The influence of type of diet and time after feeding on concentration of isoacids in rumen fluid of 6 fistulated sheep were investigated. The concentration of isoacids in rumen fluid was higher in diets rich in concentrate and protein (5.6) than in roughage diets (3.4) or in straw-starch-urea diets poor in native protein (highest concentration: 2.1 mmol/l rumen fluid). Feeding of roughage diet or straw-starch-urea-diet effected a significant decrease of concentration of isoacids in the rumen fluid after morning feeding, but concentrate-roughage diet, increased the isoacids-concentration. Reasons for decreased concentration of isoacids may be a shortage of corresponding amino acids and a high activity of cellulolytic microbes. Infusion of isoacids (3 g per day) in the rumen of sheep fed with a straw-starch-sugar-urea diet did not significantly influence the in sacco dry matter degradability of untreated wheat straw, but increased the dry matter loss of ammonia treated wheat straw from 16.0; 26.6; 39.4; 54.0 and 58.8% to 17.3; 29.7; 43.1; 56.3 and 63.0% after 6; 12; 24; 48 and 72 h rumen incubation time respectively. Further experiments seem to be necessary. The effect of isoacids may be expected to occur with poor quality feeds, rich in fibre low in protein and hence low in branched-chain amino acids.  相似文献

Effects of heat stress on multiparous lactating Holstein cows were evaluated in a continuous design in natural environments (a shade management system vs no shade, n = 6/treatment, early- to mid-lactation cows) and in a single-reversal design in climatic chambers (thermoneutral vs thermal stress, n = 4/treatment, mid- to late-lactation cows). Objectives were to compare effects of nycterohemeral heat stress vs nonheat stress environments on acid-base status, mineral concentrations and rates of ruminal liquid dilution and solid phase digesta turnover. In the shade vs no shade experiment, the complete mixed diet consisted of 38% corn silage and 62% corn-based concentrate mix. In the chamber experiment, the diet consisted of 40% alfalfa haylage and 60% corn-based concentrate mix. In both experiments, respiration rates, rectal temperatures, blood and urine were sampled hourly for 26 h. Measurements of rectal temperature, respiration rate and blood gas composition indicated that chamber heat stress simulated natural heat stress. In both experiments, cows exhibited signs of respiratory alkalosis only during hours of heat stress. Ruminal turnover rates of liquid were measured with chromium-ethylenediaminetetraacetate and solid turnover rates with ytterbium-marked fecal fiber, both dosed through the ruminal cannula. Turnover rates of liquid and solid digesta and total volatile fatty acids were lower in the heat stress vs the thermoneutral environment. The results illustrate the necessity of frequent sampling to characterize nycterohemeral patterns of physiological measurements during heat stress.  相似文献

A 1-year-old, female spayed domestic shorthair cat was presented with a 4-week history of dysphagia and regurgitation soon after oral treatment with clindamycin. Fluoroscopic and endoscopic examinations confirmed the presence of a single cervical oesophageal stricture 4 cm caudal to the pharynx. A fluoroscopically and endoscopically guided balloon dilation was performed six times consecutively over a period of 3 weeks as reformation of the stricture appeared within 3-7 days. Feeding via percutaneous endoscopic gastrostomy-tube as long-term management of the condition was declined by the owner. A self-expanding metal oesophageal stent with the following dimension was subsequently implanted: fully open diameter 16 mm, length 30 mm. After stent implantation, the cat was fed on mashed canned food and did not show any clinical signs for 12 months. Twelve months post-implantation the cat was no longer able to eat even liquid food, became lethargic and the owner opted for euthanasia. On post-mortem examination the stent surfaces were overgrown by oesophageal mucosa by approximately 50%. Stent obstruction was detected and caused by swallowed hair which also seemed to have hampered mucosal integration in the distal part of the stent.  相似文献

