This study was conducted to assess the abundance of iron-oxidizing bacteria and biological sulfur oxidation potential from soil impacted by coal and coal refuse from two coal-burning electric power facilities located at the U.S. Department of Energy's Savannah River Site (Aiken, S.C.) and the South Carolina Electric and Gas Site at Beech Island, S. C. Significantly higher MPN counts of iron-oxidizing bacteria were obtained from samples collected at the confluence of a coal storage runoff containment basin, a coal reject (refuse) pile, and an adjacent wetland at the Savannah River Site. Significant differences in pH, sulfate-S, ferrous- and ferric-iron were also obtained between sampling locations. No significant differences in ferric/ferrous ratios were determined. These ratios however, exceeded a value of 2.0 when sample pH values were less than 4.5. Under optimal conditions, biological thiosulfate-S oxidation potentials (in vitro) showed a 4- to 7-day lag in the appearance of sulfate-S, and a final pH (after twenty-four days of perfusion) of 1.97 to 3.90. These results indicate that contamination of subsurface water by acidic leachate derived from thionic bacterial activity will occur if coal and coal refuse piles are not confined by an impermeable surface or containment facility. 相似文献

Summary To evaluate the role of plasmids in soil communities antibiotic-resistant bacteria have been isolated from soil. Among them, 419 l-aminopeptidase positive strains (Gram-negative) and 28 l-aminopeptidase negative strains (Gram-positive) were screened for the presence of plasmids. Chromatographic, chemical, and spectroscopic techniques were used to characterize the physicochemical properties of napin purified by preparative chromatography. The molar extinction coefficient was determined (epsilon = 0.56), and static and dynamic light scattering measurements enabled the average molecular weight (M(w) = 13919), the second virial coefficient (A(2) = 23.95 x 10(-)(5) mol cm(3) g(-)(2)), and the hydrodynamic radius (R(H) = 1.98 nm) to be determined. No conformational changes were observed by fluorescence and circular dichroism measurements in different buffers at pH 3, 4.6, 7, and 12, confirming the high pH stability of this protein. From MALDI-TOF analysis and after enzymatic digestion, it was found that this purified sample, extracted from the rapeseed variety Express, contained mainly isoform 2SS3_BRANA. 相似文献

Present research in Germany on water, wind, and tillage erosion is examined. Existing soil‐protection strategies, laws, and guidelines are outlined. These are the result of extensive scientific investigations and the adoption of existing international knowledge. The most common methodologies currently used in Germany are reviewed. They allow an estimation of the water‐ and wind‐erosion risks as well as methods for their reduction. The results enable an aim‐orientated, site‐specific, risk‐adequate approach to implementing control measures in an easily understood framework. Purpose
Flood events are expected to increase both in intensity and frequency due to climate change in the near future. From an environmental toxicology perspective, there is concern that such flood events could lead to the remobilization of contaminated sediment layers in rivers. The aim of this pilot study was to establish a novel and interdisciplinary framework combining methods of hydrodynamic engineering and ecotoxicological assessment to enable investigation of the potential risks associated with such remobilization events.Materials and methods
Formulated sediment was prepared according to OECD guideline 218 and spiked with a mixture of four polycyclic aromatic hydrocarbons (phenanthrene, chrysene, pyrene, benzo[a]pyrene) at concentrations of 3.3?C8.3?mg?kg?1 dry weight. Rainbow trout (Oncorhynchus mykiss) were exposed as test animals to re-suspended sediments in three out of five experiments. The experiments were carried out in an annular flume designed to investigate transport behaviour of fine-grained sediments. Several physicochemical (e.g. pH) and sedimentological (e.g. turbidity) parameters were measured to characterise environmental conditions and erosion behaviour of sediments. Furthermore, exposure concentrations were measured by means of an in vitro assay (7-ethoxyresorufin-o-deethylase (EROD), RTL-W1 cell assay) and chemical analysis.Results and discussion
Preparation and spiking of large amounts of formulated sediments were feasible but not practical. Successful spiking could be confirmed by the bioanalytical methods with the spiked sediments showing significantly elevated EROD induction compared to control sediments. Conditions within the annular flume remained stable throughout all experiments and were adequate to support rainbow trout. Flood events were successfully simulated, resulting in re-suspension of formulated sediment. Different erosion behaviours of sediments during the simulated flood events were observed and could be associated with changes in microbial composition of sediments due to differences in storage conditions. Therefore, maintaining constant storage conditions of formulated sediments is crucial to enable consistency and comparability among erosion experiments.Conclusions
This study clearly demonstrated the feasibility of a combined hydro-toxicological approach in support of the investigation of the potential ecotoxicological relevance of sediment re-suspension events. However, based on the results presented here, it is recommended to include additional physicochemical parameters, such as redox potential and conductivity, and to extend the experimental setup to natural sediments and different aquatic organisms. Future studies will use natural sediments containing representative microbial communities and extracellular polymeric substances to enable extrapolation from the annular flume to conditions in natural flowing waters. 相似文献128.

The response of soil microbial communities following changes in land-use is governed by multiple factors. The objectives of this study were to investigate (i) whether soil microbial communities track the changes in aboveground vegetation during succession; and (ii) whether microbial communities return to their native state over time. Two successional gradients with different vegetation were studied at the W. K. Kellogg Biological Station, Michigan. The first gradient comprised a conventionally tilled cropland (CT), mid-succession forest (SF) abandoned from cultivation prior to 1951, and native deciduous forest (DF). The second gradient comprised the CT cropland, early-succession grassland (ES) restored in 1989, and long-term mowed grassland (MG). With succession, the total microbial PLFAs and soil microbial biomass C consistently increased in both gradients. While bacterial rRNA gene diversity remained unchanged, the abundance and composition of many bacterial phyla changed significantly. Moreover, microbial communities in the relatively pristine DF and MG soils were very similar despite major differences in soil properties and vegetation. After >50 years of succession, and despite different vegetation, microbial communities in SF were more similar to those in mature DF than in CT. In contrast, even after 17 years of succession, microbial communities in ES were more similar to CT than endpoint MG despite very different vegetation between CT and ES. This result suggested a lasting impact of cultivation history on the soil microbial community. With conversion of deciduous to conifer forest (CF), there was a significant change in multiple soil properties that correlated with changes in microbial biomass, rRNA gene diversity and community composition. In conclusion, history of land-use was a stronger determinant of the composition of microbial communities than vegetation and soil properties. Seed crops of white clover (Trifolium repens L.) are usually established with a cover crop. Provided sufficient light, white clover may compensate for low plant density by stoloniferous growth. Our objectives were (1) to compare spring barley or spring wheat used as cover crops for white clover and (2) to find the optimal seeding rate/row distance for white clover. Seven field trials were conducted in Southeast Norway from 2000 to 2003. Barley was seeded at 360 and 240 seeds m?2 and wheat at 525 and 350 seeds m?2. White clover was seeded perpendicularly to the cover crop at 400 seeds m?2/13 cm row distance or 200 seeds m?2/26 cm. Results showed that light penetration in spring and early summer was better in wheat than in barley. On average for seven trials, this resulted in 11% higher seed yield after establishment in wheat than in barley. The 33% reduction in cover crop seeding rate had no effect on white clover seed yield for any of the cover crops. Reducing the seeding rate/doubling the row distance of white clover had no effect on seed yield but resulted in slightly earlier maturation of the seed crop. 相似文献
