The residues from metered doses of cypermethrin were recovered from excised portions of grasshopper cuticle to determine losses by volatilisation. Residues from similar doses were also recovered from the cuticles of intact grasshoppers and from whole body homogenates after rinsing the cuticle, to determine rates of absorption and losses from metabolism. Residues were recovered from males and females one week and three weeks after fledging in grasshoppers infected with Malameba locustae and in uninfected ones. They were maintained at 15 or 30°C and sampled from each combination of factors at 4, 8, 16, 24, 30 and 48 h after treatment. There was no measurable loss of cypermethrin by volatilisation up to 72 h after application to excised portions of cuticle at either 15 or 30°C. In all combinations of factors, more cypermethrin was recovered externally (P <0.05) from grasshoppers held at 15°C than those held at 30°C and more from infected grasshoppers than from those that were uninfected. Significantly more cypermethrin was also recovered externally from males than from females (P <0.05) and more from infected one-week-old grasshoppers than those that were three weeks old. At 15°C there was a gradual trend towards accumulation of cypermethrin internally, but at 30°C an initial trend towards accumulation was followed by one of decline. Residual cypermethrin recovered could in general be related to the negative temperature coefficient of toxicity, to a decreased sensitivity to cypermethrin in infected grasshoppers and to an increased sensitivity in older grasshoppers.  相似文献

Clostridium difficile is widely known as a cause of disease in humans, and has emerged as an important problem in neonatal swine. No commercial product is available for immunoprophylaxis of C. difficile-associated disease, but success in preventing experimental infections in hamsters by use of nontoxigenic strains to competitively exclude toxigenic strains led us to try this method in neonatal pigs. Spores were administered orally to newborn pigs or were sprayed onto perineum and teats of dams. Significantly more piglets were weaned among litters receiving spores orally, and average weaning weights were significantly higher for both treatment groups than for controls. Toxins A and B were detected in 44.8% of litters and 16.5% of piglets born to sprayed sows and 58.3% of litters and 15.4% of piglets in the control group. However, toxins were detected in only 13.8% of litters and 3.4% of piglets given spores orally. These data support a contention that precolonization by a nontoxigenic strain can ameliorate the pre-weaning growth retardation associated with C. difficile infection in piglets.  相似文献

Loss of native grasslands by anthropogenic disturbances has reduced availability and connectivity of habitat for many grassland species. A primary threat to contiguous grasslands is the encroachment of woody vegetation, which is spurred by disturbances that take on many forms from energy development, fire suppression, and grazing. These disturbances are exacerbated by natural- and human-driven cycles of changes in climate punctuated by drought and desertification conditions. Encroachment of honey mesquite (Prosopis glandulosa) into the prairies of southeastern New Mexico has potentially limited habitat for numerous grassland species, including lesser prairie-chickens (Tympanuchus pallidicinctus). To determine the magnitude of impacts of distribution of mesquite and how lesser prairie-chickens respond to mesquite presence on the landscape in southeastern New Mexico, we evaluated seasonal space use of lesser prairie-chickens in the breeding and nonbreeding seasons. We derived several remotely sensed spatial metrics to characterize the distribution of mesquite. We then used these data to create population-level resource utilization functions and predict intensity of use of lesser prairie-chickens across our study area. Home ranges were smaller in the breeding season compared with the nonbreeding season; however, habitat use was similar across seasons. During both seasons, lesser prairie-chickens used areas closer to leks and largely avoided areas with mesquite. Relative to the breeding season, during the nonbreeding season habitat use suggested a marginal increase in mesquite within areas of low intensity of use, yet aversion to mesquite was strong in areas of medium to high intensity of use. To our knowledge, our study is the first to demonstrate a negative behavioral response by lesser prairie-chickens to woody encroachment in native grasslands. To mitigate one of the possible limiting factors for lesser prairie-chickens, we suggest future conservation strategies be employed by land managers to reduce mesquite abundance in the southern portion of their current range.  相似文献

Isoflavones have potential for preventing and treating several chronic health conditions, such as osteoporosis, cardiovascular disease, and cancer. In this study, radiolabeled isoflavones were recovered from kudzu (Pueraria lobata) root cultures after incubation with uniformly labeled (14)C-sucrose in the culture medium for 21 days. Approximately 19% of administered label was recovered in the isoflavone-rich dried extracts of kudzu root cultures (90.2 microCi/g or 3.3 MBq/g extract). HPLC-PDA analysis revealed the predominant isoflavones isolated from kudzu root cultures to be puerarin, daidzin, and malonyl-daidzin. The average concentration of the major isoflavone puerarin in kudzu root cultures was 33.6 mg/g extract, with a specific activity of 63.5 microCi/g (2.3 MBq/g). The isolated isoflavones were sufficiently (14)C-labeled to permit utilization for subsequent in vivo metabolic tracking studies.  相似文献

