with growth rates of lambs, although this effect was largely explained by variation in the liveweight of ewes at mating.

Twin lambs born to ewes shorn mid-pregnancy were significantly (p<0.05) heavier at birth (by 0.4 kg) and at weaning (p<0.05) than those born to unshorn ewes. The heavier weaning weights of lambs born to ewes shorn mid-pregnancy were due to a heavier birthweights rather than increased growth rate.

CONCLUSIONS: Liveweight and condition score of ewes at mating had small positive effects on growth rates to weaning of twin lambs. Shearing mid-pregnancy increased birthweights and weaning weights of twin lambs. To achieve high weaning weights of twin lambs, farmers should ensure ewes attain high liveweights and condition scores at mating and shear mid-pregnancy. It is speculated that the effects of liveweight and condition score of ewes at mating on weaning weights of twin lambs would be greater than those observed in the present study under conditions in which nutrition of the dam is below optimum.  相似文献

Three greenhouse experiments were carried out to determine the growth, yield, nitrate, total N and S concentration in shoots, and water uptake of hydroponically grown Brassica rapa L. subsp. nipposinica var. chinoleifera and Brassica juncea L. In each experiment, daily photosynthetically active radiation (PAR) level was 5.0 mol m−2 (low), 6.8 mol m−2 (medium) or 9.0 mol m−2 (high). Plants were supplied with nutrient solutions having equal N concentrations of 11 mM in different forms: 100% NH4, 50% NH4 + 50% NO3, and 100% NO3. Nitrogen supplied as 100% NH4 reduced fresh and dry shoot biomass, leaf area, and leaf number in both Brassica species, especially at low and medium PAR levels. In both Brassica species, S concentrations were highest, while nitrate concentrations were lowest in leaves of plants grown at N supplied as 100% NH4. No differences in leaf nitrate concentrations were observed between 50% NH4 + 50% NO3 and 100% NO3 treatments. Low and high PAR levels increased the nitrate concentrations and decreased the N/S ratio in leaves of both crops compared to medium PAR level. Fresh shoot biomass was maximized in Brassica rapa when PAR level was above the medium value and nitrate was supplied in the nutrient solution as NO3 or as a mixture of 50% NO3 and 50% NH4. The highest fresh shoot biomass of Brassica juncea was observed in all nutrient solution treatments at high PAR level.  相似文献
