Global scale modeling is reviewed with respect to global circulation models, biosphere-atmosphere models, and climate-biosphere models. These different models focus on short to long time scale interactions between atmospheric and surface systems. Remote sensing is shown to play a central role in acquisition of data for these models, and an experiment, termed FIFE, is described, which is the first attempt to take simultaneous land surface observations of meteorological and biophysical parameters at sufficient resolution to test hypotheses linking the vegetated surface and circulation within the lower atmosphere.  相似文献

The interpretation of muscle structure by scanning electron microscopy (SEM) has not been consistent among various studies. Consequently, the literature is confusing with respect to the identity of T-tubules, transverse ridges, Z-disks, and intermyofibrillar connections. The objective of this research was to evaluate the effects of different methods of sample preparation and imaging on ultrastructural details of previously identified transverse structures and intermyofibrillar connections and to verify or disprove the commonality of these structures under different viewing conditions. Scanning electron microscopy coupled with a cold stage, SEM at room temperature, and transmission electron microscopy (TEM) of thin sections were most appropriate for exposing detail of inter- and intracellular structures and for measuring sarcomere length and spacing of intermyofibrillar connections. Scanning electron microscopy of samples mounted on a cold stage, fractured, and sublimed provided excellent images of meat and muscle ultrastructure and may be used in correlative microscopy. Sarcomere length and spacing between intermyofibrillar connections were similar among most specimen preparation techniques and were affected similarly by heat treatments. Results indicate that the regularly spaced transverse structures viewed by conventional SEM and the intermyofibrillar connections viewed by low-temperature SEM are Z-disks.  相似文献

Ventilation and gas exchange were studied in healthy, adult horses and cows, two large species with different lung structures and different breathing patterns. The oxygen uptake (VO2), carbon dioxide output (VCO2), respiratory rate (fR), minute ventilation (VE), alveolar ventilation (VA), alveolar oxygen pressure (PAO2), and VE/VO2 ratio were higher in the cows, while the tidal volume (VT) and physiological dead space (VD) were larger in the horses. The arterial blood gases, alveolar-arterial oxygen pressure difference (PAO2-PaO2) and VD/VT ratio did not differ between the two species. The higher VO2 in the cows was most likely due to the energy cost of standing, and possibly to a higher cost of digestion. The higher VE, VA, VE/VO2 and PAO2 were most likely secondary to the increased VO2 and the slightly higher respiratory exchange ratio (R) in the cows. In contrast to hypotheses based on allometric equations, the PAO2 of horses and cows did not appear to differ from that of smaller mammals. The VD was larger than that predicted from allometric equations, and even though the VD/VT ratio (0.50) was lower than the previously reported values for horses and cows, it was significantly larger than the predicted weight-independent value of 0.36. Re-examination of the data used to derive the equation for VD raised questions as to the validity of this equation, and it is suggested that caution be exercised in the use of allometric equations for prediction.  相似文献

A complex interaction of multiple factors has resulted in an oak decline event in oak-hickory forests of the Ozark Mountains of Arkansas and Missouri, U.S.A. The most striking feature of this situation is an unprecedented population explosion of red oak borer, a species of cerambycid beetle, Enaphalodes rufulus (Haldeman), which appears to be causing extensive mortality to mature red oaks (Quercus, subgenus Erythrobalanus). The insect is a native species, historically a minor pest of oaks, found throughout the eastern United States. Beetles normally reproduce in living oaks, as larvae initially feed in phloem tissue and subsequently bore into xylem where pupation occurs. The life cycle is two years in length and synchronous adult emergence occurs in odd-numbered years. Data from previous research indicate average attack densities of less than four per tree with a high of 71 on a single tree. Historical emergence densities are similarly low and the highest reported was 15 adults from one tree. Our research is concerned with understanding factors contributing to this outbreak; developing sampling methods for red oak borer; assessing oak mortality; and evaluating site and stand conditions associated with the current outbreak. Results of our initial sampling reveal dramatically higher average attack densities of 244 per m2 and emergence densities of 18 per m2 of bark surface area. We confirm a three-week period of adult emergence and activity during mid-June to early July. We also report on possible management responses by federal and state agencies to this remarkable epidemic and oak mortality crisis.  相似文献

