Transplantation of human amnion epithelial cells improves learning and memory function in Alzheimer's disease-like pathology rat model

AIM: To observe the treatment effect and its immune regulation of human amnion epithelial cells (hAECs) on Alzheimer's disease (AD)-like pathology rat model. METHODS: The hAECs were isolated from amnion with trypsin digestion, and the phenotype of hAECs was analyzed by flow cytometry. SD rats (n=48) were randomly divided into sham control group, model group, medium group and hAECs group. AD-like pathology rat model was induced by bilateral intraventricular injection of lipopolysaccharide (LPS). hAECs (5×10⁵) were injected into the hippocampus of the AD-like pathology rats. At 2 weeks after transplantation, the animals were tested by Morris water maze to observe the function of learning and memory. The pathological change of the brain was observed by HE staining. The expression of amyloid β-protein 42(Aβ42) and Tau protein and the level of acetylcholine (ACh) in the injury brain were determined by immunohistochemistry. The survival and differentiation of hAECs in the hippocampus were measured by immunofluorescent technique. The percentages of lymphocyte subsets in the peripheral blood mononuclear cells were analyzed by flow cytometry. The contents of serum cytokines were detected by cytometric bead array. RESULTS: Compared with model group and medium group, hAECs group showed shortened escape latency (P<0.01), increased frequency of going through the platform (P<0.05), reduced loss of hippocampal neurons, decreased expression of Tau protein and Aβ42 in the hippocampus (P<0.05), increased ACh level in the hippocampus (P<0.05), decreased percentages of Th1 and Th17 subsets, increased percentages of Th2 and Treg cells (P<0.05), decreased concentrations of IFN-γ and IL-2 in the serum, and increased concentration of IL-4(P<0.05). CONCLUSION: hAECs improve the cognitive learning and memory function and alleviate pathologic damage of hippocampus through immune regulation in AD-like pathology rats.     

EZH2-mediated regulation of NF-κB target gene expression in gastric cancer

AIM: To explore the mechanism by which over-expression of enhancer of zeste homolog 2 (EZH2) in a panel of gastric cancer cell lines is involved in tumorigenesis of gastric cancer. METHODS: Real-time PCR and Western blot were employed to examine the mRNA and protein levels of EZH2, respectively. MTS assay, cell migration and soft agar assay were performed to investigate the role of EZH2 in the regulation of stomach cancer behaviors. The effect of EZH2 on NF-κB target gene expression was determined by Luciferase reporter and real-time PCR. Co-immunoprecipitation was used to analyze the interaction of EZH2 and p65 in HEK293T cells. RESULTS: The expression levels of EZH2 were significantly increased in the gastric cancer cells compared with normal gastric epithelial cells. Pharmacological inhibition by DZNep or knockdown of EZH2 significantly compromised AGS and SNU-16 cell activity, cell migration and anchorage-independent cell growth. Moreover, siRNA knockdown of EZH2 impaired NF-κB downstream targets, such as IL-8, CXCL5 and CCL20. In addition, the interaction of EZH2 and p65 was detected. CONCLUSION: EZH2 mediates the growth of gastric cancer cells through the regulation of NF-κB downstream gene expression.     

美洲黑杨无性系苗期光合生理特性研究

研究了9个美洲黑杨无性系的光合生理特性,为优良新无性系的选育提供参考。采用LI-6400便携式光合作用测定系统测定美洲黑杨的光合作用日变化及其光响应曲线,利用非直角双曲线Farquhar模型、二次曲线及直线方式拟合光响应曲线,分析比较光响应特征参数及影响净光合速率因子。结果表明:各无性系的净光合速率均具有明显的日变化规律,tn01-78、tn01-38、tn04-n52呈双峰曲线现象,2个峰值出现的时间在11:00和14:00左右,其余呈单峰曲线,净光合速率最大值出现在12:00左右;不同无性系最大净光合速率、暗呼吸速率、表观量子效率、光饱和点和光补偿点的大小范围分别为20.50~31.42μmol·m-2s-1、1.67~3.19μmol·m-2s-1、0.055~0.279μmol·μmol-1、950.83~1 360.83μmol·m-2s-1和29.03~55.20μmol·m-2s-1;净光合速率(Pn)变化与气孔导度Gs、水压亏缺Vpdl、空气温度Tair和光合有效辐射VPAR呈正相关,与胞间CO2浓度Ci和大气相对湿度(HR)呈负相关;净光合速率与影响因子的相关程度依次为气孔导度>光合有效辐射>胞间CO2浓度>大气相对湿度>空气温度>水压亏缺。

肾上腺皮质肿瘤良恶性鉴别方法探究

肾上腺皮质肿瘤（Adrenocortical Tumor,ACT）是一种常见的肿瘤,存在于人体的内分泌系统中,并且是引发肾上腺皮质癌（Adrcnocortical Carcinoma,ACC）进而导致死亡的主要原因.经典的＂Weiss系统＂在诊断明确性上存在种种无法解决的问题,但随着科技水平的发展,人们开始探索新的途径来鉴别ACT的良恶性,于是开始有了影像学和分子生物学技术等特异性的检查方法,从而进一步增强了诊断的明确性.现通过对比的方式,探究肾上腺皮质肿瘤（Adrenocortical Tumor,ACT）影像学检查和分子生物学技术检查的结果,从而能更准确地鉴别其良恶性.     

基于EXCEL的作物灌水率图自动绘制与修正

在灌区的规划设计中,传统作物灌水率图的绘制一般都是手工方法,不仅效率低且修正起来很不方便,而且现有的灌水率图绘制软件一般存在着灌水率计算和灌水率图绘制相互独立、无法实时动态修正等不足.基此,综合利用EXCEL软件强大的数据处理、分析功能以及图形自动绘制功能,将灌水率计算与灌水率图绘制有机结合,从而实现灌水率图的快速绘制和动态调整.这种方法操作方便、简单,值得在工程设计部门推广使用.

Study on Anti-viral Effect of Rhubarb on Porcine Epidemic Diarrhea Virus in vitro

The inhibitory effects of rhubarb on cell lesion, which was caused by porcine epidemic diarrhea virus (PEDV), and its mechanism had been studied in this research.The maximum safe concentration of rhubarb solution applied to Vero cells was determined through morphological observation, and then the PEDV infected Vero cell model in vitro was established, by which the inhibitory effect of rhubarb on virus was studied.The experimental method had also been established based on this cell model.The result showed that the maximum safe concentration was 3.28 mg/mL.Both morphological observation and MTT assay were indicated that rhubarb could not only block the attachment of the virus to Vero cell and inhibit the synthesis and reproduction of the virus, but also could directly inactivate the virus.This research showed that rhubarb had an inhibitory effect on PEDV, and provided a theoretical basis for the prevention and treatment of clinical porcine epidemic diarrhea.
