Monoclonal antibodies (MAbs) reported here were produced against the porcinophilic foot-and-mouth disease virus (FMDV) that caused the devastating swine disease on 1997 in Taiwan. A panel (25) of MAbs were found to react with VP1 of O/Taiwan/97 (O/97) by ELISA with various potencies. The biological identities of these VP1 reacting MAbs, such as neutralization activity, isotype and capability to distinguish between two serotype O FMDVs, O/97 and O/Taiwan/KM1/99 (O/99), were further analyzed. Eleven out of the total eighteen O/97 neutralizing MAbs were able to neutralize heterologous O/99. Eight O/97 neutralizing and five non-neutralizing MAbs could differentiate two serotype O FMDVs by immunofluorescence assay (IFA) implied that these thirteen MAbs recognized O/97 specific epitope(s). Furthermore, reactivities of the VP1 reacting MAbs with a 29 amino acids synthetic peptide (P29) representing the betaG-betaH loop of VP1 were analyzed by ELISA and fourteen were found positive. MAb clone Q10E-3 reacting strongest with VP1 and P29, neutralizing both but not differentiating two serotype O viruses suggested that the antibody binding site might involve the RGD motif and its C terminal conserved region on betaG-betaH loop. MAbs with diverse characters presented in this study were the first raised against porcinophilic FMDV. The complete set of MAbs may be used for further studies of vaccine, diagnostic methods, prophylaxis, etiological and immunological researches on FMDV. 相似文献

通过对影响蓄积量的因子进行相关性分析,筛选出与蓄积量存在较好相关性的指标作为自变量。但其自变量间存在多重共线性,会对模型稳定性、预测精度产生影响。通过多元统计分析中的主成分分析法,构造出影响密云县森林蓄积量的主成分,然后与蓄积量进行回归,得到主成分回归,并与一般线性回归模型进行比较。结果表明:主成分线性模型在拟合度、模型适用性与预测精度上都优于一般线性模型。主成分回归模型的复相关系数为0.809,预测精度达到88.26%。 相似文献

The extent to which terrestrial ecosystems can sequester carbon to mitigate climate change is a matter of debate. The stimulation of arbuscular mycorrhizal fungi (AMF) by elevated atmospheric carbon dioxide (CO(2)) has been assumed to be a major mechanism facilitating soil carbon sequestration by increasing carbon inputs to soil and by protecting organic carbon from decomposition via aggregation. We present evidence from four independent microcosm and field experiments demonstrating that CO(2) enhancement of AMF results in considerable soil carbon losses. Our findings challenge the assumption that AMF protect against degradation of organic carbon in soil and raise questions about the current prediction of terrestrial ecosystem carbon balance under future climate-change scenarios. 相似文献

从草鱼(Ctenopharyngodon idellus)出血病疑似病料提取物感染的草鱼肾细胞(CIK)中提取病毒基因组dsRNA,电泳显示出水生呼肠孤病毒基因组的典型特征。应用简化的FLAC(full-length amplification of cDNA)技术扩增得到病毒的4个全长基因组片段进行测序分析。核酸序列BLAST分析表明:其中3个基因组片段与GCRV-873第5、9、10片段高度同源,另外1个基因组片段与GCRV-HZ08第11片段高度同源;因此推测病料中同时存在两种不同的病毒核酸,但也可能存在一种杂合病毒。根据已发表的GCRV-HZ08第5、9、10片段设计了3对引物对分离的病毒总RNA进行RT-PCR检测并测序,结果显示这3个片段与GCRV-HZ08第5、9、10片段均具有99%同源性,表明是由于混合感染而存在两株不同病毒的核酸dsRNA,两株病毒分别命名为GCRV-JX01和GCRV-JX02。首次运用dsRNA测序法检测出了GCRV病毒的混合感染,为草鱼呼肠孤病毒的流行病学和防控提供了一种新的技术选择。 相似文献

Survey on the contamination of Toxoplasma gondii oocysts in the soil of public parks of Wuhan, China

Toxoplasma gondii of warm-blooded animals and humans is an important pathogenic agent throughout the world. Soil is increasingly recognized as an important source in the transmission of Toxoplasma. To attain the contamination status of T. gondii in the soil of public parks, a total of 252 soil samples were collected from September 2009 to August 2010 at different sites located in 6 public parks of Wuhan, Hubei, China and detected by PCR and loop-mediated isothermal amplification (LAMP). The detection limit of PCR/B1, PCR/529 and LAMP was determined to be 50, 5, and 5 tachyzoites in soil, respectively. Forty-one samples were found positive for Toxoplasma DNA by PCR on both genes, whereas LAMP products were generated in 58 samples (χ(2)=3.6328, P=0.0567). All parks were found contaminated and no significant difference was found among the parks (PCR: χ(2)=0.0072, P=0.9325; LAMP: χ(2)=0.6101, P=0.4347). However, contamination was found with significantly different among the four seasons (PCR: χ(2)=11.6066, P=0.0007; LAMP: χ(2)=12.4636, P=0.0004), with a gradual decrease in the prevalence from spring to winter on both analyses. This is the first investigation on soil contamination of public parks in China by T. gondii oocysts. The results indicate that the soil of public parks contaminated with T. gondii oocysts may play a role in the epidemiology of toxoplasmosis and effective preventive measures should be considered. Moreover, the conventional PCR and LAMP used in the present study are applicable to detect T. gondii oocysts in soil samples. 相似文献