The Muyong forest, an indigenous secondary forest in Banaue, Ifugao, Philippines, plays a crucial role in the Muyong–Payoh system, a continuum of secondary forest and rice terrace, of the Banaue rice terraces by providing water and nutrients to the rice plants in the Payoh terraces. In recent decades, the planting of introduced tree species in the Muyong forest has threatened the sustainable provision of ecosystem services such as water balance and nutrient cycling. To further understand nutrient cycling in the Muyong–Payoh systems, this study was conducted in Poitan, Banaue, Ifugao to gather preliminary baseline data on floral diversity, leaf litterfall rate, leaf litter decomposition rate, and diversity and succession of arthropods in decomposing leaf litter in a Muyong forest. Vegetation analysis was done by identifying and describing the trees growing inside the five 10 m × 10 m quadrat sampling plots. Monthly leaf litter fall was collected in 1 m × 1 m litter traps, and the dry weight was determined after oven-drying at 65 °C for 48 h. Leaf litter decomposition experiment was established by laying out 12 nylon mesh bags containing fresh leaf litter in each of the four sites on the forest floor and one bag was retrieved every month to determine the change in dry weight of the leaf litter. Six bulk soil samples were collected from the Muyong forest floor and analyzed for organic matter, pH, available P and exchangeable K. Fresh leaf litter samples were analyzed for total N, P and K contents. Arthropods in the collected decomposing leaf litter were extracted using Berlese funnel and later identified up to families level using arthropod taxonomic key. The diversity of plants in the Muyong forest includes thirty-eight tree species belonging to 19 families dominated by indigenous tree species. Results showed that the monthly leaf litter fall was higher during the dry months of March to May and lower during the wet months. The estimated total leaf litter fall in Muyong forest was comparable to published litter fall from tropical secondary forests. The N, P and K contents of fresh leaf litter range from 1.0 to 1.2, 0.11 and 0.40%, respectively. The first month of decomposition has the fastest rate while the decomposition rate during the next 4 months ranged from 0.125 to 0.251. Complete decomposition or mass lost in the leaf litter in the Muyong forest took place within 5 months. The soil arthropods identified in the decomposing leaf litter were composed of 13 orders and 28 families. Majority of the collected arthropods were insects while other species including mites, spiders, millipedes and sowbugs were also present. Detrivore and fungivore Families were found to be dominant in the decomposing leaf litter. Moreover, the composition and succession of arthropod decomposer community varied in the three sampling methods and with the changing quality of the litter material as decomposition progressed. The wide diversity and succession of leaf litter decomposers consisting of detritivores, predators, fungivore and herbivore coupled with abundant rainfall and warm temperature are the two main factors that contribute to the fast rate of leaf litter decomposition and nutrient turn over in the Muyong forest. Thus, the Muyong forest can sustain the productivity of rice planted in the adjoining downstream Payoh terraces. Hence, the conservation and management of the Muyong forest is critical in maintaining the ecological functions of the Muyong–Payoh continuum.  相似文献

Bread flour was spiked with folic acid (1.40 mg/lb or 3.08 μg/g of flour) and processed into bread by the sponge and dough method. Changes that occurred to added folic acid and endogenous folate contents through different processing stages, including sponge formation, proofing, and baking, were assessed by reversed‐phase ion‐pair HPLC combined with UV and fluorometric detection. Sample extraction required α‐amylase and rat plasma deconjugase digestion, and sample preparation required purification by solid‐phase extraction. Added folic acid was measured by monitoring UV absorption at 280 nm. Four selected forms of endogenous folates including tetrahydrofolate (THF), 5‐formyl‐THF, 10‐formylfolate, and 5‐methyl‐THF were identified and quantified throughout the bread processing using a fluorescence excitation wavelength of 290 nm and emission wavelength of 350 or 450 nm. Data indicate a relatively good stability of added folic acid and native folates to the baking process, and increased endogenous folate contents in dough and bread as compared with the flour from which they were made.  相似文献

