The human T-lymphotropic viruses types I and II (HTLV-I and -II) have been etiologically linked with certain T-cell leukemias and lymphomas that characteristically display membrane receptors for interleukin-2. The relation of these viruses to this growth factor receptor has remained unexplained. It is demonstrated here that introduction of the trans-activator (tat) gene of HTLV-II into the Jurkat T-lymphoid cell line results in the induction of both interleukin-2 receptor and interleukin-2 gene expression. The coexpression of these cellular genes may play a role in the altering T-cell growth following retroviral infection. 相似文献

Over the last two decades, there have been significant advances in the use of assisted reproductive technology for genetic and reproductive management of captive dolphin populations, including evaluation of sperm DNA quality. This study validated a customized sperm chromatin dispersion test (SCDt) for the bottlenose dolphin (Tursiops truncatus) as a means of assessing sperm DNA damage both in the field and in the laboratory. After performing the SCDt, two different sperm morphotypes were identified: (i) sperm with fragmented DNA showed large haloes of dispersed DNA fragments emerging from a compact sperm nucleoid core and (ii) sperm containing non‐fragmented DNA displayed small compact haloes surrounded by a dense core of non‐dispersed DNA and protein complex. Estimates of sperm DNA fragmentation by means of SCDt were directly comparable to results obtained following a two‐tailed comet assay and showed a significant degree of correlation (r = 0.961; p < 0.001). This investigation also revealed that the SCDt, with minor modifications to the standard protocol, can be successfully conducted in the field using a LED florescence microscopy obtaining a high correlation (r = 0.993; p = 0.01) between the data obtained in the laboratory and in the field. 相似文献

Potato yield and quality are highly dependent on an adequate supply of water. In this study, 3 years of information from thermal and RGB images were collected to evaluate water status in potato fields. Irrigation experiments were conducted in commercial potato fields (Desiree; drippers). Two water-deficit scenarios were tested: a short-term water deficit (by suppressing irrigation for a number of days before image acquisition), and a long-term cumulative water deficit. Ground and aerial images were acquired in various phenological stages along the potato growing season. Effects of irrigation treatments were recorded by thermal indices and biophysical measurements of stomatal conductance (SC), leaf water potential, leaf osmotic potential and gravimetric water potential in soil. Canopy temperature was delineated from the thermal images with and without fused information from the RGB image. Crop water stress index (CWSI) was calculated, using three forms of minimum baseline temperature: empirical, theoretical and statistical. An empirical evaluation of maximum baseline temperature of Tair + 7 °C was used in all CWSI forms examined. Statistical tests and comparison of CWSI with biophysical measurements were performed to evaluate the responses to irrigation treatments. The results indicated a high correlation of CWSI with SC from tuber initiation to maturity based on ground and aerial data (0.64 ≤ R2 ≤ 0.99). Similar trends of increasing CWSI from well to deficit-irrigated treatments were found in all three growing seasons. The results also showed that CWSI may be calculated based merely on thermal imagery data. 相似文献

Noninvasive angiography with magnetic resonance is demonstrated. Signal arising in all structures except vessels that carry pulsatile flow is eliminated by means of velocity-dependent phase contrast, electrocardiographic gating, and image subtraction. Background structures become in effect transparent, enabling the three-dimensional vascular tree to be imaged by projection to a two-dimensional image plane. Image acquisition and processing are accomplished with entirely conventional two-dimensional Fourier transform magnetic resonance imaging techniques. When imaged at 0.6 tesla, vessels 1 to 2 millimeters in diameter are routinely detected in a 50-centimeter field of view with data acquisition times less than 15 minutes. Studies of normal and pathologic anatomy are illustrated in human subjects. 相似文献

Analyses of fossil mammal faunas from 2945 localities in the United States demonstrate that the geographic ranges of individual species shifted at different times, in different directions, and at different rates in response to late Quaternary environmental fluctuations. The geographic pattern of faunal provinces was similar for the late Pleistocene and late Holocene, but differing environmental gradients resulted in dissimilar species composition for these biogeographic regions. Modern community patterns emerged only in the last few thousand years, and many late Pleistocene communities do not have modern analogs. Faunal heterogeneity was greater in the late Pleistocene. 相似文献