This study was conducted to evaluate the influence of age, breed, epididymectomy and semen processing on the concentration of estradiol-17 beta (E2) in bovine semen. Semen was collected either by electroejaculation or with an artificial vagina. Neat semen samples were stored at -20 C until analysis. Processed, frozen semen and an egg yolk-citrate semen extender were obtained from a commercial semen processing firm and stored in liquid nitrogen at -196 C. The concentration of E2 in semen was determined by radioimmunoassay. Semen from mature (greater than 24 mo), fertile Brahman (n = 19), Brangus (n = 16), Charolais (n = 29), Holstein (n = 15) and Santa Gertrudis (n = 25) bulls was analyzed for E2 concentration, and no difference (P greater than .10) between breeds was found. There was no difference (P greater than .10) in seminal E2 concentration between mature, fertile bulls (n = 104) and epididymectomized bulls (n = 22). In semen collected from prepuberal (12 to 16 mo, n = 21), peripuberal (17 to 20 mo, n = 17) and mature (greater than 24 mo, n = 19), Brahman bulls, the mature bulls had a lower (P less than .01) semen E2 concentration than peripuberal and prepuberal bulls. To evaluate the impacts of fishing on marine ecosystems, the total extraction of fish must be known. Putting a figure on total extraction entails the difficult task of estimating, in addition to reported landings, discards, illegal and unmandated catches. Unreported catches cast various types of shadow, which may be tracked and estimated quantitatively. Some shadows of unreported catches are reviewed, for example, an innovative, well‐funded NGO publicizes illegal catch in the Southern Ocean. For various reasons, official figures often have the implicit but unacceptable assumption that such categories are null. We present an estimation procedure based on adjustment factors taken from observer reports, correspondents and published information that track changes in a regulatory regime, and hence reflect incentives and disincentives to misreport. Monte Carlo simulations address uncertainty using multiple sources of information to provide upper and lower estimates. Once in place, this method provides preliminary estimates that may be refined without disruption. The method is demonstrated for fisheries in Iceland and Morocco. We use a ‘by‐species’ approach for Icelandic cod and haddock, while the Moroccan catch is divided into demersal and pelagic categories. Results suggest that Icelandic cod catches may have been underestimated by between 1 and 14% at different times, and haddock by between 1 and 28%. Underestimation of Moroccan catches appears to have been as much as by 50%. These case studies show that it is possible to obtain estimates of misreporting, even when direct data are lacking. Our method encourages transparency because sources of information are presented so that uncertain values are easily identified, offering a basis for comment, collaboration and refinement in estimating illegal and unreported fishing.  相似文献

It is of nutritional significance to fortify processed dairy products (e.g., cheese, yogurt, and ice cream) with vitamin D3; however, the inherent complexity of these foods may influence the stability and bioavailability of this nutrient. In the present study, the interactions of vitamin D3 with beta-lactoglobulin A and beta-casein were investigated under various environmental conditions (i.e., pH and ionic strength) using fluorescence and circular dichroism spectroscopic techniques. The results indicated that vitamin D3 was bound to both beta-lactoglobulin A and beta-casein depending on the solution conditions. The apparent dissociation constants ranged from 0.02 to 0.29 microM for beta-lactoglobulin A, whereas the beta-casein apparent dissociation constants ranged from 0.06 to 0.26 microM. The apparent mole ratios were also comparable, i.e., 0.51-2.04 and 1.16-2.05 mol of vitamin D3 were bound per mole of beta-lactoglobulin A and beta-casein, respectively. It was concluded that these interactions may strongly influence vitamin D3 stability and, hence, bioavailability in processed dairy products.  相似文献