Ambrosiapilze sind in ihrer Toleranz gegenüber klimatischen Bedingungen weitgehend eingeschränkt und in ihrer Verbreitung auf den Insektensymbionten angewiesen, während die Käfer in ihrer Ernährung von den Symbionten abhängig sind. Ein spezielles Organ, das Mycetangium, wurde zur Erhaltung der Pilzflora außerhalb der Brutanlagen entwikkelt. Dieses Organ entstand polyphyletisch in den versichiedenen systematischen Einheiten. Die Symbiose ist in den Familien Scolytidae und Platypodidae und deren Untereinheiten mehrfach unabhängig voneinander entzanden. Die Reareaktionen auf die unterschiedlichen ökologischen Gegebenheiten sind daher sehr variabel.In many ambrosia beetles (wood boring Scolytidae and all Platypodidae) a special fungus transmitting organ, the mycetangium, ensures fungus survival during hibernation and/or flight as well as the inoculation of new galleries. Fatty nutritional and probably selective sheltering fluids are secreted by holocrinous gland cells connected with the mycetangium during the growth of funguis inside this organ and only then. Consequently, tolerance levels of true ambrosia fungi towards climatic conditions are narrowed down compared with other auxiliary or free blue stain fungi. Survival and proparation is dependent on the insect partner, while the ambrosia beetles depend on their microsymbionts for their nutrition. The ambrosia symbiosis has developed independently in the differen axonomic groups. Adaptations to the different ecological requisitions of the beetle results in a great variability of symbiotic patterns.

AIMS: To investigate the seroprevalence of antibodies to Toxoplasma gondii in New Zealand sea lions (Phocarctos hookeri), as a potential contributor to reproductive failure.

METHODS: Archived sera were sourced from New Zealand sea lions from two recolonising mainland populations in the Otago Peninsula (n=15) and Stewart Island (n=12), as well as a declining population at Enderby Island (n=28) in the New Zealand sub-Antarctic. Sera were tested for antibodies to T. gondii using a commercially available ELISA (with samples considered positive if the sample to positive ratio was?>30%), and latex agglutination test (LAT; with titres ≥1:32 considered positive). Western blot analysis was used to validate the results of a subset of 14 samples.

RESULTS: Five samples from sea lions in mainland locations were confirmed positive for antibodies to T. gondii. Two adult females exhibited high LAT antibody titres (min 1:2048, max 1:4096) on both occasions when sampled 1 and 2 years apart, respectively. No animals from Enderby Island were seropositive.

CONCLUSIONS: Toxoplasma gondii infection is unlikely to be a major contributor to poor reproductive success in New Zealand sea lions. However, continued surveillance is pertinent to assess subclinical and clinical impacts of the parasite on these threatened populations. The commercial tests evaluated here, with further species-specific threshold refinement could provide a fast, inexpensive and reliable indicator of T. gondii exposure in New Zealand sea lions.  相似文献

A pharmacokinetic/pharmacodynamic modelling approach was used to determine a dosage regimen which maximizes diuretic efficiency of torasemide in dogs. Kinetic profiles of plasma concentration, torasemide excretion rate in urine (TERU) and diuresis were investigated in 10 dogs after single oral administrations at 3 dose levels, 0.2, 0.8 and 1.6 mg/kg, and an intravenous injection of 0.2 mg/kg. Endogenous regulation was evidenced by a proteresis loop between TERU and diuresis. To describe the diuresis–time profile, TERU served as input into a turnover model with inhibition of loss of response, extended by a moderator acting on both loss and production of response. Estimated maximum inhibition of loss of response, Imax, was 0.984 showing that torasemide is an efficacious diuretic able to suppress almost total water reabsorption. A TERU50, value producing half of Imax, of 1.45 μg/kg/h was estimated from the model. Pharmacokinetic and pharmacodynamic parameters were used to simulate the torasemide dose–effect relationship after oral administration. Model predictions were in good agreement with diuresis measured in a validation study conducted in 10 dogs, which were administered oral doses of 0.15, 0.4, 0.75, 1.5 and 4.5 mg/kg for 5 days. Finally, oral dose associated with the highest daily diuretic efficiency was predicted to be 0.1 mg/kg.  相似文献