Protein microarrays provide a powerful tool for the study of protein function. However, they are not widely used, in part because of the challenges in producing proteins to spot on the arrays. We generated protein microarrays by printing complementary DNAs onto glass slides and then translating target proteins with mammalian reticulocyte lysate. Epitope tags fused to the proteins allowed them to be immobilized in situ. This obviated the need to purify proteins, avoided protein stability problems during storage, and captured sufficient protein for functional studies. We used the technology to map pairwise interactions among 29 human DNA replication initiation proteins, recapitulate the regulation of Cdt1 binding to select replication proteins, and map its geminin-binding domain. 相似文献

The low vine vigor and high N requirement of Russet Norkotah may lead to N loss and groundwater contamination on coarse-textured soils. Recent clonal selections from Texas have produced strains that have larger and stronger vines, which may alter N requirements. This twoyear study examined the N use efficiency (NUE), yield, and quality of Russet Norkotah strain selections fertilized with different N levels on a Hubbard loamy sand in central Minnesota. The selections, Texas Norkotah Strain (TXNS) 112, TXNS 223, and TXNS 278 were grown with standard Russet Norkotah under irrigated conditions and received total N levels of 28, 112, 224, or 336 kg ha-1. Total, marketable, and large (>340 g) tuber yields increased linearly (P>0.05) with rate of N application in 1998 but not in 1997. The genotype main effect was not significant for any of the tuber yield parameters measured based on fresh weight. Vine, tuber, and total dry biomass yields were 116%, 5.8%, and 13.2%, respectively, higher with the selections than Russet Norkotah. Harvest index (HI), or the proportion of total dry matter partitioned to tubers, was 7% greater for Russet Norkotah than the TXNS selections, reflecting the larger vine growth of the selections. The selections accumulated significantly higher N in the vines (0.113 kg kg-1 N) than the standard clone (0.053 kg kg-1 N) as N rate increased from 28 to 336 kg ha-1, and the difference between the selections and the standard clone was larger at higher N rates than at lower N rates. Russet Norkotah partitioned 10% more N to tubers than did the TXNS selections, reflecting the difference in HI between the standard cultivar and its clones. Nitrogen recovered from fertilizer N applied in addition to the 28 kg ha-1 starter N (NUE28) averaged 36% and varied little with genotype, N rate, or cropping year. Biomass accumulation from similar N additions (AUE), however, was significantly higher for the selections than Russet Norkotah at 112 kg N ha-1 in 1997 only. At low N rate (112 kg ha-1), the selections had higher physiological use efficiency (PUE28) (mean 45.9 g g1) than Russet Norkotah (25.9 g g1). Results from this study demonstrate that, although N recovery was similar for the four genotypes, the Texas Norkotah strains produced greater biomass than Russet Norkotah per kg N applied at low rate in 1997 and per kg of fertilizer N absorbed by the plant in both years. The aim of this study was to evaluate the viability in the effect of open pulled straw (OPS) vitrification procedure of sheep embryos after direct transference. Embryos were produced in vivo and cryopreserved in slow freezing or OPS vitrification. The survival rates of cryopreserved embryos were compared to non-frozen standard pattern. In a first set of experiments, embryos at morula and blastocyst stages were dived in ethylene glycol (1.5 M) and frozen in an automatic freezer. After being thawed, they were directly or indirectly transferred to ewes recipient. A second group of embryos were drawn into OPS and plunged into liquid nitrogen after being exposed at room temperature for 1 min and 45 s in 10% EG plus 10% dimethyl sulphoxide (DMSO), then again for 30 s in 20% EG + 20% DMSO + 0.5 M sucrose. After being warmed, embryos were also directly transferred using a French mini straw as the catheter for the transplantation process or after in vitro dilution of cryoprotectants (two-step-process). No significant difference was observed among fresh, frozen or vitrified embryos on pregnancy rate (50.0%, 38.6% and 55.8%). However, when we evaluated only the direct transference, the pregnancy rate of OPS vitrified embryos was higher than that of frozen embryos (57.1% vs 34.8%) (p = 0.07). In addition, vitrified morulae had a higher pregnancy rate than the one with frozen embryos (64.0% vs 38.9%) (p = 0.07). Finally, our results indicate that OPS vitrification technique in association with direct transference improves the viability of sheep embryos with potential applications to field conditions. 相似文献