We monitored the decomposition of mixed leaf litter (Quercus spp., Carya spp., and Pinusechinata) in a Missouri Ozark forest eight years after experimental harvest. Leaf litter mass losses and changes in carbon chemistry (extractive, acid soluble, and acid insoluble fractions) were measured over 32 months in field incubations to determine the effects of litter composition and stand manipulation on decomposition and nitrogen (N) concentration in the remaining litter. The decay (k) rate over this period ranged between 0.39 (±0.010) and 0.51 (±0.002) year−1 for oak, oak–hickory, and oak–pine litter. There were significant main effects of stand manipulation (p = 0.03) and litter type (p < 0.01) on decay. Mass losses of oak and oak–hickory litter were 7% (p = 0.02) and 4% (p = 0.04) higher on harvested stands than controls, respectively. Mass loss of oak–hickory litter was 3% faster than oak–pine (p = 0.03) and 6% faster than oak (p = 0.02) litter on control stands, whereas the oak–hickory litter mass loss was 5% higher than oak litter on harvested stands (p = 0.01). The decay (k) rate had a linear relationship with initial leaf litter nitrogen content and lignin-to-N ratio. The nitrogen concentration in remaining litter had a nonlinear relationship to cumulative mass loss suggesting an exogenous source of N. In summary, this study demonstrated significant effects of timber harvest and litter mixtures on decomposition and N dynamics in a managed Missouri Ozark forest.  相似文献

It has long been recognized that limitations exist in the analytical methodology for amylose determination. This study was conducted to evaluate various amylose determination methods. Purified amylose and amylopectin fractions were obtained from corn, rice, wheat, and potato and then mixed in proportion to make 10, 20, 30, 50, and 80% amylose content starch samples for each source. These samples, considered amylose standards, were analyzed using differential scanning calorimetry (DSC), high-performance size-exclusion chromatography (HPSEC), and iodine binding procedures to generate standard curves for each of the methods. A single DSC standard equation for cereal starches was developed. The standard curve of potato starch was significantly different. Amylose standard curves prepared using the iodine binding method were also similar for the cereal starches, but different for potato starch. An iodine binding procedure using wavelengths at 620 nm and 510 nm increased the precision of the method. When HPSEC was used to determine % amylose, calculations based on dividing the injected starch mass by amylose peak mass, rather than calculations based on the apparent amylose/amylopectin ratio, decreased the inaccuracies associated with sample dispersion and made the generation of a cereal amylose standard curve possible. Amylose contents of pure starch, starch mixtures from different sources with different amylose ranges, and tortillas were measured using DSC, HPSEC, iodine binding, and the Megazyme amylose/amylopectin kit. All the methods were reproducible (±3.0%). Amylose contents measured by these methods were significantly different (P < 0.05). Amylose measurements using iodine binding, DSC, and Megazyme procedures were highly correlated (correlation coefficient >0.95). DSC and traditional iodine binding procedures likely overestimated true amylose contents as residual butanol in the amylose standards caused interference. The modified two-wavelength iodine binding procedure seemed to be the most precise and generally applicable method. Each amylose determination method has its benefits and limitations.  相似文献

The physical and biochemical properties of Atlantic cod (Gadus morhua) wastes were analysed, and the waste remediation potential of blue mussels (Mytilus edulis) was assessed. Waste generated daily by Atlantic cod represented 24.9% of the cod feed added to the system. Particle distributions determined using a Coulter Multisizer and image analysis revealed that the majority of the particles in terms of numbers occupy the smaller size ranges; however, larger particles occupy a larger proportion of the volume. Effluent was composed of particles <70 μm (36%), 70–500 μm (31%) and particles >500 μm (33%) by weight. The amount of dissolved carbon and nitrogen associated with the effluent represented 3.1% and 3.7%, respectively, of the total feed added to the system daily. Particles <70 μm had significantly less organic matter, lipids and fatty acids and were expected to be ingested more by mussels than larger particles. The major lipid classes present in effluent were free fatty acids, triacylglycerols, phospholipids, acetone mobile polar lipids and sterol. Cod effluent contained two essential fatty acids DHA and EPA, a diatom marker (16:1ω7), as well as two zooplankton markers 22:1ω11 and 20:1ω9, which accumulated in mussels and may serve as markers for aquaculture wastes. Although only 36% of the effluent was of a size suitable for mussel ingestion, this size fraction has the greatest potential to spread to surrounding areas. These particulates may be useful as an alternate food source when natural seston is low.  相似文献
